1.Expression of the pituitary transcription activator-1 gene in human pituitary adenomas

Objective To in ve stigate quantitatively the expression of pituitary transcription activator (Pit) -1 mRNA in different types of human pituitary adenomas by RT-PCR. Methods Samples were obtained from 35 patients who had unde rgone therapeutic surgery for pituitary adenomas. The type of pituitary adenomas was determined according to serum hormone levels and clinical features, and the grade and the stage of pituitary adenomas were depended on imaging and operatio n observation. Expression of Pit-1 mRNA of pituitary adenomas was assayed by RT -PCR. Results Thirty five patients with pituit ary adenomas consisted of 13 cases with PRL adenomas, 6 cases with GH adenomas, 2 cases with GH/PRL mixed adenomas, 11 cases with clinically non-functioning ad enomas and 3 cases of ACTH adenomas. Pit-1 mRNA was expressed in all PRL, GH an d GH-PRL mixed adenomas and in some non-functioning adenomas (9/11). No Pit-1 mRNA expression was found in ACTH adenomas. Objective To investigate the expression of estrogen receptor (ER), progesterone recptor (PR) and c-erbB-2 in breast cancer tissues and its relationship with prognostic factors. Methods The clinical data of 1 008 cases with breast cancer were retrospectively analysed. The relationship between the expression of ER, PR and c-erbB-2 in breast cancer tissues and the prognostic factors such as menopausal status, axillary lymph nodes metastasis and TNM stage was investigated. Results The mean presentation age of the 1 008 cases was (53.53?12.60) years. Invasive ductal cancer was the most common pathological type (80.5%). Objective To examine the expressions of estrogen receptor α (ERα), estrogen receptor β (ERβ) and pituitary-specific truncated estrogen receptor product-1 (TERP-1) in diethylstilbestrol (DES) induced prolactinoma of rats. Methods Ovariectomised Wistar rats were subcutaneously implanted with an implant containing 20 mg DES. Rats were implanted with a blank implant as controls. Eight weeks later, serum prolactin level of rats was measured by RIA. Prolactin immunoreactive cells of anterior pituitary were detected by immunohistochemistry assay. ERα, ERβ and TERP-1 mRNAs in pituitaries were examined by RT-PCR. Results Serum prolactin levels, pituitary weights and pituitary prolactin immunoreactive cell countings in experimental group rats were all obviously higher than those in control group (P<0.001 respectively). ERα, ERβ and TERP-1 mRNAs were all transcripted in DES induced rat prolactinomas. ERα and TERP-1 mRNA levels were significantly higher in experimental group than those in control group (P<0.001 and P<0.05). Conclusion Estrogen plays a direct role in regulating pituitary prolactin cells through estrogen receptors. Further functional studies will be required to determine whether coexpression of ER variants along with normal ER confers a pathophysiological role in pituitary hormone regulation and (or) tumor cell proliferation.

6.The clinical and laboratory characteristics of male patients with antiphospholipid syndrome

Objective To investigate the clinical and laboratory features of male patients with anti-phospholipid syndrome (APS).Methods Thirty-three male APS patients admitted to Peking University People's Hospital from January 2003 to April 2015 were enrolled.One hundred and fourteen female APS patients were selected as controls.Female patients with thrombosis were analyzed.x2 test, Fisher's exact test and t test were used for statistical analysis.Results Among male patients, 39.4% were primary APS and 60.6% were secondary APS.Compare with female patients,the prevalence of venous thrombosis was significantly higher than that in male APS (72.7% vs 29.8%,x2=19.719, P=0.000).The male APS patients experienced more episodes of thrombosis (75.8% vs 22.8%, x2=31.669, P=0.000) and not less than two locations of thrombosis than female (69.7% vs 22.8%, x2=25.321, P=0.000), there was still significant difference between male patients and female patients with thrombosis.There was no difference of antiphopholipid antibodies between the two groups.Conclusion Male patients with antiphospholipid syndrome are more prone to have thrombosis.More clinical interventions should be implemented in male patients with APS.
7.Analysis on medication principles for cough based on experience of Xu Di-hua, descendant of Meng He Medical School.

