Objective To approach a way to induce MSCs to dopaminergic neuron by mesencephalic conditional media and cytokines in vitro,and supply an ideal cells source for the treatment of Parkinson's disease.Methods The rat MSCs were isolated primarily from the femurs and tibias of the Wistar rats.MSCs were cultured,proliferated and purified by passage culture.Cultuered MSCs were divided into the control and the experimental group.In control group,MSCs were cultured without any induction medium.MSCs of experimental group were first cultured at medium containing bFGF for 24 hours.Then media were replaced with induction media which contained the agents as follows,respectively:GM_1,GDNF, GDNF+GM_1,GDNF+GM_1+mesencephalic conditional media.The surface markers of the differentiated neuron,such as NSE and TH were detected by immunocytoehemistry after MSCs were cultured in induction media for 3 and 7 days.Results In control groups,the NSE expression of MSCs was very lower than experimental groups.The percentage of NSE-positive cells of GDNF+GM_1+mesencephalic conditional media group in 7 day((45.257?5.999)/HP)was significantly more than other groups(control group is 2.214?0.779,GM_1 group is 22.014?3.624,GDNF group is 31.345?2.850,GDNF?GM_1 group is 40.314?4.203,P
2.Clone and Expression of Mat Peptide Gene of 47 kD Pro tein in Orientia Tsutsugamushi Karp Strain

Objective To know the expression of 47 kD protein of orientia tsutsugamushi(Ot) Karp strain and test its immunogenicity and antigenicity.Methods Forty-seven kD protein gene that code the whole mat peptide was amplified from the genomic DNA of Ot.Karp strain by PCR and constructed to a recombinant plasmid pGEX-47.The E. coli cells BL21 were transformed with pGEX-47,then the transformants were induced to express the recombinant 47 kD protein by IPTG. The antigenicity and immunogeni-(city) were identified by Dot Blot method and the experiment of immunizing mice respectively.Results 1. The PCR product of 47 kD protein gene of Ot.Karp strain was obtained and its sequence was the same as the published 47 kD protein gene.2. The E.coli expression plasmid pGEX-47 constructed was identified containing the 47 kD protein gene fragment by restrication analysis and PCR identification,and an expression band about 47 kD was detected by SDS-PAGE of the purified product from GST-fusson protein expressed by pGEX-47. 3.Dot Blot analysis and the experiment of immunizing mice testified its antigenicity and immunogenicity.Conclusions The 47 kD protein gene of Ot Karp strain is amplified that code the whole mat peptide, which can be expressed in prokaryotic cells and the expressed protein has antigenicity and immunogenicity.

4.Preparation and evaluation of monolithic molecularly imprinted stationary phase for S-naproxen

An S-naproxen (S-NAP) molecularly imprinted monolithic stationary phase (MIMSP) with specific recognition for S-NAP and naproxen (NAP) was prepared by in situ technique,utilizing 4-vinylpridine (4-VP) as a function monomer,ethylene glycol dimethacrylate (EDMA) as a cross-linking agent,and low-polar solvents (toluene and dodecanol) as porogenic solvents.The selectivity of the polymers for S-NAP and NAP was evaluated by high performance liquid chromatography (HPLC).The binding characteristics were tested by Scatchard analysis.Racemic NAP could be specifically separated to some extent.At the same time,NAP could be separated from ibuprofen under optimized conditions.Scatchard analysis showed that two classes of binding sites existed in the S-NAP-imprinted polymers,with their dissociation constants estimated to be 1.045 and 5.496 μM,respectively.The results demonstrate that S-NAP and NAP can be recognized specifically on the obtained MIMSP.
5.Studies on the Capacity of Phosphate Dissolving and the Identification of Strain BL-11

The morphology,cultural characteristics,physiological and biochemical properties of phosphate solubilizing strain BL-11 were studied.Combine with its 16S rDNA sequence analysis result strain BL-11 was identified as Brevibacillus laterosporus.Results showed that the phosphate solubilizing efficiency of strain BL-11was up to 10.91% when with Ca_ 3 (PO_ 3 )_ 2 as the sole phosphorus source in the medium,while the efficiency of that was up to 1.56% when with sands as the sole phosphorus source.An optimal media of BL-11 strain for solubilizing phosphate was obtained by orthogonal test.It was composed of sugar 20g/L,(NH_ 4 )_ 2 HCO_ 3 0.3g/L,MgSO_ 4 .7H_ 2 O 0.5g/L,NaCl 0.3{g/L},KCl 0.5g/L,FeSO_ 4 0.03g/L,MnSO_ 4 .H_ 2 O 0.03{g/L.}_ While other conditions were consisted of initial pH7.0-8.0,180 r/min,and 30℃.

