1.Expression of OBR and NPY in mouse hypothalamus

Objective To observe the expression of OBR and NPY in mouse hypothalamus.Methods In mouse hypothalamus,the location and coexpression of OBR and NPY were observed with immunohistochemistry and double immunohistochemistry.Results OBR positive cells distributed as clump in hypothalamus ME,ARC and VMN,having obscure boundary.OBR positive cells were also present in choroid plexus,brain ependymal layer cell and vascular endothelial cells.In hypothalamus ARC,NPY positive neurons were present with bright red color in cell plasma.The NPY positive neurons were found as round or ellipse,having many neurites.NPY positive fibers were present in ME.In double immunohistochemistry result,the coexpression of OBR and NPY showed black color,because that OBR positive cells showed brown purple or dark purple granula near the NPY positive neurons.Conclusion OBR distributed in ME,ARC,VMN of mouse hypothalamus,choroid plexus,brain ependymal layer cells and vascular endothelial cells.Meanwhile NPY also distributed in ME,ARC,cerebral cortex and hippocampus and so on.Moreover the coexpression of OBR and NPY was present in mouse hypothalamus ARC.
2.Investigation of the clinical features and risk factors in elderly patients with acute pulmonary thromboembolism

Objective To investigate the differences in clinical symptoms and risk factors between non-elderly and elderly patients with acute pulmonary thromboembolism(PTE).Methods Totally141 patients with acute PTE and 100 sex and age-matched healthy controls were enrolled in the study.The final diagnosis was confirmed by CT pulmonary angiography (CTPA) and/or pulmonary angiography and/or pulmonary perfusion imaging as well as medical history.The patients with acute PTE were divided into two groups according age:elderly group (n=78,age≥60 years),young group (n =63,age < 60 years).Multivariate Logistic regression analysis was performed to identify independent risk factors of PTE and to confirm the risk of each factor.Results The morbidity rate of dyspnea,cough,chest distress and chest pain were 88.5% (69 cases),76.9% (60 cases),56.4% (44 cases),15.4% (12 cases) in elderly group,which was similar to 84.1% (53 cases),69.8% (44 cases),63.5% (40 cases),25.4% (16 cases) in young group (all P>0.05); while the morbidity rate of hemoptysis in elderly group [9.0% (7 cases)] was lower than in young group [23.8%(15 cases)](P<0.05).9% (7 cases) in elderly patients without any symptoms were more than 1.6% (1 cases) in young patients,but no significant differene (P>0.05).There were more risk factors in the elderly group accompanying with diabetes (x2 =7.41,P < 0.01),hypertension (x2 =14.51,P< 0.01),chronic obstructive pulmonary diseases (COPD) (x2 =7.42,P<0.01) and previous stroke (x2 =4.91,P<0.05)compared with young group.Independent risk factors for elderly acute PTE included COPD (OR:3.29,95%CI:1.04-10.46),previous deep veneus thrombosis(DVT) (OR:4.72,95%CI:1.68-13.27),cancers (OR:4.35,95%CI:1.00-18.13) and previous stroke (OR:5.13,95%CI:1.15-24.52).Independent risk factors for non-elderly PTE included previous DVT (OR:11.94,95% CI:3.35-42.60)and cancers (OR:11.44,95%CI:1.44-89.92).Conclusions Much attention should be paid to the identification of diagnosis of acute PTE depending on the non-specific clinical features,although dyspnea may be the most frequency symptoms,but unexplained cough and chest distress should be alert for PTE.COPD,cancer,DVT and previous stroke are independent and important risk factors for elderly acute PTE.
3.The clinical characteristics of 169 cases of severe fever with thrombocytopenia syndrome

Objective To analyze the clinical characteristics of severe fever with thrombocytopenia syndrome (SFTS) so as to improve the recognition of the emerging infectious disease.Methods A retrospective analysis was performed upon clinical manifestations,laboratory test results and prognostic features of 169 patients with SFTS admitted to the 154 Hospital,Chinese People's Liberation Army from October 2010 to May 2011.The patients were divided into moderate disease group and severe disease group according to the prognosis.The differences between two groups were compared to explore the prognostic indicator of severe type.Results All patients with SFTS inhabited in hilly ground with history of field work.The main clinical symptoms were severe fever (98.8%,167/169),headache (52.1%,88/169),muscle soreness (95.9%,162/169),nausea (73.4%,124/169),vomiting (67.5%,114/169),coughing (61.5%,104/169),etc.Superficial lymph node enlargement with haphalgesia was observed in 45.0%(76/169) patients.Petechia or ecchymosis was observed in 36.7% (62/169) patients.Critical patients were mainly aged > 60 years,associated with nerve and circulatory system syndrome,prominent hemoptysis,dyspnea,and nearly 73.3% (22/30) of severe thrombocytopenia.Serum levels of aspartate aminotransferase,creatinine and urea nitrogen were significantly higher in severe disease group than those in moderate disease group [235 (47-1750) U/L vs 88 (14-2000) U/L,997 (281-2601) U/L vs 399 (26-2633)U/L,101 (62-291) μmol/L vs 70(26-205) μmol/L,7.0(2.2-20.0) mmol/L vs 4.8(1.4-18.5) mmol/L,all P values <0.001].Conclusion Nerves system syndrome,transaminase and urea nitrogen are risk factors of prognosis of severe SFTS to whom deserves paying attention.
5.Clinical studies on the combiantion of single lower limb traction and massage therapy for the lumbosacral tunnel syndrome

