Objective To investigate the effects of CTX-d from venoms of cobra (Naja naja atra) on inducing NB4 apoptosis and its mechanism. Methods MTT was used to detect the antitumor effect of CTX-d in vitro; Electron microscope and flow cytometry were used to observe the apoptotic inducing effect of CTX-d in NB4 cells; Mitochondrial transmembrane potential change (??m) was analyzed by flow cytometry; The levels of caspase-9, caspase-3, and cytochrome C in the cytosol fraction were analyzed by Western blotting. Results The IC50 values of CTX-d affected on NB4 cell for 6 and 12 h were 1.8 and 1.35 ?g/mL, respectively. CTX-d could induce morphological changes, such as condensed chromatin and swelling mitochondria in NB4 cells. Analyzed by flow cytometry, CTX-d induced apoptosis in NB4 cells evidenced by increasing sub G1 cell population in a dose- and time-dependent manner. The mitochondrial membrane potential of NB4 cells had already decreased when incubated with CTX-d (1.0 ?g/mL) for 0.5 h, and cytochrome C in the cytosol was detected simultaneously, which indicated the release of cytochrome C from mitochondria to cytosol. The caspase-9 was activated initially at 1 h after 1.0 ?g/mL CTX-d treatment, whereas the cleavage of caspase-3 was detected at 0.5 h. This suggested that some other mechanism may be involved in caspase-3 activation. Conclusion The results suggest that the loss of mitochondrial membrane potential and the release of cytochrome C from the mitochondria into the cytosol are the early events of CTX-d on NB4 apoptosis. Once release into the cytosol, cytochrome C precedes the activation of caspase-9 and -3 to leading to the apoptosis and there are maybe some other mechanism involved in caspase-3 activation.

Objective To evaluate the biomechanical characteristics of the long U-shaped hollow pedicle screw system for percutaneous fixation of thoracolumbar fractures.Methods Models of vertebral compression fracture were created by flexion in 8 fresh cadaveric samples of thoracolumbar spine.The models were divided into 2 equal groups according to the fixation system applied.In group A conventional pedicle screws were used while in group B long U-shaped hollow pedicle screws were used.The yield compressive strength,axial compression stiffness,bending stiffness,torque and torsional stiffness of all the fixated spines in the 2 groups were tested by the bio-mechanical electrical measurement and compared as well.Results The conventional pedicle screws and long U-shaped hollow pedicle screws showed no significant differences regarding yield compressive strength (3450.02 ± 110.03 MPa vs.3430.04 ± 102.12 MPa),axial compressionstiffness (407.52 ± 33.02 N/mm vs.395.04 ± 26.46 N/mm),bending stiffness (305.14 ± 26.53 N · cm/Degvs.295.12±23.80 N · cm/Deg),torque (6.45±0.25 N · m vs.6.25±0.21 N · m) or torsional stiffness (0.54±0.04N·cm/Degvs.0.52±0.03N·cm/Deg) (P＞ 0.05).Conclusion The long U-shaped hollow pedicle screw system can satisfy the biomechanical requirements for fixation of thoracolumbar fractures and can function as well as the conventional pedicle screw system.

Objective: To estabalish the quality control standard for Yangyanan Lotion Fructus Cnidii; Herba Solidaginis; Cortex Phellodendri; Rhizoma Brainea insuignis. Methods: Fructus Cnidii , Herba Solidaginis, Cortex Phellodendri, Rhizoma Brainea insuignis were respectively identified by TLC. The cnidiadin in Yangyanan Lotion was purified by weak aqua ammonia and determined by HPLC.Results: The average recovery of cnidiadin was 97.96% ( n =6). with RSD was 2.2%.Conclusion: The identification was highly specific, and the method was simple, specific and accurate, and can effectively control quality of Yang Yunan Lotion.

Objective :To establish a RP-HPLC method for determination of isofraxidin in Pientzehuang oral tablet. Methods: RP-HPLC analysis was carried out on Nava-Pak C18 column and with acetonitrile: 0.1%phosphoric acid (15∶85) as a mobile phase. Results: The linear range was 12.9～90.3 ?g/mL(r=0.9999,n=7).The average recovery rate was 102.65 %and RSD=0.71 %.The intra-day and inter-day RSD was less than 3 %.Conclusion :This method is simple, rapid and accurate and suitable for the quality control of the preparations of Chinese herbal medicine containing isofraxidin.

Medical safety management and non-medical safety management are two major systems of hospital safety management.With the deepening of the medical and health system reform and the change of the mode of medical treatment,the connotation and extension of non medical safety management have changed,and the non-medical safety management field is faced with more risk factors and security risks.Combined with the.status quo of non medical safety management in hospital,the problems existing in non-medical safety management are analyzed and some suggestions for improvement are put forward.