Objective To promote the high quality nursing service,explore the measures and effects of the high quality nursing service.Methods Develop the nursing service model by the change of the stuff management,and improving the health education to patients.Results The degree of satisfaction from patients increased to 99% and positive service consciousness increased,and the rate of nursing chaperonage decreased to 60%.Conclusions Carrying out the high quality nursing service in rehabilitation ward could improve the nursing quality.

Objective To evaluate the diagnostic value of thick-slab single-shot turbo spin-echo (SSTSE)T2 ?-weighted sequence in magnetic resonance fetography (MRF)for fetal abnormalities.Methods 328 of 1 990 pregnant women with the diagnosis of fetal congenital defects on prenatal ultrasound screening or chromosome examination were randomly selected,and 338 fetuses were ob-tained.These fetuses were scanned by conventional magnetic resonance imaging (MRI)and MRF.The diagnostic results from the two MR methods were compared.Results Six hundred and twenty-four lesions were detected by MRF.The primary diagnosis based on conventional MRI was changed for 14 lesions (2.2%).New findings were identified for 48 fetal lesions (8.4%)and 66 ma-ternal lesions.However,78 fetal lesions could not to be identified by MRF.MRF could increase the diagnostic confidence for fetal lesions with high water content (56.1% of the lesions).Conclusion MRF can yield more precise information for fetal extremities, fluid-filled cavities,pathological hydrops and cystic lesions.As an additional aid to the conventional multi-slice T2-weighted se-quence.
10.Role of pulmonary stem cells labeled with bromodeoxyuridine and telomerase reverse transcriptase in hyperoxic lung injury in neonatal rats.

OBJECTIVETo investigate characteristics of pulmonary stem cells labeled with bromodeoxyuridine (Brdu) and telomerase reverse transcriptase (TERT) in lung tissue, as well as the effects of proliferation and differentiation of the stem cells on lung development and repair of pulmonary injury.
METHODSA model of hyperoxia in neonatal rats was made by exposing the rats to 95% O2 for 7 d. Before sacrificing the rats, Brdu was injected through peritoneum, and immune staining positive cells were analyzed after the rats were sacrificed. TERT positive cells were stained by an immunohistochemical method. At the same time, the double staining for surfactant protein C (SPC) and Brdu or SPC and TERT were performed. Lung histologic study was done on HE stained tissue slices.
RESULTS(1) The lung with hyperoxic injury had thinner walls of alveoli, simple alveolar structure, fewer and larger alveoli, expanded and shrunken alveoli, and there were many fell-off alveolar epithelial cells in the alveolar cavities as well. (2) The cells positively stained with Brdu located in septa, mucosa and submucosa of various bronchi, scattering in epithelium of bronchi, and the number of positive cells was low, having a large nucleus. The TERT-positive cells were apparent in the septa and alveolar walls of peripheral lung tissue, characterized by uneven distribution in the lung lobes, the number of positive cells was less than that of Brdu-positive cells [integral of expression (1.61 +/- 0.83) vs. (0.62 +/- 0.55), P < 0.05]. The number of Brdu- and TERT-positive cells had no significant difference in hyperoxic rats compared to that in controls [integral of expression (1.43 +/- 0.85) vs. (1.61 +/- 0.83); (0.62 +/- 0.55) vs. (0.83 +/- 0.84), P > 0.05]. (3) After double staining, a few positive cells were found in double-stained tissues with SPC and Brdu or TERT. (4) The cells positively stained with SPC antibody had different size. The percentage of positive cells was not significantly different between the hyperoxia group (80.3%) and control group (78.6%). The Brdu positive staining located in nucleus of cells that had larger size than the cells not stained, round nucleus with intense staining (seldom, pole-shaped) and the number of such cells was less than that of the SPC positive cells. The percentage of positive cells was not significantly different between the hyperoxia group (28.5%) and control group (21.4%). (5) The TERT staining located in nucleus of cell that had smaller size than the cells not stained, various nuclear shape, including round intensively stained, round slightly stained, pole-shaped and divided shape. The percentage of positive cells was not significantly different between the hyperoxia group (2.3%) and control group (1.5%).
CONCLUSIONS(1) Brdu and TERT, as markers of stem cells having different capability of differentiation, possess special characteristics, respectively. The cells with Brdu could be transit amplifying cell (TAC) which retains characteristics of stem cells originated from differentiated stem cells, while, the cells stained with TERT especially reflects the characteristics of stem cells. (2) The proliferation and differentiation of pulmonary stem cells during hypoxic lung injury are limited and may be related with arrest of alveolization.
