3.Pittsburgh Sleep Quality Index Investigation of Depressive Disorder Patients in the East-region of Shenyang

ObjectiveTo investigate sleep disorder of patients with depression and find out the regularity.Methods78 depression cases in the east-region of Shenyang were evaluated by Pittsburgh Sleep Quality Index (PSQI) questionnaire and mental health symptom checklist-90 (SCL-90). The results were contrasted with normal control group.Results63 depression cases (80%) had sleep disorder (PSQI scores≥8) and were significantly higher than normal control group. The PSQI evaluation showed that sleep quality, enter-sleep time, sleep efficiency, sleep disorder, hypnotige drugs, function of daytime and total scores of PSQI of the depression group were significantly higher than that of normal control group (P<0.05~0.01). The SCL-90 test showed that scores of somatic, obsessive, depressive, anxiety, phobia, paranoid and mental-illness, etc. and total quota of bad-sleep quality group were significantly higher than that of the fine-sleep quality group (P<0.05~0.01).ConclusionDepression patients have significantly descent in sleep quality, latter is often closely related to the mental psychotic expressiveness of somatic, obsession, depression, anxiety, phobia, etc.
4.Long-term effect of pseudomonas aeruginosa preparation on advanced colorectal cancer during surgery

Objective: This study aimed to observe the long-term effect of Pseudomonas aeruginosa preparation used in peritoneal injection of advanced colorectal cancer patients during surgery. Methods: A total of 83 colorectal cancer patients who received surgery between September 2006 and March 2008 were enrolled in this study. The patients were divided into two groups. Palliative resection and a 10 ml P. aeruginosa peritoneal injection were performed in 30 of 83 patients in the treatment group. Simple palliative resection was conducted in the other 53 patients, which comprised the control group. Both groups were then treated by regular chemotherapy and radiotherapy. Results:The follow-up visit was completed in 79 of 83 patients, with a high follow-up rate of 95.2%. No significant difference was found in the five-year overall survival time between the two groups (P=0.403). However, the five-year median survival time in the control group was only 13.9 ± 2.14 months, whereas that in the treatment group was 17.2 ± 2.12 months. Conclusion: Within a short period, peritoneal injection of P. aeruginosa during surgery could confer certain survival advantages for advanced colorectal cancer patients. However, the long-term effect of this therapy remains unknown.

Background Diabetiretinopathy(DR) ione of the leading causeof blindness.To understand the risk factorof Dpathogenesiiimportanfothe prevention and treatmenof DR.Objective The purpose of thistudy wato survey the prevalence and relevanfactorof Damong diabetipatientaged ove45 yearold.Methodcross-sectional study wadesigned.physical examination waperformed on 7300 subjectfrom July,2010 to March,2011 athe FirsHospital of Shanxi Medical University,and 5012 of them received non-mydriatidigital funduphotography.Type 2 diabetimellituwadetermined in 574 subjects,and 480 casethaconformed to the criteriof the currenstudy were recruited.questionnaire survey wacarried ouin the 480 patients,and the prevalence of diabeteand Dwacalculated fothe analysiof the causative factorof DR.ResultGradable funduphotographwere available in 480 subjects,and 98 of them presented with Dcomplication aprevalence rate of 19.8%.The morbidity rateof Dwere 14.3%,19.4%,23.7% and 21.5% in the 45-55 yeargroup,>55-65 yeargroup,>65-75 yeargroup and >75 yeargroup,respectively,withousignificandifference among them (x2 =3.824,P=0.281).The incidence rateof Dwere 11.8%,13.2%,15.4%,27.0% and 62.5% in patientwith disease courseof lesthan 5 years,5-10 years,> 10-15 years,> 15-20 yearand ove20 years,respectively,with significandifference among differendisease duration(x2 =57.518,P =0.000).No significandifference waseen in the prevalence of Damong groupwith body weighindiceof <18.5,18.5-22.9,23.0-24.9 and ≥25.0 (16.7%,23.3%,16.8%,19.7%) (x2 =2.099,P =0.718) and differentreatmenmethod,such aoral antidiabetidrugs,insulin injection ocombination treatmen(18.8%,25.9%,19.8%,respectively) (x2 =1.477,P=0.478).The questionnaire survey showed close relevancy between gender,disease duration,fasting glucose concentration,family history of diabetes,and whethefunduexamination waperformed (P =0.025,0.000,0.001,0.003,0.039).ConclusionThe prevalence rate of Dwaassociated with the awarenesfodiabeteand treatmenmodes.Gender,disease duration,family history of diabetes,fasting glucose concentration and submission to eye examination are the independenrisk factorof DR.
8.Correlation Research between Pharmacological Method State and Natural Attribute of Chinese Herbs.

Pharmacological method state is a method to explain the principle of Chinese herb according to its external phenomenon such as shape, color, texture, features, and so on. The natural attribute of Chinese herb include shape, color, texture, smell, harvesting time, medicinal parts, chemical components, and so on. Though both of them have different key points, the natural attribute of Chinese herb can also be used to explain its medicinal mechanism. Therefore, the correlation research between pharmacological method state and the natural attribute of Chinese herb has some significance.

9.Effects of ketamine on anoxia-reoxygenation induced glutamate release from cerebral cortex neurons of fetal rats

Objective To investigate the effects of ketamine on anoxia-reoxygenation(A/R)induced glutamate release from cerebral cortex neurons.Methods Primary cultured neurons obtained from cerebral cortex of fetal Wistar rats(16-18 d)were randomly divided into 3 groups:Ⅰcontrol group;ⅡA/R group andⅢketamine pretreatment+I/R group.The control group was not subjected to A/R while A/R group was exposed to anoxic air(95% N_2+5% CO_2)for 5 h followed by 24 h reoxygenation.In groupⅢdifferent doses of ketamine were added to the culture media before anoxia and the final ketamine concentrations were 1,20 and 100?mol?L~(-1) respectively.The extracellular glutamate concentration was detected at the end of 24 h reoxygenation.Results The extracellular glutamate concentration was significantly higher after 24 h reoxygenation in A/R group than in control group.Ketamine 20 and 100?mol?L~(-1) significantly inhibited glutamate release from the neurons induced by A/R in a dose-dependent manner.Conclusion Ketamine can inhibit glutamate release from neurons induced by A/R in a dose-dependent manner.
10.Cloning and Prokaryotic Expression of Human Recombinant Calreticulin

Objective: Clone, express and purify human recombinant calreticulin (CRT). Methods: Human CRT cDNA was amplified from total RNA of human lung cancer cell line A549 cells by RT-PCR. Then, PCR product was subcloned into prokaryotic expression vector pET-15b. After sequencing, this recombinant plasmid was transformed into E.coli. Rossetta. Recombinant CRT was expressed in host cells by IPTG induction. Resulted protein was purified by Ni-NTA resin under denature condition and dialyzed to recover its native structure. SDS-PAGE and Western blot method were used to identify the expression and purification of reconbinant CRT. Results: Human CRT cDNA was cloned from total RNA of A549 cells. CRT prokaryotic expression vector pET-15b-crt was constructed. Reconbinant CRT was induced to express in E.coli and purified by Ni-NTA affinity chromatograph. Conclusion: A method for prokaryotic expression and purification of human recombinant CRT was successfully established. This method laid a foundation for the subsequent CRT research.
