1.Characterization and comparison of interferon reference standards using UPLC-MS.

The study aims to characterize and compare interferon reference standards from 5 manufacturers. By testing molecular mass and trypsin-digested peptide mass mapping, the amino acid sequence was verified and post-translational modifications such as disulfide bond were identified. Results show that the molecular mass and amino acid sequence were consistent with theory; the disulfide bonds of 4 lots of interferon were Cys1-Cys98/Cys29-Cys138, 1 lot was Cys29-Cys139/Cys86-Cys99; N-terminal "+Met", acetyl N-terminal and Met oxidation were identified in part of the sample. UPLC-MS can be used to characterize and compare interferon reference standards from different manufacturers.

2.Changes of fast and slow components of brainstem auditory evoked potentials in the rat pups with hyperbilirubinemia.

OBJECTIVETo probe the changes of fast component of brainstem auditory evoked potentials (FC-BAEP), slow component of brainstem auditory evoked potentials (SC-BAEP) and the mitochondrial ultrastructures of the neurons in the brainstem in the rat pups with hyperbilirubinemia.
METHODS7 days old SD rat pups were randomly divided into control group (C, 17 rat pups) and two test groups (T1, 17 rat pups and T2, 17 rat pups). Bilirubin solutions (2 g/L) were injected into the abdominal cavity of the rat pups in the group T1 and T2 at the postnatal day 7 and 10. Six hours after the second injection, seven rat pups of each group were randomly selected to test serum bilirubin concentration via a micro-gauge. FC-BAEP and SC-BAEP were examined with an evoked potential recorder in the rest rat pups of each group at postnatal day 17 and 20. At the postnatal day 20, the endocardial perfusion was performed in these rat pups for the fixation of the brain, and then the brains were taken out. The cochlear nuclei were used for observation via electron microscope.
RESULTSSix hours after the injection of bilirubin solution at the postnatal day 10, the serum bilirubin concentrations of the rat pups in group T1 and T2 were increased significantly. Except for II-IV inter-peak latency(IPL), all the peak latency(PL) and IPL of FC-BAEP evoked via three sound stimulating rates (10/s, 40/s,80/s) at the postnatal day 17 prolonged significantly in the rat pups of group T1 and T2, and the PL in group T2 were much longer than that in group T1. Except for II-IV IPL of FC-BAEP evoked via sound stimulating rates of 10/s and 40/s, all the PL and IPL at the postnatal day 20 prolonged significantly in the rat pups of group T1 and T2. The PL of SC-BAEP evoked via sound stimulating rate of 10/s at the postnatal day 17 and 20 in the rat pups of group T1 and T2 prolonged significantly, and the PL at the postnatal day 17 in group T2 were much longer than that of group T1. The changes of mitochondria of the neurons in the cochlear nuclei at the postnatal day 20 in the rat pups of group T1 and T2 were characterized by swell, the slurred membranes, the broken crista and so on.
CONCLUSIONThere were the abnormal changes of FC-BAEP, SC-BAEP and the mitochondrial ultrastructures of the neurons in the brainstem in the rat pups with hyperbilirubinemia. The PL and IPL of FC-BAEP and SC-BAEP could be taken as the objective and sensitive indexes for early monitoring the bilirubin-induced hearing loss and brain injury.

3.Research progress of pharmacological activities and analytical methods for plant origin proteins.

As one of the important active components of traditional Chinese medicine (TCM), plant origin active proteins have many significant pharmacological functions. According to researches on the plant origin active proteins reported in recent years, pharmacological effects include anti-tumor, immune regulation, anti-oxidant, anti-pathogeny microorganism, anti-thrombus, as well as hypolipidemic and hypoglycemic activities of plant origin were reviewed, respectively. On the other hand, the analytical methods including chromatography, spectroscopy, electrophoresis and mass spectrometry for plant origin proteins analysis were also summarized. The main purpose of this paper is providing a reference for future development and application of plant active proteins.

4.Prognostic factors of penis-sparing surgery for early-stage penile cancer.

OBJECTIVETo investigate the factors influencing the prognosis of penis-sparing surgery (PSS) for early-stage penile cancer.
METHODSWe retrospectively studied the clinical data about 45 cases of early-stage penile cancer treated by PSS from January 2007 to December 2014. We calculated the rate of local recurrence-free survival by the Kaplan-Meier method, and conducted univariate and multivariate COX regression analyses on the relevant factors including the patient's age, marital status, tumor location, tumor size, postoperative sexual life, histological grade, and TNM stage.
RESULTSOne-year and three-year local recurrence-free survival rates were 95.5% and 52.2%, respectively. Multivariate analysis demonstrated that the histological grade (P = 0.039) and postoperative sexual life (P = 0.049) were independent factors for the prognosis of PSS. Logistic regression showed the patients age to be significantly associated with histological grade (P = 0.014).
CONCLUSIONHistological grade and postoperative sexual life are important independent prognostic factors of PSS for early-stage penile cancer, and the patients age is associated with the prognosis of PSS through its influence on the tumor grade.

