2.Preparation and pharmacokinetic evaluation of long-acting injectable oily suspensions for ophiopogonis radix polysaccharide MDG-1.

OBJECTIVETo evaluate in vivo pharmacokinetics of Ophiopogonis Radix polysaccharide MDG-1 oily suspension injection prepared with different prescriptions in rats, and explore the feasibility of the long-acting drug delivery of MDG-1 Injection by using the oily suspension drug release system.
METHODMDG-1 microparticles were prepared by the anti-solvent precipitation method. Their size and size distribution were characterized. Castor oil with a high viscosity or aluminum stearate were added into soybean oil with a low viscosity, in order to prepare oily media with different viscosities, detect their rheological properties and screen out superior prescriptions for in vivo evaluation.
RESULTThe average size of microparticles was 21.81 microm, and the span between them was 2.63. The in vivo evaluation was conducted for prescriptions of mixed oil (soybean oil/castor oil, 2: 3) and soybean oils gelled by 2% and 4% aluminium stearate. Among them, the prescription of soybean gelled by 4% aluminium stearate could significantly reduce C(max) and prolong the apparent t1/2, with the MDG-1 release time of several days.
CONCLUSIONIt is feasible to achieve the long-acting MDG-1 drug delivery by using oily media with a high viscosity.

3.Primary observing pulmonary function variety following three-dimensional conformal radiation therapy of Ⅲ phase non-small cell lung cancer

Objective To explore the pulmonary function,DVH and radiation pneumonitis after three-dimensional conformal radiation treatment ofⅢphase non-small-cell lung cancer.Methods 71 pa- tients (male 52,female 19,median age 63,KPS≥80) were evaluated by pulmonary function tests before radiotherapy and in M1 and M3 after radiotherapy respectively.After 3 months of follow-up time,it reviewed the appearance and grade of radiation pneumonitis.Then V_(20),V_(30) and MLD were worked out from dose vol- ume histogram.Results All patients completed radiotherapy,and total dose was 66-70 Gy.FVC (L), FEV1 (L) and CLCO were (2.58?0.65) L,(1.85?0.58) L and (15.15?4.65)ml/(min)before radio- therapy,with (2.96?0.76) L,(2.13?0.65) L and (14.71?3.92) ml/(min) in Ml after radiotherapy, with (2.65?0.61) L,(1.92?0.52) L and (13.15?3.71)ml/(min)in M3 after radiotherapy.The ac- cidence of radiation pneumonitis was 30%,moderate and severe radiation pneumonitis was 7%.With V_(20), V_(30) and MLD increasing,the grade of radiation pneumonitis was increasing.V_(20),V_(30) and MLD were related to the change in CLCO value among before,M1 and M3 after radiotherapy,and the correlation coefficient was more than 0.2.Conclusions There is a relationship in the pulmonary function,DVH and radiation pneu- monitis surely.The change in C_LCO value between before radiotherapy and M1 after radiotherapy could pre- dict the radiation pneumonitis.V_(20),V_(30) and MLD are not only correlated to radiation pneumonitis evidently but the change in FEV_1 and C_LCO after radiotherapy.
4.Inhibitory effect of arctigenin on lymphocyte activation stimulated with PMA/ionomycin.

This study investigated the effect of arctigenin (Arc) on the cell activation, cytokines expression, proliferation, and cell-cycle distribution of mouse T lymphocytes. Mouse lymphocytes were prepared from lymph node and treated with Phorbol-12-myristate-13-acetate (PMA)/Ionimycin (Ion) and/or Arc. CD69, CD25, cytokines, proliferation and cell cycle were assayed by flow cytometry. The results showed that, at concentrations of less than 1.00 micromol x L(-1), Arc expressed non-obvious cell damage to cultured lymphocytes, however, it could significantly down-regulate the expression of CD69 and CD25, as well as TNF-alpha, IFN-gamma, IL-2, IL-4, IL-6 and IL-10 on PMA/Ion stimulated lymphocytes. At the same time, Arc could also inhibit the proliferation of PMA/Ion-activated lymphocytes and exhibited lymphocyte G 0/G1 phase cycle arrest. These results suggest that Arc possesses significant anti-inflammatory effects that may be mediated through the regulation of cell activation, cytokines expression and cell proliferation.

