1.Cloning and Expression of EGFR Tyrosine Kinase and Construction of the Screening Model for the Enzymatic Inhibitors

As a member of tyrosine kinases(TK) family,Epidermal growth factor receptor(EGFR) has an activity of intrinsic protein tyrosine kinase,which plays an essential role in the regulation of signal transduction in the cells.Due to the abnormal expression of EGFR-TK,the certain type cancers may developed and progressed.Based on that,the inhibitors of EGFR-TK could be effective medicines for the treatment of cancer.The EGFR-TK domain was amplified by RT-PCR with RNA of HUVCEs cells as the template and expressed in E.coliBL21(DE3) using plasmid pET30a as vector.The recombinant protein was purified with the affinity chromatography(Ni-NTA),which was identified to have kinase activity catalyzing the substrate phosphorylated with ATP in the enzymatic reaction.Using the recombinant EGFR-TK as target,the screening model for enzymatic inhibitors was constructed.
3.Effect of Specific Growth Rate and Nitrogen Source on the Production of Recombinant Lateolabrax japonicus Growth Hormone by Pichia pastoris

The bioreactor production of recombinant Lateolabrax japonicus growth hormone (rljGH) expressed intracellularly by Pichia pastoris was investigated. A strategy of feeding methanol at the exponential rate was established and the effect of specific growth rate on the rljGH production was examined. The results indicated that the average specific production rate increased and the rljGH production duration decreased as the specific growth rate increased. The maximum specific rljGH production (0.58 mg/g WCW) was achieved at a specific growth rate of 0.029/h. The effect of supplementing ammonium sulfate, peptone and yeast ex- tract on the rljGH production was further investigated. The results indicated that the effects of ammonium sulfate and peptone were not significant. Supplementing yeast extract of 2.5 g/L was advantageous for the rljGH production. The duration of the rljGH production was increased to 23 h from 17 h and the fermenta- tion stability of run-to-run could be improved.
4.Current situation of undergraduate education of rehabilitation therapeutics in colleges and universities in China

Rehabilitation therapeutics has become a popular specialty in colleges and universities in China. At present, 160 colleges and universities have set up rehabilitation therapeutics specialty, of which 73.75% are public colleges and 26.25% are private colleges (including independent colleges). The types of those colleges and universities are mainly concentrated on western medicine colleges (40.6%), traditional Chinese medicine colleges (18.1%) and comprehensive colleges (31.3%), with an imbalance in regional distribution between the east and west of China. Some problems are becoming prominent with the increase of the number of these colleges and universities, such as differences in discipline cultivation modes, compositions of colleges and universities, teaching quality, profession/industrial standards and distribution of rehabilitation education resources. In order to promote the healthy development of undergraduate education of rehabilitation therapeutics in colleges and universities in China, we should define the connotation and extension of rehabilitation therapeutics, establish the professional training mode of rehabilitation therapeutics with Chinese characteristics, classify the professional training objectives, strengthen the construction of teachers, formulate professional standards of rehabilitation therapeutics, and improve the uneven distribution of rehabilitation education resources.
5.Pediatric Nosocomial Infection:Reasons Analysis and Prevention Strategies

OBJECTIVE To investigate the reasons of pediatric nosocomial infection,analyze the possible risk factors,and provide the effective control and prevention strategies. METHODS A total of 18002 patients were investigated in 2007 by prospective study. The data were analyzed via SPSS 11.5 statistical analysis software using mainly descriptive and contrast analysis. RESULTS The nosocomial infection rate was 2.44%. The children were relatively highly infected on both of age from 1-month-old to 1-year-old (3.67%) and stay in hospital over 10 days (7.85%) ,the season in third and fourth quarters (3.13% and 2.64%). The nosocomial infection occurred mainly in respiratory tract and gastrointestinal tract,and more respiratory infections occurred in winter and spring,but more gastrointestinal tract infections occurred in summer and autumn. CONCLUSIONS Pediatric patients are more easy infected of the baby age or the longer stay in the hospital. Pediatric nosocomial infection more occurrs in the summer and autumn. Hospitals should focus on preventing and controlling the respiratory and gastrointestinal infections.

Recently, neuroimging techniques are widely applied in the nervous systemic disease. With the development of neuroimaging, the neuroimaging studies of Alzheimer's disease (AD) are more and more in-depth. The main purpose is to identify AD patients in the early stage (clinically normal), to treat and prevent the disease in the early stage. This article reviewed the progress of various of imaging studies in AD disease.
7.The Influence of ?-Synuclein Overexpression on Mitochondrial Membrane Structure with Atomic Force Microscopy

Objective:To identify the effect of ?-synuclein overexpression on mitochondrial membrane structure with atomic force microscopy. Methods:?-syn expression was mediated by AAV (adeno-associated viral vector) and Recombinant AAV/?-syn and AAV/LacZ viral particles were stereotaxically injected in the left side of rat substantia nigra (SN) for rat model of ?-synuclein overexpression. Mitochondria were isolated from rats SN of Brain. Mitochondria were analysis with JC-1 staining,atomic force microscopy and Western blot. Results:By 16 weeks post-infection of AAV-?-syn,the level of ?-syn increased about 2 times in mitochondrial fraction with Western blot and mitochondrial membrane potential (??) decreased with JC-1 staining. Furthermore,mitochondria swelling and porous like structure formed on the mitochondrial membrane with atomic force microscopy. Conclusion:The data suggested that ?-syn could accumulate in mitochondria,might form mitochondrial membrane pores and lead to ?? decreases. ?-syn might lead to mitochondrial dysfunction in Parkinson's disease.
8.Selective Screening of Inborn Errors of Metabolism by Urease Pretreatment-Gas Ch romatography-Mass Spectrometry:Pilot Study of 327 Patients at High Risk

