1.Protection of indole-3-carbinol against ethanol-induced hepatotoxicity in precision liver-cut slices of rats

AIM To study the hepato-protection of indole-3-carbinol (I3C), one of glucosinolate compounds in cruciferous vegetables, on ethanol-induced hepatotoxicity. METHODS Ethanol 50 mmol?L -1 was used to induce hepatotoxicity in precision liver-cut slices of rats. The release of glutamic pyruvic transaminase(GTP), glutamic oxaloacetic transaminase (GOT), lactate dehydrogenase (LDH) and glutathione S-transferase (GST) into culture medium, and aniline hydroxylase (ANH) and alcohol dehydrogenase (ADH) in cytosol of hepatocytes in the precision liver-cut were measured with different doses of I3C. The histopathological observation of liver slices was also done to evaluate the effect of I3C. RESULTS The release of GTP, GOT, GST and LDH in the medium increased significantly when the slices were treated with 50 mmol?L -1 ethanol for 4 h. I3C at concentrations of 100~400 ?mol?L -1 effectively reduced the release of GTP, GOT, GST and LDH in the medium of ethanol-treated slices. Meanwhile, the activities of cytosol ANH and ADH decreased to the normal levels. The histopathological changes of liver slices induced by ethanol were lessened by I3C treatment. CONCLUSION I3C can markedly protect rat liver slices from ethanol-induced hepatotoxicity, which partly results from the change of ethanol metabolism in liver.
3.Investigation on the effect of defluoride project in Yantai economic & technological development area

Objective In order to explore the effectiveness of defluoride project on the control of endemic fluorosis in Yantai economic & technological development area. Methods Nine hundred forty-five children aged 7-12 were selected in 4 endemic fluorosis disease areas (Guxian,Bajiao,Dajijia Streets and Changjiang community) and the control area of Yantai economic & technological development area. Prevalence of children's dental fluorosis was determined by Dean method. Water fluoride content was measured in the 4 endemic fluomsis disease area by means of fluorides ion selective electrode. Results Childr dental fluorosis prevalence rate in Guxian,Bajiao,Dajijia Streets and Changjiang community was respectively 30.1% (55/183),41.2% (73/177),33.7% (66/196),31.8% (64/201),higher than those in the control area [13.3% (25/188)]; the median of water fluoride in the 4 disease areas was 1.84,3.46,2.39,2.10 mg/L,respectively,higher than the National Health on the control of endemic fluorosis is not obvious,Standard (1.00 mg/L). Conclusions Effect of the defluoride project in Yantai economic & technological development area needs to be improved.
4.Construction of eukaryotic expression vector for rat Akt1 gene and its expression in 293 cells

BACKGROUND: Akt gene plays an important role in cell survival, proliferation, metabolism, and apoptosis. Although mechanism of Akt gene still remains unclear, it, a major element of survival signal, has aroused much attention in the domain of molecular biological research. OBJECTIVE: To construct eukaryotic expression vector pDC316-Akt1 for Aktl gene and to observe the expression in 293 cells. DESIGN, TIME AND SETTING: An observational study was performed at Molecular Biological Laboratory of Academy of Military Medical Sciences of Chinese PLA between September and December 2006. MATERIALS: pCD316 plasmid was provided by Benyuan Zhengyang Company; DH5α and 293 cells were reserved in our laboratory. METHODS: Aktl DNA segment was cloned from total RNA of hepatic tissue using RT-PCR and inserted into eukaryotic expressing vector pDC316 between EcoRI and Hind Ⅲ sites after sequencing. And then, 293 cells were transfected with the recombinant by liposome. MAIN OUTCOME MEASURES: Aktl gene content in transfected 293 cells using Westem blotting. RESULTS: Sequencing test indicated that Aktl amino acid sequence was in accordance with wild Aktl sequence. Akt-pDC316 was transcripted in 293 cells, and Akt1 protein with relative molecular weight of 55 000 was highly expressed in 293 cells when assayed using Western blotting. CONCLUSION: Eukaryotic expression vector for Aktl gene highly expresses in 293 cells.
5.Long-term effect of pseudomonas aeruginosa preparation on advanced colorectal cancer during surgery

Objective: This study aimed to observe the long-term effect of Pseudomonas aeruginosa preparation used in peritoneal injection of advanced colorectal cancer patients during surgery. Methods: A total of 83 colorectal cancer patients who received surgery between September 2006 and March 2008 were enrolled in this study. The patients were divided into two groups. Palliative resection and a 10 ml P. aeruginosa peritoneal injection were performed in 30 of 83 patients in the treatment group. Simple palliative resection was conducted in the other 53 patients, which comprised the control group. Both groups were then treated by regular chemotherapy and radiotherapy. Results:The follow-up visit was completed in 79 of 83 patients, with a high follow-up rate of 95.2%. No significant difference was found in the five-year overall survival time between the two groups (P=0.403). However, the five-year median survival time in the control group was only 13.9 ± 2.14 months, whereas that in the treatment group was 17.2 ± 2.12 months. Conclusion: Within a short period, peritoneal injection of P. aeruginosa during surgery could confer certain survival advantages for advanced colorectal cancer patients. However, the long-term effect of this therapy remains unknown.
6.Effect of different acupoint prescriptions on prevention and treatment of stress gastric ulcer.

OBJECTIVETo investigate the effects of different acupoint combination on prevention and treatment of stress gastric ulcer and to search for a better acupoint combination.
METHODSThe theoretical basis and experimental research on treatment of stomach diseases with the combination of he-(sea) points and front-mu points as well as back-shu points and front-mu points are analyzed in this article.
RESULTSThe combination of he-sea points and front-mu points is suitable to be applied on acute diseases, febrile diseases and diseases of fu organs. While the combination of back-shu points and front-mu points is more applicable on chronic gastric diseases.
CONCLUSIONIt is proved that the combination of he-sea point (Zusanli ST 36) and front-mu point (Zhongwan CV 12) has reliable effect on prevention and treatment of stress gastric ulcer.

7.Reconstruction of facial nerve defect in parotidectomy of parotid carcinoma using sternocleidomastoid muscle-great auricular nerve flap

Objective To develop the clinical application and the method for functional repair in fa- cial nerve defect in parotideetomy of parotid carainoma.Methods Defect of facial nerve in parotidectomy was repaired by transplantation of sternocleidomastoid muscle-great auricular nerve flap with anastomosis of great auricular nerve-facial nerve under microscope.Results Eight eases of facial nerve defect in parotid carcinoma were repaired by this method.The facial nerve function almost recovered and access to normal dur- ing 3 to 6 month after operation in this series.6 of 8 patients achieved a gradeⅡ,2 of 8 patients achieved a gradeⅢ.Conclusion Reconstruction of facial nerve defect using sternocleidomastoid muscle-great auricu- lar nerve flap can provide better blood supply for the plerosis and regeneration of nerve.The nerve flap also ac- celerate the functional recovery after nerve grafting.
