1.Clinical experience and bibliography retrospection of multiple primary lung cancer

Objective:To analyze the diagnosis and surgical treatment of multiple primary lung cancer.Methods:The clinical data of 11 cases of multiple primary lung cancer were analyzed retrospectively,9 males and 2 females.5 cases developed bilateral synchronous lung cancer and 6 cases developed metachronous multiple primary lung cancer.Different operative procedures were adapted to the various growing sites of tumor,8 cases with normative pulmonary lobectomy, 3 cases with lump resection for the poor pulmonary function.Results:All cases were successful,10 cases were in follow-up,1 case was out of follow-up.8 patients survived one year (72.7%),4 patients survived 3 years (36.4%),1 patient survived 5 years (9.1%),2 patients were still in follow-up.Conclusion:Operative treatment is the key therapy for multiple primary lung cancer.Excessive treatment and long course of chemotherapy should be prevented after operation.
2.Protein kinase Cβ-mediated matrix metalloproteinase 9 activation by acute femoral artery injury

Objective To investigate the activation of matrix metalloproteinase (MMP)9 by acute arterial injury and the involved signaling mechanism in murine femoral artery. Methods In the C57BL/6 mice femoral artery denudation injury were performed. Total protein and membrane protein extracts were prepared from targeted arteries. The MMP 9 activity was measured by zymography assay, the expressions of MMP 9 antigen and protein kinase C (PKC) isoforms were measured by Western blot. Seventy-two hours after mice fed with PKCβ inhibitor (ruboxistaurin), the denudation injury triggered MMP 9 activation was reassessed. Results Within 4-24 hours after denudation injury, MMP 9 activity in femoral arteries was significantly increased, with a peak induction of (99.3±9.5) times the sham control (F=51.49,P<0.01) at 8 h. MMP 9 antigen increased in parallel with MMP 9 activity. Within 15-120 minutes after denudation injury, there was a significant induction of PKCβⅡ in membrane fraction of femoral arteries, with a maximum induction of (7.50±0.60) times the sham control (F=207.06,P<0.01)at 30 min. Injury-induced MMP 9 activation was significantly inhibited by ruboxistaurin. Conclusions MMP 9 activation is, at least in part, mediated by PKCβ in acute arterial denudation injury, it highlights the new target for therapeutic intervention to suppress the over-activation of MMP 9, which plays a critical role in restenosis.
3.Cardiac stem cells in cardiac tissue engineering:present and future

BACKGROUND:For a long time, the myocardium of adult mammalians is the terminal y differentiated tissue with no regeneration capacity. If damaged, myocardial cells wil be replaced by fibrous connective tissue. OBJECTIVE:To rediscover the myocardial cells and to do a review for cardiac stem cells, in order to define the existence of myocardial cells. METHODS:A computer-based online research of CNKI and PubMed databases was performed to col ect articles published between 2003 and 2014 with the key words of“cardiac stem cells, stem cells, cardiac regeneration”in Chinese and English, respectively. There were 82 articles after the initial survey, and final y 40 articles were included in result analysis. RESULTS AND CONCLUSION:Cardiac stem cells exist in the heart, and some surface markers of cardiac stem cells have been discovered. Cardiac stem cells for some diseases with myocardial cellinjury have opened up a new way. Because of the smal number of myocardial cells, how to purification, culture, identification and proliferation wil be further studied to meet the need for regenerative medicine and tissue engineering. Cardiac stem cellresearch wil open a new approach for cardiac tissue engineering.
4.Detection of Lead Ions in Water Based on Surface Energy Transfer between Gold Nanoparticles and Fluorescenct Dyes

We established a sensitive method for the detection of lead ions in aqueous solution based on the surface energy transfer between gold nanoparticles and fluorescent dyes. The fluorescein-modified thrombin aptamer (5′-FAM-GGT TGG TGT GGT TGG-3′) can be selectively transformed to G-quadruplex induced by lead ions. The conformation changes of the aptamer can alter the distance between the energy donor of fluorescent dyes and the energy receptor of gold nanoparticles, and further enhance the fluorescence intensity. The fluorescence recovery efficiency ( F/F0 ) is proportional to the concentration of lead ions in the range of 12. 5-100 nmol/L following the linear regression equation of y=0. 910﹢0. 007c(R2=0. 997)and the limit of determination is about 10 nmol/L. The proposed method has been applied to the determination of lead ions in tap water with satisfied results.

