Objective To investigate the relationship between anti-lymphocyte antibody (ALA) and clinical and immunological parameters in primary Sj(o)gren syndrome (pSS).The significance of ALA and lymphopenia in pSS was explored.Methods Serum samples were obtained from 143 patients with pSS.Clinical and laboratory data were collected.Sera from 45 healthy individuals were collected as the controls.Serum ALA IgG was detected by indirect immunofluorescence test (IIFT).x2 test and t test were used for statistical analysis.The reasons of lymphopenia in pSS patients were analyzed.The correlation between ALA,lymphopenia and clinical manifestations as well as laboratory parameters was analyzed.Results ① Of the 143 pSS patients,25.2％(36/143) were serological positive for ALA,which was significantly higher than the normal group (6.5％,5/77) (x2=27.496,P＜O.01).② The prevalence of leukopenia (55.6％,20/36),lymphopenia (77.8％,28/36),anti-nuclear antibody(80.5％,29/36) and C3 decrease(50.0％,18/36) was higher in pSS patients with ALA than those without (x2=8.296,25.348,3.929,3.866,P＜0.05).③ 46.2％(66/143) patients had lymphopenia.Comparing with patients with normal lymphocyte count,the incidence of ALA positive,leukopenia,PLT decrease and anti-SSB antibodies positivity was significantly higher in patients with lymphopenia (x2=27.496,51.344,5.320,8.950,7.782; P＜0.05).Conclusion The ALA is likely to be a meaningful sera marker for pSS patients with lymphopenia,and this suggests that it has an important role in the clinical progress of SS.

Aim To investigate the effects of isoliquiritigenin(ISL) on cognition dysfunction and brain energy metabolism following repeated cerebral ischemia-reperfusion in mice.Methods Mouse models of learning and memory deficits were made by repeated occlusion of bilateral common carotid arteries and reperfusion. Step-down test and Y-maze task were used to examine the learning and memory abilities of mice. The brain energy state was analyzed by reversed-phase high performance liquid chromatogra phy. Results ISL 10, 20 and 40 mg?kg -1 ig reduced the n umber of errors and extended the latency of memory in step-down test, decreased the number of cumulated stimulation and increased the number of right choices i n Y-maze task. ISL(10, 20, 40 mg?kg -1) increased the levels of ATP, ener gy charge (EC) and total adenine nucleotides(TAN) in a dose-dependent manner. Conclusion The results suggest that ISL improves cognition dysfun ction induced by repeated cerebral ischemia-reperfusion in mice, which may be r elated to the improvement of brain energy metabolism.

OBJECTIVETo observe the effect of American Ginseng Capsule (AGC) on the liver oxidative injury and the Nrf2 protein expression in the liver tissue of rats exposed by 900 MHz cell phone electromagnetic radiation.

METHODSTotally 40 male SD rats were randomly divided into the normal control group, the model group, the Shuifei Jibin Capsule (SJC) group, and the AGC group,10 in each group. Rats in the normal control group were not irradiated. Rats in the rest three groups were exposed by imitated 900 MHz cellular phone for 4 h in 12 consecutive days. Meanwhile, rats in the SJC group and the AGC group were intragastrically administrated with suspension of SJC and AGC (1 mL/200 g body weight) respectively. Normal saline was administered to rats in the normal control group and the model group. The histolomorphological changes of the liver tissue were observed by HE staining. Contents of malonic dialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), and glutathione peroxidase (GSH-PX)were detected by colorimetry. The Nrf2 protein expression of hepatocytes was detected by immunohistochemical assay and Western blot.

RESULTSCompared with the normal control group, hepatocyte nucleus was atrophied or partially disappeared, the contents of liver MDA and Nrf2 protein obviously increased (P <0. 05, P <0. 01); contents of liver SOD and GSH decreased (P <0. 05) in the model group. Compared with the model group, karyopyknosis was obviously attenuated and approached to the normal level in the SJC group and the AGC group. The contents of liver MDA and Nrf2 protein expression decreased (P <0. 05), and the contents of liver SOD, GSH, and GSH-PX obviously increased (P < 0.05) in the SJC group. The contents of liver MDA and the Nrf2 protein expression decreased (P < 0.05), and contents of SOD and GSH obviously increased in the AGC group (P <0.01, P <0.05).

