2.Prevalence of Enterococcus faecalis in saliva and filled root canals of teeth associated with apical periodontitis.

To investigate the prevalence of Enterococcus faecalis in saliva and filled root canals of patients requiring endodontic retreatment for apical periodontitis. Patients with apical periodontitis who were referred for endodontic retreatment were examined. The type and quality of the restoration, symptoms, quality of obturation were recorded. During retreatment, an oral rinse sample and root canal sample were cultured using brain-heart infusion agar and bile esculinazide agar to select for E. faecalis. The 16S rRNA technique was used to identify E. faecalis. A total of 32 women and 22 men (mean age: 38 years; s.d.: 11 years) and 58 teeth were studied. The prevalence of E. faecalis was 19% in the saliva and 38% in the root canals. The odds that root canals harbored E. faecalis were increased if the saliva habored this bacterium (odds ratio=9.7; 95% confidence interval=1.8-51.6; P<0.05). Teeth with unsatisfactory root obturation had more cultivable bacterial species in root canals than teeth with satisfactory root obturation (P<0.05). E. faecalis is more common in root canals of teeth with apical periodontitis than in saliva. The prevalence of E. faecalis in root canals is associated with the presence of E. faecalis in saliva.

3.Effect of movie enjoyment on emotion of male patients with chronic schizophrenia

Objective To investigate the effect of movie enjoyment on emotion of male patients with chronic schizophrenia.Methods A total of 62 standard patients with chronic schizophrenia from September to November 2011were selected and randomly divided into the experimental group(30) and the control group(32) by using a random number table.Patients in experimental group received movie enjoyment and routine rehabilitation project,while those in the control group received only routine rehabilitation project.Before the intervention and 12 weeks after the intervention,the patients were investigated with the Positive and Negative Affect Scale for efficacy assessments.Results After the nursing intervention,the scores of positive and negative affect were (30.79 ± 7.54) and (18.07 ± 5.97) respectively in the experimental group in contrast to (22.29 ± 8.14) and (18.00 ± 7.79) before the intervention,the difference was statistically significant (t =3.97,4.85,respectively;P <0.01).After the nursing intervention,the scores of positive and negative affect were(21.41 ±7.40) and (17.72 ± 7.03)respectively in the control group while were (23.25 ± 6.37)and (16.50 ± 6.08) before the intervention,the difference was statistically significant (t =2.67,1.98,respectively; P < 0.05).There was statistically significant between the two groups in the scores of positive and negative affect after intervention(t =5.69,6.78 ;P < 0.01).Conclusions The movie enjoyment can help improve positive emotions affect of patients with chronic schizophrenia.

5.Polypropylene mesh for testicular prothesis implantation: A report of 57 cases.

OBJECTIVETo search for an optimum method for testicular prothesis implantation in the treatment of testis loss.
METHODSWe retrospectively analyzed the surgical methods and outcomes of 53 cases of terminal prostate cancer and 4 cases of unilateral testicular torsion treated by implantation of testicular prothesis with the polypropylene mesh.
RESULTSThe 57 male patients all received testicular prothesis with the polypropylene mesh. All the patients were satisfied with the appearance and size of the scrotum after surgery. No scrotal hematoma, prosthesis infection, or autoimmune disease occurred postoperatively.
CONCLUSIONTestis loss is not a rare condition clinically, for the treatment of which surgical implantation of testicular prothesis with the polypropylene mesh can achieve both a fine tissue compatibility and a desirable scrotal appearance.

7.Protective effects of oxymatrine on adriamycin-induced cardiotoxicity in rabbits and its mechanism

Objective To establish the in vivo models of adriamycin(ADR)-induced cardiotoxicity in rabbits, investigate the protective effect of oxymatrine (OMT) on ADR-induced cardiotoxicity, and explore the possible mechanism. Methods Twenty-six rabbits were randomly divided into ADR group (n=8, 2 mg/kg ADR), OMT group (n=5, 10 mg/kg OMT), ADR + OMT group (n=8, 10 mg/kg OMT was injected 30 min before ADR injection) and saline group (n=5, same quantity of normal saline), and rabbits in each group were infused with medicine or normal saline through ear marginal vein once a week for 8 weeks. The apoptosis of myocardial cells was detected by TUNEL methods, and the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) was determined. Results After treatment, the body weight of ADR group was significantly lower than that of the other groups(P < 0.05), the activity of SOD and GSH-Px significantly decreased and the apoptosis index (AI) was significantly higher than that of the other groups(P <0.01). There were similar while minor changes in ADR + OMT group. There was no significant adverse effects in OMT group. Conclusion OMT protects heart from adriamycin-induced injury in rabbits, which may relate to the decrease in level of antioxidant and apoptosis of myocardial cells.
8.Triptolide-induced apoptosis by inactivating nuclear factor-kappa B apoptotic pathway in multiple myeloma in vitro.