6.Application of Microsoft Excel Worksheet to Management of Drugs in Hospital

OBJECTIVE: To strengthen drug management in hospital.METHODS: Hospital Information System(HIS)-based drug management was supplemented and consummated with the help of Microsoft Excel worksheet's functions such as ranking,computing and advanced screening etc.RESULTS: Using the Microsoft Excel worksheet to help monitor inventory control,warehousing,and shortage of drug supply,unsalable drugs and expiration date in HIS-based drug management has facilitated the effective management on both the quality and the quantity of drugs in our hospital.CONCLUSIONS: Microsoft Excel worksheet can be utilized to consummate HIS-based electronic management of drugs in hospital.
7.The Biotransformation of Atrazine and Influence Factors by Genetically Engineered Microorganism

The co-metabolic carbon sources,dynamics and influence factors of atrazine biotransformation by genetically engineered microor- ganism(GEM)were investigated in the paper.The results showed that glucose was superior to acetate as co-metabolic carbon sources.The concentration of carbon sources affected little on atrazine transformation but much on GEM growth.The specific transformation rate(STR)of atrazine was influenced not by cell density of GEM but by atrazine concentration.Monod model was used to simulate atrazine transformation dynamics and the parameters were v_(max)=0.168/h and Ks=30.49 mg/L.When temperature reduced,STR decreased markedly.The trans- formation was efficient in weak alkaline condition and was inhibited in acidic condition.To some extent,salinity did not affect transformation activity of GEM.Co~(2+),Fe~(2+),Fe~(3+),and Zn~(2+) improved atrazine transformation,but Mn~(2+),Ni~(2+) and Cu~(2+) inhibited atrazine transforma- tion.The results also showed that there existed a direct correlation between adsorption and transformation of atrazine by GEM.
8.Analysis Clinical Drug Use of Amikacin in a Hosiptal

OBJECTIVE:To reference clinical utilization of amikacin of a hospital.METHODS:By retrospective survey,the clinical information of 30 inpatients who were treated with amikacin and received plasma concentration monitoring from Apr.2006 to Feb.2010 were analyzed statistically in respect of primary disease,results of pathogen culture and drug sensitivity test,dosages and usage,plasma concentration,clinical efficacy,adverse drug reactions,etc.RESULTS:Clinical response rate was 55.2% and dosage of amikacin was small as well as measured value and reference value of plasma concentration.No adverse drug reaction was observed.CONCLUSION:The proportion of amikacin should be increased and plasma concentration monitoring of amikacin should be strengthened.Individual regimen is realized to improve effectiveness and safety of drug use.
9.Effect of IL-21 on Treg cells in peripheral blood mononuclear cells in pathogenesis of Graves disease

Objective:To explore the role of IL-21 in regulating the expression of peripheral blood Treg cells in the pathogenesis of Graves disease ( GD ) . Methods: Electrochemical luminescence detection was used to determine levels of thyroid function indexes and autoantibodies. Peripheral blood mononuclear cells (PBMCs) were extracted,then,cultured in the presence or absence of IL-21 in vitro. The level of IL-10 protein was measured by enzyme-linked immunosorbent assay ( ELISA) ,and expressions of Foxp3 and IL-10 mRNA were examined by Real-time PCR. Results: Compared to eGD and control group, there were significant differences in the levels of thyroid function indexes in GD group (P<0. 05),whereas there was no difference between control group and eGD group (P>0. 05). Before IL-21 stimulation,compared to eGD and control groups,expressions of Foxp3 and IL-10 mRNA and level of IL-10 protein were significantly higher in GD group ( P<0. 05 );but there were no significant differences between eGD group and control group (P>0. 05). After IL-21 stimulation,expressions of Foxp3,IL-10 mRNA and IL-10 protein levels were decreased markedly in all three groups,among which GD group showed the greatest change(P<0. 05). Conclusion:IL-21 may inhibit the differentiation of Treg cells and the production of IL-10,therefore decreasing the number of Treg cells and the ability of suppressing effector T cells ,thus should be involved in the pathogenesis of GD.