CONCLUSIONSThe electromagnetic radiation induced by 900 MHz cell phone could affect the expression of Nrf2 protein, induce oxidative injury, and induce abnormal morphology of liver cells. SJC and AGC could promote the morphological recovery of the liver cells. Its mechanism might be related to affecting the expression of Nrf2 protein and attenuating oxidative damage of liver cells.

Objective To study clinical features of liver damage in primary Sj(o)gren syndrome (pSS) and its related factors.Methods One hundred and forty-nine patients of confirmed pSS hospitalized at Peking University People's Hospital were analyzed retrospectively.Results Seventeen of 149 patients of pSS (11.4%) showed liver damage,10 diagnosed as autoimmune hepatitis and seven as primary biliary liver cirrhosis.Increased serum levels of IgG,IgM and γ-globulin accounted for 88.2% and 50.8%,35.3% and 5.3%,and 94.1% and 47.7% of those complicated with and without liver damage,respectively,with statistically significant difference (P ＜ 0.05).Positive serum anti-mitochondria antibody subtype M2 (AMA-M2) was 35.3% in pSS patients with liver damage,significantly higher than that in those without liver damage (1.5%) (P ＜ 0.05).Independent risk factors significantly associated with liver damage included age (OR = 1.013,95% CI 0.971-1.058),course of illness (OR = 1.089,95% CI 1.032-1.150),serum level of γ-globulin (OR = 4.021,95% CI 1.246-12.982),positive AMA-M2 (OR = 11.82,95% CI 0.005-0.157),and positive anti-SSA (OR = 101.333,95% CI 12.653-811.560).Conclusions Liver damage occurred rather high in pSS patients and increased serum levels of IgG,IgM,γ-globulin and anti-SSA can be used to predict their complication with liver damage.Age,course of illness,serum level of γ-globulin,positive AMA-M2 and positive anti-SSA are all significantly associated with liver damage in pSS patients.

Objective To investigate the significance and distribution of lymphopenia and antilymphocyte antibody(ALA) in systemic lupus erythematosus (SLE) and to explore the role of ALA in lympho penia.Methods One hundred and ten in-patients who were admitted during February 2003 to February 2008 were retrospectively reviewed.Indirect immunofluorescence test was used to detect ALA.Results ① Lymphopenia (＜1.5×109/L) was observed in 68.2% patients.Lymphopenia was associated with skin rashes,serositis,renal involvement,NPSLE,leucopenia,ANA,ds-DNA antibody,ESR and IgG levels.The lymphocyte count and SLEDAI scores was more closely correlated than total white blood cell counts.②ALA was positive in 51/110 (46.4%) patients.ALA was associated with renal involvement,NPSLE,WBC count,C3 level decrease,ANA,dsDNA antibody and SLEDAI scores.Conclusion Lymphopenia is significantly associated with SLEDAI scores.ALA is possibly one cause of lymphcytopenia.In addition,ALA is also a parameter for disease activity,and is associated with organ involvement and outcomes.

AAV-ITR gene expression mini vector is a double-strand or single-strand DNA that only contains inverted terminal repeats of adeno-associated virus, cis-elements and gene of interest and does not contain any other foreign DNA sequences. We prepared Bac-ITR-EGFP and Bac-inrep. Spodoptera frugiperda cells were infected with Bac-ITR-EGFP (P3) and Bac-inrep (P3). Up to 100 μg of AAV-ITR-EGFP gene expression mini vectors were extracted from 2 x 10(7) cells of Sf9 72 h after infection. The gel electrophoresis analysis shows that most forms of AAV-ITR-EGFP gene expression mini vector were monomer and dimer. The mini vector expression efficacy was examined in vitro with HEK 293T cells. The EGFP expression was observed at 24 h after transfection, and the positive ratio reached 65% at 48 h after transfection.