Objective To observe the therapeutic effect of single lower limb traction plus massage for the lumbosacral tunnel syndrome.Methods The selected 80 candidate patients were divided into the observation group with 38 patients and treatment group with 39 patients. The observation group received single lower limb traction plus massage and the treatment group received pelvic traction plus massage. Both groups therapy last 5 days, The Japanese orthopaedic association scores (JOA) was evaluated for subjective symptoms, signs, functional activity and bladder function score. The therapeutic effect was evaluated.Results The total effective rate (94.7% vs. 82.1%;χ2=12.160,P=0.007) in the observation group was significantly higher than that in the control group. O, The scores of JOA at one week after treatment (20.2 ± 2.0 vs. 18.3 ± 2.8;t=3.419, P<0.01) and two weeks after treatment (24.4 ± 1. 4vs. 22.6 ± 3.0;t=3.359,P<0.01) in the observation group were significantly higher than those in the control group.Conclusions The single lower limb traction plus massage can improve the clinical symptoms of patients with lumbosacral tunnel syndrome, and be superior to the pelvic traction plus massage.
6.N-terminal of ?-Synuclein Involved in Regulation of Mitochondrial Function

Objective: To identify the functional domain of ?-Synuclein in affecting mitochondrial function and how the function to be impaired,especially,the mitochondrial membrane potential and the release of Cytochrome c.Methods: Harvest of ?-Syn-N and ?-Syn-△N by PCR,then subcloned into the pCMV-Myc mammalian expression vector.The recombinant plasmids were transfected into HEK293T cells by Lipofectamine 2000.After detecting the protein expression by Western blot,the functional domain was detected by co-immunoprecipitation.The mitochondrial membrane potential through flow cytometry and immunofluorescence,at the same time,the release of Cytochrome c through flow cytometry to detect.Results: The recombinant plasmids were constructed successfully.CO-IP has proved that N-terminal may be the functional domain of ?-Synuclein in affecting mitochondria.Over-expression of N-terminal could depolarize the mitochondrial membrane potential and induce the Cytochrome c releasing in MN9D cells.Conclusion: N-terminal may be the functional domain of ?-synuclein and over-expression of N-terminal could decrease mitochondrial activity.
7.Assessment for health education in plague affected area in Dingbian country, Shaanxi province in 2008

Objective To improve the knowledge about controlling plague in cadres, masses, and the medical staff in plague affected area in Dinghian county of Shaanxi province and to assess the efforts of health education activities. Methods In 2008, the education activities carded out by the government-related departments were investigated. The awareness of plague control and assessment was obtained through a written survey, questionnaire and interviews on clinic. Results Some education activities were carried out in plague area of Dingbian county, such as training, issuing educational materials, broadcasting plague scientific educational films and arranging knowledge grand prix. The rates of knowing plague clinic, epidemiology, prevention and the "three prohibitions and three alerts to report" were as follows: the cadres were 50.50%(101/200),63.69%(414/650),78.67%(118/150), the masses were 64.71% (66/102),87.91% (269/306),76.47% (78/102) and the medical staff were 64.18% (543/846) ,68.51%(322/470),67.02%(63/94). The passing rates of the cadres, the masses and the medical staff were 70.00% (35/50),92.16% (47/51),74.47% (35/47). Conclusions Health education can strengthen health consciousness of cadres and masses and improve the ability of the medical stalf on controlling sudden epidemic situation. Reinforcing plague control knowledge and training of medical staff are still important for health education in the future.
8.Combined hepatectomy in radical resection for hilar cholangiocarinoma