The effect of triptolide on proliferation and apoptosis of human multiple myeloma RPMI-8226 cells in vitro, as well as the roles of nuclear factor-kappa B (NF-κB) and IκBα was investigated. The effect of tritptolide on the growth of RPMI-8226 cells was studied by MTT assay. Apoptosis was detected by Hoechest 33258 staining and Annexin V/PI double staining assay. The expression of NF-κB and IκBα was observed by Western blot and confocal microscopy. The results showed that triptolide inactivated NF-κB apoptotic pathway in human multiple myeloma RPMI-8226 cells. Triptolide at nM range induced proliferation inhibition in a dose- and time-dependent manner and apoptosis in a dose-dependent fashion in RPMI-8226 cells. Besides, we observed the inhibition of NF-κB /p65 in the nuclear fraction was correlated with the increase in the protein expression of IκBα in the cytosol. These results suggested that triptolide might exhibit its strong anti-tumor effects via inactivation of NF-κB/p65 and IκBα.
9.Magnetic resonance imaging of congenital fibrosis of extraocular muscle associated with limb movement disorder in a family

Background Congenital fibrosis of extraocular muscles (CFEOM) affects patient' s appearance and quality of life,and no effective treatment for this disease is available.Imaging study is helpful for exploring the pathogenesis of CFEOM.Objective This study was to describe the characteristics of CFEOM associated with limb movement disorder using magnetic resonance imaging (MRI).Methods A family with CFEOM associated with limb movement disorder was investigated in Renmin Hospital of Wuhan University.Disease history was collected and the pedigree was investigated.Ophthalmologic examinations,including corrected visual acuity,refractive error,slitlamp examination,ophthalmoscopic examination,force of levator palpebrae superioris,ocular movement,eye position,forced duction test,and bell phenomenon examination,were performed.Ocular orbital and cranial MRI was performed in 4 CFEOM patients and 10 normal subjects to compare the structures of the extraocular muscles,motor nerve and cranium.Oral informed consent was obtained from each patient prior to any medical examination.Results A total of 1 1 members from 3 generations were investigated in this study,presenting with 4 cases of disease.The mode of inheritance of this family complied with the Mendelian autosomal dominant inheritance law.Clinical signs included disturbance of eye movement,deviation of eye position,ptosis,lack of Bell sign and positive reaction of passive pull test.In addition,unstable gait,improper body limb alignment,dysphasia and mental retardation were ohserved in 1 patient,which coincided with the diagnostic criteria of type 3 CFEOM.MRI results demonstrated that the levator palpebrae superioris,superior rectus and superior oblique muscle were clearly thinner,and the medial rectus,lateral rectus,inferior rectus muscle were thinning in the patients,showing significant differences in comparison with the normal controls(P<O.05).The oculomotor and abducens nerves became thinner and even absent in the patients.Cranial MRI showed that Ⅲ-3 in the pedigree with callosum was shorter than that of the normal controls,suggesting that patient suffered from corpus callosum hypoplasia.Meanwhile,cranial MRI indicated the presence of cerebellar hypoplasia and the expansion of the fourth ventricle.Conclusions MRI demonstrates consistent abnormalities of the oculomotor nerves and abducens nerves in the affected individuals in this CFEOM 3 family,and some affected members exhibit two types of central nervous system abnormalities-corpus callosum and cerebellar hypoplasia.These findings suggest that CFEOM 3 is primarily a neuronal disease.
10.Enhancing Protective Immunity Effects of TPI DNA Vaccine against Schistosoma japonicum through Codon Optimization

Objective To study the protective effect of codon optimized TPI DNA vaccine against Schistosoma japonicum infection.Methods Sixty female BALB/c mice were randomly divided into 5 groups.The mice were injected through musculus quadriceps fexoris with 100 ?g pcDNA 3.1 control(Group A), pcDNA3.1-TPI(Group B), pcDNA 3.1-TPI-mHSP70(Group C), pcDNA3.1-TPI.opt(Group D), and pcDNA3.1-TPI.opt-mHSP70(Group E) respectively.All mice were immunized for three times with an interval of two weeks.The mice were challenged with(40?1) cercariae of S.japonicum per mouse by abdominal skin penetration 4 weeks after the last immunization, and sacrificed at 42 days post-challenge, the number of worms or hepatic eggs was counted.Blood was taken for the detection of IgG, IgG1, and IgG2a 2 days before immunization and before challenge, respectively.Spleen cells of 2 mice from each group were cultured and stimulated with ConA and rSjCTPI peptide, and the supernatant was collected for detection of IL-2, IL-4, IL-5, IFN-?, and TNF by flow cytometry.Results ELISA showed that the mice in groups B, C, D, and E produced specific IgG and IgG1, IgG2a antibody isotypes, and the ratio of IgG2a/IgG1 was 1.73, 2.06, 2.44, and 3.09, respectively.The levels of IL-2, IFN-? and TNF in groups D and E were higher than that of groups B and C.The worm reduction rate and hepatic egg reduction rate in groups D(36.03%, 41.7%) and E(39.03%, 46.85%) were higher than those of groups B(26.28%, 28.35%) and C(28.38%, 31.39%)(P