Objective To evaluate the diagnostic value of thick-slab single-shot turbo spin-echo (SSTSE)T2 ?-weighted sequence in magnetic resonance fetography (MRF)for fetal abnormalities.Methods 328 of 1 990 pregnant women with the diagnosis of fetal congenital defects on prenatal ultrasound screening or chromosome examination were randomly selected,and 338 fetuses were ob-tained.These fetuses were scanned by conventional magnetic resonance imaging (MRI)and MRF.The diagnostic results from the two MR methods were compared.Results Six hundred and twenty-four lesions were detected by MRF.The primary diagnosis based on conventional MRI was changed for 14 lesions (2.2%).New findings were identified for 48 fetal lesions (8.4%)and 66 ma-ternal lesions.However,78 fetal lesions could not to be identified by MRF.MRF could increase the diagnostic confidence for fetal lesions with high water content (56.1% of the lesions).Conclusion MRF can yield more precise information for fetal extremities, fluid-filled cavities,pathological hydrops and cystic lesions.As an additional aid to the conventional multi-slice T2-weighted se-quence.
6.Comparative analysis on residents' dietary structure and health status in 2007 and 2012 in Songjiang District of Shanghai

Objective To investigate the dietary pattern and health status of residents in Songjiang District,Shanghai from 2007 to 2012 and provide basis for nutrition education and nutrition improvement.Methods Data were systemized from "Health and Nutrition Survey among Residents in Shanghai",which was carried out in Songjiang District in 2007 and 2012.Statistical analysis was made on average every standard person daily intake of vrious foods,dietary pattern and change of health status.Results The average every standard person daily intake of cereal foods,fruits,milk,beans and nuts increased as compared with those in 2007,while the intake of vegetables,aquatic products,livestock and poultry meat decreased from 2007 to 2012 in Songjiang District.And the intake of the livestock and poultry meat declined the most (from 137.4 g to78.8 g),that was closer to the recommended intake.Residents''intake of energy and nutients both decreased except dietary fiber and VitC,and the proportion of energy supply with carbohydrates had an increasing trend while the fat for energy supply a rising trend.The prevalence rate of hyperlipidemia,high blood pressure and diabetes for residents aged 15 and above were on the rise.Conclusion The dietary pattern of residents in Songjiang District has been with irrationality and nutrition related diseases such as hyperlipidemia,high blood pressure and diabetes has had a remarkably rising trend.Specific and various nutrition education should be carried out to promote the establishment of rational diet concepts and to improve the residents'' health level.
7.Study about the clinical and pathological characteristics of salivary duct carcinoma.

OBJECTIVETo investigate the clinical and pathological characteristics of salivary duct carcinoma (SOC) so as to provide basis for the diagnosis and therapy of SOC.
METHODSEight patients suffered with SOC treated in Affiliated Stomatological Hospital of Kunming Medical College from March 2001 to February 2009 were studied retrospectively including the clinical data. Meanwhile, eight tumor specimens were examined for vascular endothelial growth factor (VEGF), cytokeratin (CK), gross cystic disease fluid protein (CC DFP)-15, androgen receptor (AR) and oncogene C-erBb-2 protein expression via immunohistochemistry.
RESULTSOf the eight patients with SOC, five tumors grew in the parotid gland, the majority of the patients had facial nerve paralysis, three had recurrence. Computerized tomography (CT) showed that no obvious margin and similar crystal. Immunohistochemical staining for the expression of VEGF, CK, GCDFP-15 and C-erbB-2 were carried out in all SOC, while AR in majority.
CONCLUSIONSOC is a high-grade malignant carcinoma among all salivary carcinoma, immunohistochemical analysis of SOC has revealed a variety of similarities to ductal carcinoma of the breast. GCDFP-15, AR and C-erBb--2 may play an important role in pathological diagnosis for SOC.

8.Scene-trait coping style of military rescuers in Wenchuan earthquake.

