3. Preparation of X-ray developable alginate-barium sulfate microspheres as material for embolic agent
Academic Journal of Second Military Medical University 2010;29(7):833-836
Objective: To prepare an X-ray developable alginate-barium sulfate microspheres as material for embolic agent. Methods: The emulsification-internal gelation technique was used to prepare alginate microspheres and the orthogonal design was adopted to optimize the technique. The diameter, shape, dispersibility, stability, suspension property, coating ratio and developability under X-ray of the microspheres were examined. Results: The average diameter of the microspheres was (53. 08± 32. 72) μm; they were well distributed and had a round shape. The ratio of the destroyed microspheres was (2. 0±1, 1) % after heated at 100°C for 2 h,(86. 0± 19. 2)% after frozen at - 4°C for 24 h, (39. 0± 14. 7)% after vibrated at 37°C for 24 h, and (10. 3±3. 2)% after irradiated with 60Co(10kGy) for 0. 5 h. The ratio of sedimentation volume in 0. 25 % suspension of sodium alginate microspheres was (0. 92±0. 018). The coating rate was (69. 2±13.2) % when the feeding amount of barium sulfate was 2. 0 g, and the result of X-ray development was good. Conclusion: Alginate-chitosan can be used for the preparation of barium sulfate microspheres; and the emulsification-internal gelation technique can prepare the microspheres with good dispersibility and shape.

Chinese Journal of Medical Imaging Technology 2001;17(5):470-471
Objective To explore the pathogeny, pathology, ultrasonic and clinical expression of upper extremity deep venous flow obstruction and superior vena cava syndrome. Methods Retrospective analysis and summarize the disease changes of 10 cases in recent 5 years. Results 7 patients of 10 cases were superior vena cava syndrome, in which 4 cases caused by upper right lung malignant tumor, 3 cases caused by upper and media mediastinal malignant tumor. 2 patients of 10 cases were found thrombosis in right subclavian vein caused by duct of artificial pacemaker. The left 1 patient was no reason thrombosis in right axillary vein. Conclusion Ultrasonography combine with color and spectral Dopper image can evaluate the clinical development and therapeutic efficiency, which is a useful and effective non-invasive examination.

BACKGROUND:Current research has shown that many cellgrowth factors can promote the proliferation and differentiation of epiphysis stem cells, and this regulation is closely related to Sox-9.
OBJECTIVE:To investigate the effects of smal interfering RNA-induced specific silence of Sox-9 gene on the proliferation and apoptosis of epiphysis stem cells.
METHODS:Epiphysis stem cells were isolated from the ring of La Croix with enzyme digestion and purified with magnetic activated cellsorting, identified by immunocytochemistry assay. The cells in good conditions were selected to be transfected by Sox-9 silenced by smal interfering RNA with fluorescent marker, and then were observed under the fluorescent microscope to check the transfection efficiency. Next step, epiphysis stem cells were divided into 2 groups:group one was cultured normal y as control group;group two was transfected by Sox-9 silenced by smal interfering RNA through LipofectamineTM 2000 as experimental group.
RESULTS AND CONCLUSION:The primary epiphysis stem cells were separated and purified, which were of stable growth and tightly attachment. The results of immunohistochemistry and immunofluorescence showed the epiphysis stem cells expressed cel-specific markers, fibroblast growth factor receptor 3. After transfection, reverse transcription-PCR results showed that Sox-9 gene expression in epiphysis stem cellwas inhibited specifical y;3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide results showed that cellproliferation in experimental group decreased significantly compared with the control group (P<0.05), and the cellapoptosis detected by flow cytometry showed that the experimental group increased as compared with the control group (P<0.05). Sox-9 gene plays an important role in regulating the proliferation and apoptosis of rat epiphysis stem cells.

Objective To investigate laparoscopic-guided selective portal vein ligation in the two-stage hepatectomy for patients with primary hepatocellular carcinoma (HCC).Methods Twenty-three patients with HCC who were not suitable for one-stage hepatectomy were admitted to the Sichuan Provincial People's Hospital from March 2009 to February 2012.Their clinical data were retrospectively analyzed.Laparoscopic-guided selective portal vein ligation was firstly performed, dynamic changes of hepatic volume and predicted volume of liver to be resected were detected by computed tomography.Two-stage open hepatectomy was performed after assessment of resectability of HCC.All data were analyzed using the analysis of variance or q test. Results Laparoscopic-guided selective portal vein ligation was successfully performed on 22 patients (2 patients received concomitant cholecystectomy because the right branch of portal vein was difficult to expose),1 patient was converted to open surgery because of hemorrhage during portal vein separation.Three patients with multiple lesions received transcatheter arterial chemoembolization at 1 week after selective portal vein ligation.Dull pain in the hepatic region, low fever, nausea and vomiting were observed in the 23 patients, while no severe complications including peritoneal hemorrhage, bile leakage, hepatapostema was observed. The levels of aspartate aminotransferase, alanine aminotransferase and total bilirubin were back to normal at 1 week after the surgery. The right liver volume at postoperative week 3 was (590 ± 154)cm3,which was significantly smaller than (698 ± 135)cm3 before surgery.Compared with right liver volume at postoperative week 1,2,3,the right liver volume before operation was significantly smaller (F=15.62,P <0.05).The left hepatic volume at postoperative week 3 was (408 ± 149)cm3,which was significantly bigger than (331 ± 68)cm3 before operation. The left liver volume before operation was significantly different from those at postoperative week 1,2,3 (F =17.48,P < 0.05).The predicted ratio of liver to be resected was 60% ± 18% at postoperative week 3,which was significantly smaller than 67% ± 15% before operation (F =12.35,P < 0.05).Two patients with insufficient hyperplasia of offside liver,2 patients with intrahepatic metastasis at postoperative week 3,2 patients were lost to follow up and 3 patients gave up hepatectomy,14 patients received hepatectomy at 2-4 weeks after laparoscopic-guided selective portal vein ligation. The resection rate was 60.9% (14/23).There were 2 patients received extended right hepatectomy,8 received right hepatectomy,4 received non-anatomical hepatectomy. All the 14 patients recovered well, and no hepatic failure, severe peritoneal effusion and infection was observed. Conclusion Laparoscopic-guided selective portal vein ligation is easy to perform, and it extends the indication of hepatectomy, increases the safety of two-stage hepatectomy.
