3.Effect of basic fibroblast growth factor gene transfection on bio-behavior of rat extraocular muscle satellite cells

BACKGROUND: Experiments have demonstrated that exogenous basic fibroblast growth factor (bFGF) promotes the growth and survival of skeletal muscle satellite cells, and endogenous bFGF also has obvious effect on muscular repair.But whether bFGF gene transfection has the same effect on extraocular muscle satellite cells is unclear.OBJECTIVE: The goal of this study was to investigate the effect of bFGF gene transfection on the bio-behavior of rat extraocular muscle satellite cells.DESIGN: Single sample observation.SETTING: Department of Ophthalmology, Qingdao University Medical College.MATERIALS: Primary rat extraocular muscle satellite cells were cultured (purity > 90%). Human PEGFP-N3-bFGF eukaryotic expression vector was successfully constructed (reported in other papers).METHODS: This study was carried out in the Central Laboratory of Qingdao University Medical College between September 2005 and December 2006. Experimental intervention and grouping: During the experiment, 3 groups were divided: Experimental group, in which, recombinant PEGFP-N3-bFGF was used for transfection; Control group A, in which, empty-load PEGFP-N3 was used for transfection; Control group B: in which, only F 10 medium (0.1 volume fraction of fetal bovine serum)was used for transfection. Recombinant PEGFP-N3-bFGF plasmid was used to transfect rat extraocular muscle satellite cells cultured in vitro by liposome-mediated transgenic technology. Experimental evaluation: bFGF expression was observed by immubohistochemical method; Transfection efficiency was detected with fluorescence microscope; The protein secretion of bFGF of satellite cells in each group was detected by SBC-ELISA method on the 1st, 3rd, 5th, 9th, 11th, 13th, 15th, 21st and 28th days of culture; The proliferation of rat extraocular muscle satellite cells in each group was detected by methyl thiazolyl tetrazolium(MTT) method; Creatine kinase (CK) activity of cells in each group was determined according to the A value of some standard whose concentration was known.MAIN OUTCOME MEASURES: ① Transfection efficiency of bFGF gene to rat extraocular muscle satellite cells cultured in vitro and bFGF gene expression after transfection; ②bFGF protein secretion after transfection, and effect of bFGF gene transfection on the growth and proliferation of extraocular muscle satellite cells.RESULTS: ① Transfection efficiency of bFGF gene to rat extraocular muscle satellite cells cultured in vitrowas (42.8±1.2)%. ② Immunohistochemical detection showed positive reaction. ②Transfected cells could secrete bFGF protein,which was the highest on the 5th day (662.935 ng/L). ④ The proliferation activity of transfected cells was obviously enhanced, and its A value was significantly higher than that of control group at the same time point (P < 0.05). ⑤CK value was higher than that of the control group from the 5th day after transfection to the end (P < 0.01).CONCLUSION: bFGF gene transfering into rat extraocular muscle satellite cells can make extraocular muscle satellite cells to secrete bFGF, promote cell proliferation, survival and differentiation.
4.TWO-DIMENSIONAL MEASUREMENT OF THE CAPUT HUMERI WITH LASER PROJECTION

One hundred Chinese adult humeral heads (50 of either side) were studied with laser. The radius, height, angle as well as the surface area of each were anteriorly, laterally and obliquely measured in two-dimensions.The main results are as follows:1) The mean angle of the articular area was 144.48?11.9?.2) The average radius and height of the humeral head were 2.15?0.16 cm and 1.56?0.24 cm respectively. The articular surface height occupied 36.05 per cent of the diameter of the approximate spherical body.3) The mean articular surface area of the humeral head was 21.02?4.03cm~2. According to the radius of the humeral head and the articular surface angle, the curvature of the articular surface can be estimated and drawn out. That will offer the theoretical data of Chinese adult humeral head for the design and manufacture of artificial joint and artificial humeral head.
5.HLA-A, B, and DRB1 polymorphism at high-resolution in Ji-nan Han population

