[ ABSTRACT] AIM: To study the autophagy of prostate cancer PC-3 cells induced by CD147 in vitro.ME-THODS:Themethod of amino acid starvation to induce autophagy was used. The expression of CD147 was detected by Western blotting. To study the functional effects of CD147 on autophagy in prostate cancer PC-3 cells, the down-regulation of CD147 expression was induced by the technique of RNAi.The conversion of autophagic marker protein LC3-I to LC3-II was determined by Western blotting. The cell death after starvation-induced autophagy was analyzed by trypan blue exclu-sion assay.RESULTS:The CD147 expression gradually increased in starvation-induced autophagy. The down-regulation of CD147 significantly increased the expression of autophagy-related protein LC3-II compared with control group.Mean-while, the cell death rates increased from (19.3 ±3.1)%and (22.3 ±3.5)%in control groups to (38.4 ±3.1)%in si-lencing the expression of CD147 in the PC-3 cells (P<0.05).CONCLUSION:CD147 inhibits starvation-induced auto-phgy and autophagy death in the prostate cancer PC-3 cells.

Objective To investigate the distribution of neurons expressing Fos protein in central nervous system (CNS) following esophageal mucosal acid exposure, and to map the contribution of spe- cific brain areas in sensitizing responsivity and emphasize the coding change of CNS to the esophageal acid stimulation. Methods Thirty-six healthy Sprague-Dwley rats were randomly divided into five groups. Group A (n=6) was normal group of home cage control animals to which no stimulation was given. Group B (n=7) was saline group which received esophageal perfusion with normal saline solution (0.9% NaCl).Group C (n=8) was treated with esophageal mucosal acid exposure containing 0.1 mol/L HCl. Group D (n=7) was sensitized by ovalbumin.Group E (n=8) received basal ovalbumin-sensitization plus esophageal mucosal acid exposure. The rat model of esophageal visceral hypersensitivity was estab- lished by the basic ovalbumin-sensitization combined with intra-esophageal mucosal acid exposure. The neuronal expressions of c-fos proto-oncogene were detected with immunohistochemical counter-staining and computerized color image analyzer under various conditions. Results The rats in model group with basic ovalbumin-sensitization plus esophageal acid perfusion initiated a high density expression of c-fos- immunoreactive(Fos-IR) neurons in multineuronal networks.A significantly higher number of Fos positive neurons was found in the model group than those in the corresponding regions of other groups (P<0.05) in the following brain areas:frontal and parietal cortex, insular cortex, cingulated cortex, central amyg- daloid nucleus, the K(?)lliker-Fuse nucleus, the nucleus ambiguus, parabrachial nucleus, hypothalamic paraventricular nucleus, paraventricular thalamic nucleus, paratrigeminal nucleus, the nucleus of solitary tract, area postrema, reticular nucleus of medulla, whereas no significant difference was found in the dorsal motor nucleus of the vagus, supraoptic nucleus, periaqueductal gray matter or orbital part of infe- rior frontal gyrus. The values of Fos-IR neurons were also increased in the central amygdala,parabrachi- al nucleus, paraventricular nucleus, the paratrigeminal nucleus and NTS in the model group than that in the corresponding regions of other groups (P<0.05).Conclusion The basic ovalbumin-sensitization fa- cilitated dramatically the c-fos expression evoked by esophageal acid perfusion, suggesting that visceral hypersensitivity induced by ovalbumin may alter cortical reactivity processing of esophageal acid stimula- tion in brain areas.

Objective To investigate the possible mechanism of the gap junctional influence on the change in permeability of the blood-brain barrier(BBB)after reperfusion subsequent to cerebral ischemia. Methods In the test laser scanning confocal microscope(LSCM)was used to investigate the change of Cx43 levels and distribution. The MCAO/R model was induced using intraluminal suture technique first described by Longa with a little modification.A total of 60 Wistar rats were divided into 4 groups:the sham-operation group, control group,octanol-treatment group and DMSO vehicle control group. Control group were further divided into seven subgroups at different time points of reperfusion after middle cerebral artery occlusion. To observe the change in permeability of BBB,Evans blue(EB)in the brain tissue was surveyed by the means of EB fluorescent quantitation.Octanol-treatment group and DMSO vehicle control group were done at the point of the peak of permeability of BBB.Octanol, the specific blocker for gap junctions(GJ)was used in an intervention study. To compare the amount of EB with the same point of groups, the influence of octanol on BBB permeability was investigated. Results At 3 h of reperfusion after cerebral ischemia for 2 h,the permeability of BBB began to increase, reached the peak at 24 h of reperfusion and was still elevated at 72 h.The Cx43 expression formed into bigger plague and remained linear disposition in the penumbra after reperfusion subsequent to cerebral ischemia.Octanol group was done at 24 h of reperfusion after cerebral ischemia. The amount of EB of octanol group((4.924?0.296)?g/g)was significantly lower than that of corresponding operation control group(5.543?0.506)?g/g.Conclusions (1)Cx43 expression is concentrated around vessels in brain. The Cx43 forms into bigger plague and the function maybe strengthens after reperfusion.Gap junction might aggravate the disruption of BBB.(2) Octanol, the specific blocker of gap junctions, could effectively prevent the permeability of BBB from increasing and has a protective effect on BBB.

PurposeAdrenal schwannoma is rare and quite difficult to diagnose before operation. This paper aims to analyze the CT findings of adrenal schwannoma to improve the recognition and diagnosis of this disease.Materials and Methods The CT findings of 6 patients with adrenal schwannomas proved by operation and pathology were retrospectively analyzed. Results Four schwannomas were located in the right adrenal and 2 in the left adrenal. The tumors, with diameters ranging from 3.0 cm to 6.4 cm, mean (4.5±1.3) cm, had well-defined margin. The densities of all the masses were close to or slightly lower than those of kidneys on plain scan images, with mean CT attenuation value of (32±7) HU. Three masses showed homogeneous density and the other 3 were mildly heterogeneous with patches of lower density. All the 6 schwannomas were enhanced mildly or moderately after intravenous administration of contrast material and showed gradual enhancement mode. They showed mild homogeneous or flocculent enhancement on arterial phase and had gradual progress on venous and delayed phases. The mean increase of CT attenuation value was (13±2) HU on arterial phase, (18±4) HU on venous phase and (23±4) HU on delayed phase. Only 1 mass showed some patches of cystic degeneration without enhancement inside. Conclusion Adrenal schwannoma should be included in the differential diagnosis of solid nonfunctioning adrenal tumors. Features such as relatively homogeneous mass, mild to moderate enhancement and gradual enhancement mode after contrast enhancement are possibly the unique CT manifestations of adrenal schwannoma.