Objective The purpose of this paper is to screen inborn errors of meta bolism (IEM) by analyzing urinary components, so as to provide laboratory guide for their diagnosis and therapy.Methods Urine samples of patients suspected to have IEM were collec- ted.Urea was de compo sed with urease and n-heptadecanoic acid was added as internal standard.Protein was denatured with ethanol and precipitate was removed by centrifugation,dried b y evaporation, the residue was trimethylsilylly derivatized with BSTFA/TMCS,and then analyzed with GC-MS for quantification of organic acids, amino acids,suga rs, polyols, purines and pyrimidines, simultaneously. This procedure is denom inated as urease pretreatment-gas chromatography-mass spectrometry (UP-GC-MS) internationally.Results Urinary samples of 327 patients from 6 provinces, cities and autonomous regions were analyzed,and 16 kinds of 27 cases of IEM were screened out with a positiv e rate of 8.26%,among which there were 3 cases of hyperphenylalaninemia,3 cases of glyceroluria,3 cases of Leigh syndrome, 2 cases of propionic acidemia, 2 case s of methylmalonic aciduria, 2 cases of von Gierke′s disease, 2 cases of fructo se-1,6-diphosphatase deficiency, 2 cases of fructosuria, 1 cases of multiple car boxylase deficiency, 1 cases of glutaric acidemia typeⅠ, 1 cases of maple sy rup urine disease, 1 cases of hyperglycinemia, 1 cases of 3-aminoisobutyric acid uria,1 cases of adult-onset typeⅡcitrullinemia,1 cases of galactosemia and 1 ca ses of Fanconi′s syndrome.Several IEM patients above had died,but satisfactory therapeutic effects had been achieved in some diseases,in cluding multiple carboxylase deficiency,methylmalonic aciduria and galactosemia. Other patients′ condition remained to be followed up.Conclusion Analysis of urinary components by UP-GC-MS provides a valuable tool for screenin g of IEM and the results will help to provide effective diagnostic and therapeut ic guide for the patients. 9.Treatment of osteomyelitis and bone defect of femoral shaft by external fixation and bone transport.

OBJECTIVETo discuss the clinical effects and superiority of applying external fixation and bone transport to treat osteomyelitis and bone defect of femoral bone.
METHODSFrom August 2008 to December 2013,16 patients with osteomyelitis and bone defect of femoral bone were treated including 11 males and 5 females with an average age of 42 years old ranging from 13 to 62 years old. The average course of disease was 18 months ranging from 2 months to 4.5 years, and the average length of bone defect was 7.8 cm ranging from 4.5 to 15 cm. The bone defect of all cases were treated by external fixation and bone transport, the bone transport began at 1 week after operation, 1 mm per day and 4 times per day.
RESULTSAll patients were followed up for 10 to 36 months (means 22.5 months). One patient did not cooperate with treatment leads to the failure, then took the amputation. The remaining 15 cases of osteomyelitis were under control, including 12 cases of bone transport achieved one stage bone union, 3 cases achieved bone union via bone graft from iliac bone. The bone union time was 5 to 13 months(means 7.9 months). Thirteen patients almost obtained the same length of two lower extremities,2 patients had shortening of 1.5 to 2 cm. The time of moving the external fixation was from 6 to 16 months (means 9.3 months).
CONCLUSIONApplication of external fixation and bone transport is an effective method in treating the osteomyelitis and bone defect that can control the infection, eradicate wounds, and be the equalization of limb length.

10.Analysis of surveillance results of drinking-water supply status in drinking-water-borne endemic fluorosis areas in Henan

Objective To understand the status of drinking-water supply and the progress of waterimproving projects in drinking-water-borne endemic fluorosis areas in Henan,and to provide scientific data for related government departments to carry out prevention and surveillance plan for those areas.Methods Questionnaire survey was carried out in all the villages in drinking-water-borne endemic fluorosis areas in Henan in 2010.Each village was given one set of questionnaire.Results By the end of 2010,the total number of fluorosis villages in Henan was 25 434,among them 11 484 villages had been conducted water-improving projects,accounting for 45.15%.Currently,9267 water-improving projects worked properly,accounting for 36.4%(9267/25 434)of all villages surveyed,and 80.7% (9267/11 484)villages had water-improving projects.The projects in 2217 villages do not work properly,accounting for 19.3%(2217/11 484) of all projects.There were 5832 water-imp.roving projects conducted amnog 2005-2010,accounting for 50.8% (5832/11 484) of all water-improving projects.Among villages with no such projects,97.5%(15 769/16 167) of them used shallow groundwater,and in villages with water-improving projects,89.6% (8303/9267) of them used deep groundwater as drinking-water supply.Conclusions After 2005,the water improving progress was significantly speed up in drinking-water-borne endemic fluorosis areas in Henan,but progress of the water improving projects is relatively slow due to changing and expanding of fluorosis areas.The government should increase investment and improve the proportion of waterimproving defluoridation.