5.Analysis of highly cited papers published in Chinese Journal of Endemiology during 2000-2006

Objective To analysis the inherent quality of highly cited papers published in Chinese Journal of Endemiology, so as to identify the direction of invitation for contributions, find high quality articles, and to improve the journal's core competitiveness. Methods Scientific papers published in Chinese Journal of Endemiology from 2000 to 2006, with citation rate equal to or higher than 20 times were retrieved in China National Knowledge Infrastructure(CNKI) by December 27, 2010. These articles were statistically analyzed according to publication year, section type, author, and geographical distribution of units. Results A total of 68 highly cited papers were obtained. The citation frequency of the highly cited papers ranged from 20 to 94, with an average of 31.09 times. The highly cited papers were published in 2000 at most, accounting for 25.0%(17/68); followed by 2001,accounting for 20.6% (14/68); at least in 2006, accounting for 2.9% (2/68). Of the highly cited papers, experimental study articles was in the highest proportion of 30.9%(21/68); followed by field epidemiological investigation articles,accounting for 23.5%(16/68); editorial, review articles accounting for 10.3%(7/68) and 8.8%(6/68), respectively;short reports, case reports, meeting records etc were not highly cited papers. The number of highly cited papers in editorial, expert forums, academic contend accounted for higher proportion of total number in corresponding section types(7/19, 3/11, 3/7). Highly cited papers came from 13 provinces(municipalities), there were 10 authors contributed two or more highly cited papers, and 8 units contributed two or more highly cited papers. Conclusions Adequate space should be given to experimental research, field epidemiological investigation, editorial, expert forums andacademic contend articles due to their high rate of citation. More attention should be paid to further consolidate and expand excellent authors group, and to intensify the invitation for key issues from influential experts.
6.Survey on knowledge, attitude and practice about hypertention in elderly patients from communities of Shanghai Xuhui District

Objective To survey on the knowledge,attitude and practice about hypertention in elderly patients and its relation to blood pressure control.Methods A sampling survey on the knowledge,atitude and practice about hypertention were carried out in a target population of 47 572 elderly hypertensive patients from 5 communities in Xuhui District of Shanghai during May 2011 to July 2011.Total 3800 questionairres were distributed and 3328 responded with a response rate of 87.5%.Results Among 3328 responders,3036 were aged above 65 years with a mean age of (76 ±7) years and 43.3% were males.The rates of knowledge about hypertension were 88.5%-97.8%.General practitioners in out-patient service were the major source of knowledge.Raults revealed that 81.9% had received health education before;81.8% requied a regular health education; 67.2% had self-tested blood pressure weekly; 89.6% controlled salt intake; 66.8% attended outdoor activities weekly with (5.6 ±2.1) times a week and (45 ±28) min each time.The blood pressure control rate in uncomplicated hypertensive patients was higher than those complicated with diabetes,coronary heart disease or stroke (62.3 % vs.17.0%,P < 0.01).Knowing high blood pressure was related to salt intake (OR =3.364,P < 0.01),hoping a regular health education about hypertension (OR =1.718,P < 0.01) were factors contributing to blood pressure control in uncomplicated hypertensive patients; while self-testing blood pressure once or twice a month(OR =2.572,P < 0.01)was contributing factor to blood pressure control in complicated hypertensive patients.Conclusions Elderly hypertensive patients from communities in Shanghai have enough knowledge,positive attitude but lack practices.General practitioners should carry out personalized and stronger health education focued on behavioral changes for those patients.

Objective Previous studies indicated that activation of Toll-like receptor4 (TLR4) was involved in the progression and instability of atherosclerotic plaque.Anti-inflammatory effects were shown in statins. However,the mechanisms underlying these effects have not been well explored.We test the hypothesis that a por- tion of these anti-inflammatory effects are mediated by regulation of TLR4 expression.Methods One hundred twenty-one subjects (22 normal persons,17 patients with stable angina and 82 patients with ACS) were recruited. 41 patients with ACS were randomized to atorvastatin 10 mg/d or atorvastatin 40 mg/d on top of routine anti-anginal treatment.Serum level of hsCRP,blood lipids,TLR4 expression on CD_(14)~+ monocytes were measuered before and after one month treatment.TLR4 expression on CD_(14)~+ monocytes were quantified via flow-cytometry.Results hsCRP and TLR4 expression on CD_(14)~+ monocytes in patients with ACS were higher than patients with stable angina and normal persons(hsCRP,ACS:11.1?14.3 vs stable angina:2.5?2.7 mg/L vs normal:2.3?4.2 mg/L,P
8.Relation between recurrent hemorrhage and both ultrastructural features of vascular and expressions of angiopoietin 1,CD68 and matrix metalloproteinase-9 in intracranial cavernous hemangioma