Objective To analyze the polymorphic character of HLA-A,B,DRB1 alleles at highresolution level in Ji-nan Han population.Methods A total of 483 unrelated and healthy Han ethnic individual from Ji-nan region were genotyped by PCR-SBT for HLA-A,B and DRB1 loci.Then allelic and haplotypic frequencies were calculated by maximum likelihood estimation method using Arlequin software.Results There were 27,56 and 41 alleles identified at HLA-A,B and DRB1 loci respectively,and the most common alleles were A * 11∶01(0.1615),B * 13∶02(0.1163) and DRB1 * 07∶01 (0.1763).The three loci haplotype was dominated by A* 30∶01-B * 13∶02-DRB1 * 07∶01 with a frequency of 0.0867.Conclusion The HLA-A,B and DRB1 loci are highly polymorphic in Ji-nan Han population.

As a new type of environmental pollutants, microplastics are widely distributed in the global ecosystem, and ingestion of microplastics may produce a number of toxic effects. Based on currently available publications, this paper describes the main pathways of exposure to microplastics, and summarizes the toxic mechanisms of microplastics in mammals, including oxidative stress, inflammatory response, immune damage, imbalance of gut microbiota, energy metabolism disorder and DNA damage, so as to provide insights into elucidation of the toxic mechanism mechanisms and health risk assessment of microplastics.
7.The Evaluation of ROC Curve in Qualitative and Quantitative Analysis of Pancreatic Diseases on Fat Saturation FSPGR Sequence

Objective To evaluate the accuracy of fatsuppressed fast spoiled gradient echo(FSPGR) T_1WI in detection of pancreaticdisease using the receiver operating characteristic curve(ROC).Methods The pancreas were evaluated in 92 subject investigated(57normal and 35 with pancreatic disease) with fat saturation FSPGR T_1WI.The signal intensity(SI) ratio of pancreas relative to liver and spleen was quantitatively measured using regions of interest(ROIs),then qualitatively assessed by one independent observers.Results The mean values of pancreas-liver ratios and pancreas-spleen ratios between a normal and an abnormal pancreas were significantlydifferent.Thearea under ROC curve of the pancreas-liver SI ratio and the pancreas-spleen SI ratio was 0.92?0.03 and 0.91?0.04,respectively.There was no statistically significant difference between quantitative and qualitative analysis for the diagnosis of pancreatic disease when using liver as comparable standard.The sensitivity of qualitative pancreas in comparison with liver was 86.7%,while specificity was 91.2%.Conclusion When the SI ratio of pancreas-liver is used to distinguish normal from abnormal pancreas,qualitative analysis by observers is as accuracy as quantitative measurement.
8.MRI Diagnosis of Meniscus Tear of Knee Joint

Objective To evaluate the value of MRI in diagnosis of meniscus tear of knee joint.Methods MRI manifestations of meniscus tear of knee joint were analyzed in 60 cases.Four-grade classification of meniscus injury was suggested according to Stoller's classification.Results Meniscus tear was seen in 65 menisci of 60 patients,including 15 medial tear,40 lateral tear,and 5 cases(n=10) of bilateral tear.Fifteen lateral discoid menisci were found among the 60 cases of meniscus tear.MRI revealed a accuracy of 90.8%.Thirty-nine cases were complicated by ligament injury,bone contusion,effusion of joint,fracture and meniscus cyst.Conclusion MRI can accurately diagnose meniscus tear and its complication.
9.MRI Manifestations of MELAS Syndrome

Objective To investigate MR imaging characteristics of MELAS syndrome.Methods The clinical data and MRI features of 13 cases with MELAS syndrome were retrospectively analyzed.Results Multiple hypointense on T1WI and hyperintense on T2WI lesions were showed predominantly in the gray matter of the temporal, parietal and occipital lobes. The lesions crossed vascular boundaries. No stenosis and occlusion of main artery were displayed by MRA.Basal ganglia calcifications and cerebral atrophy with widening of ventricles were also frequent found. Increased ADC value of the leison on DWI and appearance of Lac peak on1H-MRS were useful in diagnosis.Conclusion There are some characteristics of MRI in patients with MELAS syndrome.Combined clinical informations with neuroimaging and muscle biopsy, the diagnosis of MELAS syndrome can be made correctly.
