1.Protective effects of 10-HDA combined with AA-2G on UVA-induced photodamage in fibroblasts

Objective To explore the protective effects of 10-HDA combined with AA-2G on UVA-induced photodamage in fibroblasts. Methods The primary cultured human skin fibroblasts were divided into three groups: blank control group, UVA irradiation group and 10-HDA+AA-2G group. The cell viability and cellular senescent state were analyzed using CCK-8 and senescence associated-β-galactosidase (SA-β-gal) staining, respectively. Fluorometric assays were performed to detect the formation of reactive oxygen species (ROS) in the cells. Results Compared with those in control group, the cell viability was decreased (P < 0.05), and the SA-β-gal positive cells and ROS level were significantly increased (P<0.01 for both) in UVA-irradiated group. Compared with those in UVA-irradiated group, the cell viability was significantly increased (P < 0.01), and the SA-β-gal positive cells and ROS level were significantly reduced (P < 0.05, P < 0.01, respectively) in the 10-HDA combined with AA-2G group. Conclusion Fibroblasts treated with 10-HDA and AA-2G are significantly protected from UVA-induced cytotoxicity, cellular senescence and ROS, indicating that 10-HDA combined with AA-2G has photoprotective effects. Therefore, 10-HDA combined with AA-2G may be a potential combination for the prevention and treatment of skin photoaging.
2.Effect of tension on collagen arrangement

Objective To study the effect of mechanical tension on collagen arrangement and illustrate the relationship between tissue architecture and tension properties. Methods Cell morphologies, orientation and collagen arrangement of fibroblasts cultured in three different types of collagen gels with variation of mechanical tension were observed by phase contrast photomicrographs, light microscopy and transmission electron microscopy. Expression and distribution of a-smooth muscle actin (α-SMA) were investigated by immunohistochemistry. Results Phase contrast photomicrographs, light microscopy and transmission electron microscopy showed high level of tension distributed anisotropically in the monolayer gels and the anchored collagen gels, with bipolar shape of the fibroblasts, obvious polarity, arrangement of exogenous collagen fibres parallel to the long axis of the fibroblasts, especially prominent in monolayer gels. Low level of tension distributed isotropically was observed in floating collagen gels, with stellate morphology and arrangement of exogenous collagen fibres in a reticular array. Immunofluorescence showed that fibroblasts expressed high level of α-SMA protein distributed along the long axis of fibroblasts in the monolayer gels and the anchored collagen gels, especially in former ones. In contrast, few expression of α-SMA protein was found in floating collagen gels. Cell morphologies and orientation, expression and distribution of α-SMA as well as collagen arrangement of fibroblasts in the monolayer gels and the anchored collagen gels were similar to those in granulation tissue, whereas floating collagen gels resembled normal dermis or remodelled tissues. Conclusions Tissue architecture or morphology of the dermis are corresponding to tension proporties. Different tissue architectures are closely correlated with particular tension proporties.
3.Analysis on the long-term effects of modified double endobutton technique in the treatment of Tossy type III acromioclavicular joint dislocations.

OBJECTIVETo investigate the long-term clinical effects of modified double Endobutton technique for the treatment of acromioclavicular joint dislocations of Tossy type III.
METHODSA retrospective study was done in 42 patients with acromioclavicular joint dislocations of Tossy type III treated with modified double Endobutton technique from December 2008 to December 2010. There were 24 males and 18 females, ranging in age from 21 to 56 years old (averaged, 32.5 years old). All the patients were treated with open reduction, coracoclavicular ligament reconstruction using double Endobutton technique, and repair of acromioclavicular ligament. The Karlsson system was used to evaluate therapeutic effects. The distance from coracoid to clavicle was measured to evaluate reduction loss.
RESULTSAll the patients were followed up, and the duration ranged from 2.0 to 3.2 years (averaged,2.4 years). According to Karlsson system, 32 patients got an A degree and 10 patients got a B degree at three months post-operatively; 26 patients got an A degree and 16 patients got a B degree at the latest follow-up; 6 patients got an A degree at 3 months after operation lowered to B degree at the latest follow-up. The coracoid-clavicle distance increased from (26.91 +/- 0.91) mm at 3 months after operation to (27.41 +/- 1.10) mm at the latest follow-up. Te patients treated with over-reduction during operation or with heavy physical labour work after operation had obvious widened coracoid-clavicle distance. Bone absorption was found around the plate in most cases, mainly in the clavicular side.
CONCLUSIONTreatment for acromioclavicular joint dislocations of Tossy type III with modified double Endobutton technique has satisfactory early clinical results. But with time passing, loss of reduction and bone absorption around the plate could be observed, and clinical outcomes of some cases downgrade during the long-term follow-up.