Objective To evaluate the value of combined hepatectomy in radical resection for hilar cholangiocarinoma. Methods The clinical data and follow-up data of 67 patients of resection for hilar cholangiocarinoma in Henan Tumor Hospital from June 2005 to october 2008 were retrospectively analyzed. Results According to intraoperative exploration situation and bismuth types, tumor resection was combined performed with hepatectomy (n=38)or non-hepatectomy (n=29). The rate of R0 resection was 55.3% in hepatectomy group(n=21) and 34.5% in non-hepatectomy group(n=10), and the difference was significant(P=0.024). The incidence of complications were 39.5% in hepatectomy group(n=15) and 13.4% in non-hepatectomy group(n=4), and one patient with liver and kidney failure died in hospital. The 1, 3, 5 years of survival rate were 89.3%,53.6% and 32.1% respectively in R0 group (n=31) and 69.7%,30% and 10% respectively in R1~R2 group(n=36), there were significant differences in the postoperative survival rate between both groups(P=0.018). The 1, 3, 5 years of survival rate were 81.8%,48.5% and 24.2% in hepatectomy group and 75%,32% and 16% in non-hepatectomy group respectively, and the differences were significant(P=0.037). Conclusions Aggressive resection including combined hepatectomy for hilar cholangiocarcinoma can play an important role for curative effect and long term survival rate.
9.Purification,antiserum preparation and antigenic characters of ClpC2 in Myco-bacterium tuberculosis

Objective:To investigate the antigenicity of ClpC 2 and the feasibility of polyclonal antibodies of ClpC 2 as detected antibody.Methods:rClpC2 was induced with IPTG.The rClpC2 was identified by SDS-PAGE and Western blot ,and purified by affinity chromatography ,with which rabbit were immunized and the specificity of rabbit antiserum was detected by Western blot , the titer of rabbit antiserum against ClpC2 was detected by double immunodiffusion and indirect enzyme-linked immunosorbent assay (ELISA).The antigenicity of the rClpC2 was detected by ELISA.The polyclonal antibodies of ClpC 2 were prepared to detect the ClpC 2 in clinical serum of TB patients by ELISA.Results:SDS-PAGE showed specific protein band with a relative molecular mass of 46 kD.The rClpC2 could bind with the antibody in the blood serum of the mouse immuned by MTB.By Bandscan analysis rClpC 2 accounted for about 58.7%of the total bacteria protein ,the purity of rClpC2 was 88.5% after purification.The ClpC2 of BCG could bind with the rabbit antiserum.The titer of antiserum were 1∶32 and 1∶320 000 by double immunodiffusion and ELISA detected respectively.ELISA results showed that clinical serum positive rate of rClpC 2 antigen was 46%in TB patients,the sensitivity of this protein was 46%,and the spe-cificity of this protein was 90%.ELISA results showed that the sensitivity of rabbit antiserum against ClpC 2 was 40%, and the specificity was 90%.Conclusion: Successfully expressed and purified rClpC 2 and high titer polyclonal antibody were successfully prepared,and these results will provide basements for further study on the biological functions of ClpC 2 and its candidate potentiality as serological diagnosis and drug-target and biological functions of antiserum against ClpC 2.
10.Safety test of light-emitting diode (blue) light to rat retina

BACKGROUND: Foreign studies have demonstrated that the blue light at 470 nm inhibits melatonin secretion and displays the most obvious biorhythm regulation. To date, light-emitting diode (LED) applied in regulating biorhythm remains poorly explored. OBJECTIVE: To explore whether a certain intensity of LED (blue) light could induce retinal injury in rats. DESIGN, TIME AND SETTING: Randomized, controlled animal experiment was performed at the Animal Laboratory of Peking University Third Hospital between May 2007 and April 2008. MATERIALS: A total of 32 SD rats and 16 BN rats were provided by Animal Department of Peking University Third Hospital. METHODS: A total of 16 SD and 16 BN rats were respectively randomly divided into test and control groups. Test group rats were placed in light boxes which were controlled by blue LED (wavelength 470 nm) at a intensity of 300-350μW/cm2, 4 hours everyday for 3 days. The remaining SD rats were placed in light box which was controlled by blue LED (wavelength 470 nm) at a intensity of 120-150μW/cm2, 4 hours everyday for 3 days. The control rats were not treated. MAIN OUTCOME MEASURES: At the second day after light irradiation, the rats of all groups were sacrificed and both eyeballs were harvested. The frozen sections were subjected to hematoxylin-eosin staining to observe changes of rat retina. RESULTS: A total of 48 rats were included in final analysis. The retina of SD rats became thinning and disorderly arranged following blue LED irradiation at density of 300-350μW/cm2, but the retina of BN rat remained unchanged similar to control group. After blue LED irradiation at density of 120-150μW/cm2, the retina of SD rat remained unchanged similar to control group. CONCLUSION: Blue LED light source irradiation at a intensity of 300-350μW/cm2 is safe to pigment-protected retina, and at a intensity of 120-150μW/cm2 does not injury retina of different races of rats.