OBJECTIVEThis study aimed to explore the scene-trait coping style of military rescuers in Wenchuan earthquake in an effort to provide scientific evidences for mental intervention program for Chinese military personnel.
METHODSBy cluster sampling, a total of 151 military rescuers and 331 control servicemen were administered the military personnel scene-trait coping style scale (MPSTCSS).
RESULTSAll active coping factor scores, and passive coping factor scores of affection, health and economy in the rescuer group were significantly higher than those in control group (P < 0.05). The 21-above age subgroup, the 3-year plus service subgroup, and the officer subgroup had significantly higher active coping factor scores on military tasks, military experience and personal development than those of the 21-below age subgroup, 3-year minus service subgroup and the soldier subgroup, respectively (P < 0.05 or P < 0.01).
CONCLUSIONThe earthquake relief servicemen can cope with stressful situations better than control group by taking active coping style. The officers, servicemen older than 21 years, and servicemen with more service duration than 3 years could usually take active coping style.

9.Comparison of vasodilatation effect between quercetin and rutin in the isolated rat thoracic aorta.

OBJECTIVETo determine the possible difference in vasodialtation effect of quercetin and rutin.
METHODSThe isolated rat thoracic aorta was treated with phenylephrine (PE), and the effects of quercetin and rutin on the preconstricted aorta rings with or without endothelium were determined by organ bath technique. Nitric oxide synthase inhibitor L-N(G)-nitroarginine methyl-ester (L-NAME), guanylyl cyclase inhibitor methylene blue, cyclooxygenase inhibitor indomethacin were used to explore the mechanism.
RESULTSQuercetin (10-160 micromol/L) caused vasorelaxation of aorta rings preconstricted with PE in endothelium-intact and denuded aorta rings in a dose-dependent manner. Rutin(10-160 micromol/L) caused dose-dependent vasorelaxation in endothelium-intact rings preconstricted with phenylephrine, but not in denuded aorta rings. The maximal response (Rmax) values calculated from vasorelaxation curves of quercetin and rutin were (77.20+/-6.11)% and (44.28+/-7.48)%, respectively. There was no difference between median effective concentration (EC(50)) values of quercetin and rutin. Pretreatment with L-NAME (0.1 mmol/L) abolished the vasorelaxation by rutin,but did not influence the vasodilating effect of quercetin in endothelium-intact rings. Pretreatment with methylene blue (10 mmol/L) canceled the vasorelaxation both by quercetin and rutin. Pretreatment with indomethacin (10 micromol/L) attenuated the vasodilatation of quercetin, but did not affect the vascular effect of rutin.
CONCLUSIONThe vasodilatation effect of quercetin is more potent than rutin. The vasodilatation effect of quercetin might be mediated by guanylyl cyclase and cyclooxygenase-dependent pathway, while the vasodilatation by rutin might be via nitric oxide-guanylyl cyclase pathway.

10.High-dose daidzein affects growth and development of reproductive organs in male rats.

OBJECTIVETo explore the effect of the phytoestrogen daidzein on the growth and development of the testis and epididymis in male SD rats.
METHODSThirty 10-week old (early adult) and 30 4-week old (pubertal) male SD rats were included in the study, each age group equally divided into 5 subgroups: normal control, positive control, low-dose, medium-dose and high-dose. The normal and positive control groups were given 1 ml distilled water and the same amount of distilled water containing diethylstilbestrol (DES) at the dose of 0.1 mg/kg, and the low-, medium- and high-dose groups administered daidzein in the dose of 2mg/kg, 20 mg/ kg and 100 mg/kg, respectively, all by gavage for 90 days. Observations were made on the changes in body weight and testicular and epididymal indexes, as well as on the structural changes of the testis and epididymis by H&E staining.
RESULTSThe early adult rats showed no significant differences in body weight and testicular and epididymal indexes between the claiclzein groups and the control (P > 0.05), nor did the pubertal rats in epididymal index (P > 0.05). The testicular index differed significantly between the high-dose group (3.21 +/- 0.07) and the normal control (3.71 +/- 0.45) (P < 0.05). The body weight reduced markedly in the high-dose group (P < 0.05), but with no significant differences between the normal control and the other two dose groups (P > 0.05). No obvious changes were observed in epididymal morphology in all the daidzein groups of the early adult and pubertal rats, but high-dose daidzein resulted in smaller testes and impaired spermatogenesis.
CONCLUSIONThe phytoestrogen daidzein, administered in a high dose, could delay the growth and development of the testis and induce structural changes of testicular tissues in pubertal SD rats.