4.Simultaneous Determination of Three Components of Puma Nose Drops by RP-HPLC

OBJECTIVE:To establish a RP-HPLC method for simultaneous determination of chlorapheniramine maleate,furacilin and ephedrine hydrochloride in Puma nose drops.METHODS:The analysis was carried on a XDB C 8 column;the mo?bile phase was composed of methanol(A),acetonitrile(B)and0.02mol/L potassium dihydrogen phosphate solutions(containing0.2%triethylamine and adjusted to pH3.0with phosphoric acid,C)with linear gradient elution(0min~3.5min,A∶B∶C=6∶13∶18,8.5min,A∶B∶C=6∶30∶64)and the flow rate was1.0ml/min;the detection wavelength was254nm and the column temperature was30℃.Chloramphenicol was used as the internal standard.RESULTS:The linear ranges were0.04~0.20mg/ml for chlorapheniramine maleate,0.02~0.10mg/ml for furacilin,0.50~2.50mg/ml for ephedrine hydrochlo?ride.The average recoveries were99.44%(RSD=0.48%,n=3)for chlorapheniramine maleate,101.36%(RSD=0.41%,n=3)for furacilin and99.43%(RSD=0.59%,n=3)for ephedrine hydrochloride.CONCLUSION:The method is reliable,accurate and suitable for quality control of Puma nose drops.
5.An Ethical Discussion on Special Population during the Standard Diagnosis and Treatment of STDs

To discuss the ethical factors influencing the standard diagnosis and treatment of sexual transmitted diseases(STDs) on special population including senile patients,pregnant and lying-in women and children.The senile patients with STDs had a decline of physiological function and might have foundational diseases,some of these patients had the psychological characteristics such as unclear talk,uncertain memory,stronger self-esteem,and had a decline of life quality.Therefore hte main ethical measures during the senile patients with STDs should include considering the pathophysiological and psychological characteristics,adopting individual diagnosis and treatment,making the skill of inquiring and physical examination,and attaching importance to the improvement of life quality.The main ethical measures during the pregnant and lying-in women with STDs should include attaching importance to the marriage examination and pregnant examination,finding potential STDs as soon as possible,and weighing potential medical risk.The main ethical measures during the children with STDs should include paying attention to the infection of family members or sex-aggression,making the protective medicine,doing well the propaganda and education,and making the children away from STDs.
6.High-dose intravenous immunoglobulin in the treatment of some severe skin diseases: a clinical analysis

Objective To analyze the therapeutic outcomes and adverse effects of high-dose intravenous immunoglobulin (hd-IVIg) in the treatment of some severe skin diseases (toxic epidermal necrolysis, drug hypersensitivity syndrome, connective tissue disease, autoimmune bullous disease, acute graft-versus-host disease). Methods Twenty-five cases of severe skin diseases were treated with hd-IVIg (0.4 g/kg/day for a course of 5 days). Results The therapeutic outcomes were different from each other. Of all the cases, 21 improved, especially those of acute onset of toxic epidermal necrolysis and drug hypersensitivity syndrome; 1 adult dermatomyositis and 2 elder bullous pemphigoid were not relieved. A patient with acute graft-versus-host disease died. Three patients presented with minor adverse effects (headache and blood pressure rising). Conclusions hd-IVIg is effective and safe in the treatment of some severe skin diseases. More importantly, it has a substantial effect on shortening disease course and decreasing the dosage of glucocorticoids and immunosuppressants as well as on preventing infections.
7.Intervention of empowerment with multi-stage change theory on diet behavior for the influence of metabolic indices in patients with diabetes

Objective To investigate the influence of different educational methods on the metabolic indices in patients with diabetes .Methods A total of 218 patients with diabetes in the First Affiliated Hospital of Guangxi Medical University were ran-domized divided into two groups .Different intervention ways were used :patients in the control group received conventional educa-tional methods ,while patients in experiment group received the educational methods combined with empowerment and multi-stage theory of change .The C-DES-SF scale scores and the HbA1c ,FPG ,2 h PG ,TC ,TG ,LDL ,HDL indices was compared between the two groups .Results Indices and empowered ability of HbA1c ,FPG ,2 h PG ,TC ,LDL were changed in accordance with time .As compared with control group ,the educational method in intervention group had better change of FPG ,2 h PG ,TC ,LDL(P< 0 .05) . Indices and empowered ability of HbA1c ,FPG ,2 h PG ,TC ,LDL had inter-action with time ,and indexes in intervention group were better than those in the control group(P< 0 .05) .At the 3rd and 6th month after invention in the intervention group ,patients in movement stage and movement maintaining stage were higher than those in control group(P< 0 .05) .Conclusion Well correction of bad diet behaviors by application of educational methods of combined with empowerment and multi-stage theory of change for the type 2 diabetes patients would help the patients to improve the ability of self-care and control the level of metabolic indices .

9.A preliminary study on the resistance mechanism of Ureaplasma urealyticum to erythromycin

Objective To study the resistance mechanism of Ureaplasma urealyticum (Uu) to erythromycin.Methods The susceptibility of 73 clinical isolates of Uu to erythromycin was evaluated by using broth dilution techniques. PCR and DNA sequencing were carried out to screen hot spot mutations at the variable region of 23S ribosomal RNA in erythromycin-resistant strains of Uu. Moreover, erythromycin resistance methylase genes (ermA, ermB, ermC) and efflux pump genes (mefA/E, msrA/B, mreA) were screened by using PCR with specific primers. Results There were 35 (47.95%) resistant Uu strains out of the 73 isolates, and the minimal inhibitory concentration varied from 8 to 32 mg/L among these resistant strains. The ermB gene was detected in 19 (54.29%) resistant strains, and msrA/B gene in 9 (25.71%) resistant strains. Two resistant strains harbored both ermB gene and msrA/B gene. No mutation at 23S ribosomal RNA or amplification of resistance-associated genes was noted in sensitive or reference strains of Uu. Conclusion The ermB and msrA/B genes may be responsible for the erythromycin resistance of Uu.