Based on the software of traditional Chinese medicine inheritance support system (TCMISS), this article aims to analyze the experience and composition rules for cough from the descendant of Meng He Medical School, Xu Di-hua. The cough cases treated by Xu Di-hua were collected, and recorded into TCMISS (V2.0). Data mining methods such as Apriori algorithm and complex system entropy cluster were used to analyze the medication principles of Xu Di-hua for cough from pathogenesis and therapeutie aspects, and dig out the frequency of the herbs in prescription, core medicine and new combinations. The experience of curing cough from Professor Xu Di-hua were well found in the research. He is good at choosing prescriptions accurately, and pays attention to simultaneous use of cold and moisture drugs with combination of tonification and purgation. He is skilled in adding or reducing materia medica flexibly, as well as regulating lung to relieve cough and eliminating phlegm by clearing heat.

8.Spontaneous regression of a giant basilar artery aneurysm in a young adult after surgical injury:case report and literature review

A giant basilar artery aneurysm of young woman with endocrine disturbance was misdiagnosed as a large pituitary adenoma and treated surgically via a trans-sphenoidal approach was planned.But the neurosurgery was finally aborted because of massive bleeding during the procedure. One year later, a cerebral angiography confirmed this basilar artery aneurysm was obviously regressed and then endovascular coiling was successfully performed. No neurological complication occurred post-procedure and the endocrine dysfunction symptom was obviously improved.
10.Expression of hypoxia inducible factor-1a and vascular endothelial growth factor in retina during rat embryon development

Background Research determined that hypoxia inducible factor-1(HIF-1)is associated with the hypoxia response in normal organ,and it plays an important role during the embryon development.It is also proved that the expression of vascular endothelial growth factor(VEGF)is upregulated in embryon retina.But,whether the action of HIF-1 and VEGF is correlated is unclear. Objective The aim of this study was to investigate the dynamic change of expression of HIF-1α and VEGF in retina during embryonic development and explore the role of HIF-1α and VEGF along with retinal evolvement.Methods The different embryon ages of SD rats were obtained from 30 clean pregnant SD rats by cesarean surgery.The retinas were isolated from embryon 10一day,12一day,14-day,16-day and 20-day rats respectively.The expressions of HIF-1 α and VEGF protein in retina were semi-quantitatively and qualitatively determined by immunochemistry,and the expressions of HIF-l α and VEGF mRNA in retina with different-embryon-phase and adult rats were detected by RT-PCR. Results The hishly level of expression of HIF-1 α and VEGF protein were found in the cellular nuclei and cytoplasm of retina in embryon 10一and 12-day rats.With the increase of embryon age.the expression of HIF-1α and VEGF protein in retina lowed(F=56.70,P<0.01;F=60.78.P<0.01).Compared with the adult rats,the expressions of HIF-1α and VEGF protein in retina were higher from embryon 0-day through 20-day rats(all P<0.01).The significantly positive correlation was found in the expression level between HIF-1α and VEGF protein throughout the experimental duration(r=0.96,P=0.00).Followed the same pattern,the expression levels of HIF-1α and VEGF mRNA in retinas were also enhanced in the embryon 10- and 12-day rats.Identically,a gradually weakened trend was seen in the expressions of HIF-1 α and VEGF mRNA in retinas as the increase of gestational age of rats(F=68.84,P<0.01;F=96.49,P<0.01),and the exDressions of HIF-1 α and VEGF mRNA decreased in the adult rats compared with different embryon-phase rats,showing a statistically significant difference between them(all P<0.01). Conclusion The expressions of HIF-1 αand VEGF in retina appear a dynamic alteration from hish to low during the embryonic development of rat retina,indicating that HIF-1α/VEGF pathway might participate in the process of embryo development of rat retina.