Objective To investigate the relation between recurrent hemorrhage and both ultrastructural features of vascular and expressions of angiopoietin 1 (Ang-l),CD68 and matrix metalloproteinase-9 (MMP-9) in intracranial cavernous hemangioma.Methods Thirty-five intracranial cavernous hemangioma samples,35 hepatic cavernous malformation samples and 4 normal brain tissues were chosen in our study; the ultrastructural features of vascular were observed with optical microscope and electron microscope; immunohistochemical methods were used to analyze the expressions of Ang-1,CD68 and MMP-9.Results The vascular wall was very thin in intracranial eavemous hemangioma samples,only with thin monolayer of endothelial cells and adventitia,but without basal lamina and elastic layer; gap junctions were obviously observed between endothelial cells; basement membrane showed unintelligible,and a large number of high electron density of sediment was presented at the surrounding blood vessels.Tissues from intracranial cavernous hemangioma obviously expressed higher levels of CD68 and MMP-9 than the normal brain tissues (P<0.05); Rank correlation analysis indicated that the expressions of CD68 and MMP-9 had a high degree of positive correlation (r=0.870,P=0.000).Tissues from intracranial cavernous hemangioma expressed less Ang-1 than normal brain tissues without significant difference (P>0.05).Conclusion The vascular structural and functional defection and instability in patients with cavernous hemangioma provide favorable conditions for the happaning of recurrent hemorrhage; and high expressions of CD68 and MMP-9 are closely related with recurrent hemorrhage.
9.Detection of TERC gene amplification by fluorescence in-situ hybridization in cervical intraepithelial lesions.

OBJECTIVETo explore the feasibility and practical value of fluorescence in situ hybridization (FISH) detection of TERC gene amplification in cervical intraepithelial lesions (CIN) and squamous cell carcinoma (SCC).
METHODSTissue microarray was constructed to cover 150 cases of various cervical conditions, including 24 cases of normal cervical mucosa, 78 cases of CINs (CINI, 25 cases; CINII, 21 cases and CINIII, 32 cases) and 48 cases of SCC. FISH was used to detect TERC gene amplification.
RESULTSTERC gene amplification was detected in 8% (2/25) CINI, 47.6% (10/21) CINII, 71.9% (23/32) CINIII and 87.5% (42/48) SCC. There were significant differences among these groups (P < 0.05). The amplification rates of TERC gene in SCC, CINIII and CINII were significantly higher than those of normal cervical epithelium and CINI (P < 0.05). Significant differences were also observed among CINI and CINII, CINIII and SCC (P < 0.05), and between CINII and SCC (P < 0.05). There were no significant differences between normal cervical epithelium and CINI, CINII and CIN III, and between CINIII and SCC (P > 0.05). FISH detection of amplification of TERC gene in CINI and CINII-III demonstrated the following statistics: sensitivity of 62.3%, specificity of 92.0%, accuracy of 71.8%, positive and negative predictive values of 94.3% and 53.5%, respectively.
CONCLUSIONSFISH detection is a reliable method in detecting TERC gene amplification using paraffin tissue sections. When histological evaluation becomes difficult, TERC amplification detectable by FISH may offer a diagnostic distinction of CINI from CINII. Moreover, TERC amplification may be used as a biomarker in predicting CIN progression to invasive cancer.

10.Impact on IL-2 expression of lymphocytes in donors after G-CSF administration and its clinical significance.

In this study, to investigate the effect on expression of IL-2 in lymphocytes from bone marrow and peripheral blood of normal donors after they were mobilized by G-CSF in allo-BMT, 7 normal donors bone marrow and peripheral blood were harvested before and after G-CSF administration. The separated lymphocytes were measured by FCM after they were stained intracellularly by anti-IL-2, and their expressions of IL-2 were compared. The degree of aGVHD in patients after bone marrow transplantation was evaluated clinically, and it was compared with the status of aGVHD of 15 patients whose donors didn't receive G-CSF administration in our department, and 2 groups of patients are comparable in age, types of diseases and status of donors. The results showed that the expression of IL-2 in lymphocytes in 7 G-CSF mobilized donors decreased significantly after G-CSF administration and more severe aGVHD than grade II didn't develop in these recipient patients, and comparing with 15 patients received the bone marrow from donors who didn't receive G-CSF, the incidence of aGVHD decreased. It is suggested that the expression of IL-2 in lymphocytes was influenced by donors' G-CSF administration, and it is likely that thereby reduces the incidence of aGVHD in patients after BMT.
