1.Management of disorder of temporomandibular joint syndrome using elastic resin occlusal pad: a clinical observation

Objective To investigate the possibility of occlusal pad in treating disorder of temporomandibular joint syndrome.Methods We treated 25 patients with disorder of temporomandibular joint syndrome using occlusal pad made of elastic resin,and evaluated the effects.Results Total effective rate was 96% .Clicking and pain dissappeared in all patients.Conclusion Occlusal pad made of elastic resin could be used for treating disorder of temporomandibular joint syndrome in clinic.
2.Screening and characterization of aptamers of human chronic myeloid leukemia (CML) fusion protein BCR-ABL and its structure analysis

Objective:To screen and characterize aptamers against BCR-ABL fusion protein.Methods:A 90bp single stranded DNA( ssDNA) random library was subjected to 13 rounds of selection against BCR-ABL fusion protein by systematic evolution of ligands by expotential enrichment ( SELEX ) method, the selected aptamers were cloned and sequenced.The primary sequences and structure of aptamers were analyzed by Clustal W and DNA Folding Sever and the percentage of the ssDNA pool bound to BCR-ABL core protein were determinated.Results: after 13 rounds selection, the percentage of ssDNA pool bound to BCR-ABL fusion protein increased from 0.3%to 47.1%,the results showed that affinities of the Aptamers were different,the second structure analysis revealed possible stem-loops for binding to BCR-ABL fusion protein,the affinity of aptamer A2 to BCR-ABL fusion protein was highest with Kd values as low as 72 nmol/L.Conclusion:Aptamers against BCR-ABL fusion protein has been identified by SELEX methods from a 90 bp single stranded DNA library.And provide certain reference for the clinical treatment of chronic myelogenous.
3.The effect of Qibaipingfei capsule on right ventricular hypertrophy index(RVHI)of rats with phlegm and blood stasis of COPD

Objectlve To explore the effect of Qibai Pingfei capsule on the right ventricular hypertrophy index (RVHI)of rats with phlegm and blood stasis of COPD.Methods 60 rats were randomly divided into 6 groups,a control group,a model group,a Ligustrazine group,a Nifedipine group,a high dose Qibai Pingfei capsule group and a low dose Qibai Pingfei capsule group.Composite factors method was adopted to establish phlegm and blood stasis COPD rat model.At the same time of modeling.Qibai Pingfei capsule,Ligustrazine,Nifedipine were also given to these rats.Observed the changes of RVHI in each group.Results RVHI did not show statistic difierence between high dose of the Qibai Pingfei capsule group and the control group(P>0.05).and between the Nifedipine group and the model group(P>0.05).RVHI manifested significant diffbrencc between the Ligustrazine group and the low dose Qibai Pingfei capsule group(P<0.01),and between the Ligustrazine group and the control group(P<0.01).Conclusion High dose Qibai Pingfei capsule can effectively prevent the right ventricular hypertrophy of model rats with the phlegm and blood stasis of COPD.Ligustrazine has certain effects but not as good as high dose Qibai Pingfei capsule.Nifedipine can not prevent the right ventricular hypertrophy of model rats.

6.Comparison of endovenous laser treatment combined with transilluminated powered phlebectomy and traditional surgical treatment for great saphenous varicosity

Objective To compare the efficacy of endovenous laser treatment combined with transilluminated powered phlebectomy and traditional surgical treatment for great saphenous varicosity. Methods From January 2014 to January 2015,77 patients diagnosed as great saphenous varicosity were enrolled. They were randomly divided into two groups, 37 patients received endovenous laser treatment combined with transilluminated powered phlebectomy and the other 40 patients underwent traditional surgical treatment. The related indicators were compared.Results The mean operation time,average bleeding volume,operative incisionquantity and average hospitalization time of EVLT+ TIPP group were (66.1±14.7) min,(24.4±10.5) ml,5.7±1.7,(5.6±1.4) d respectively,of control group were (84.3±18.5) min,(59.0±15.6) ml,8.0±1.8,(10.1±3.1) d respectively,there were significant differences between the two groups(t =-4.749,-11.460,-5.714;P<0.001) .There were no significant complications in the two groups.In the EVLT+ TIPP group,6 patientsoccurred patsubcutaneous induration, 5 patients occurred subcutaneous hematoma and 7 patients had skinparesthesias.In the traditional operation group,2 cases occurred patsubcutaneous induration,3 cases occurredsubcutaneous hematoma and 2 cases had skin paresthesias.The EVLT+ TIPP group was slightly higher than thetraditional operation group in the complications, but there were no significant differences between them ( P>0.05).During a mean follow?up time of (18±7) months(12 months to 24 months),1 patient recurrence in theEVLT+ TIPP group,2 cases recurrence in the traditional operation group.The recurrence rates of the two groupswere similar,the difference was not significant between them(P=1.000) .Conclusion Endovenous lasertreatment combined with transilluminated powered phlebectomy for great saphenous varicosity is effective andsafe.It also has the advanta and better cosmetic results.
7.Expression of specific marker molecules of long-term cultured human dermal papilla cells in vitro

Objective To investigate the expression of specific marker molecules in hair-inducing activity of long-term cultured human dermal papilla cells (HDPCs) in vitro.Methods After dissected and cultured the HDPCs in vitro,the cells of passages 1 to 8 were used for experiments.The growth appearances of HDPCs in different passages were observed under inverted microscope.To detect the expression of specific marker molecules of long-term cultured HDPCs,the alkaline phosphatase (ALP) activity of the HDPCs was examined,and the specific genes ALP and insulin-like growth factor-1 (IGF-1) expression levels of HDPCs were determined by real-time quantitative PCR.Results After long-term cultured in vitro,the ALP and IGF-1 expression levels of HDPCs gradually decreased in different passages,as well as the display of the aggregated and cartouche growth.The ALP and IGF-1 expression levels of HDPCs in passage 1 was the highest,they were almost about 6.8-fold and 3.5-fold higher than the HDPCs in passage 8.The ALP staining of the HDPCs in passage 1 and passage 2 were evident,but the cells' ALP staining gradually became much weaker than the cells in the previous passages after the long-term cultured in vitro.Conclusions The expression levels of specific marker molecules ALP and IGF-1 of the HDPCs decrease gradually after long-term cultured in vitro,and the higher passage HDPCs lost the special aggregated and cartouche growth appearance,and hence lead to the loss of hair-inducing activity of HDPCs.
8.Tissue-engineered bone repairs sheep alveolar bone defects

BACKGROUND:Fol owing physicochemical treatment and high-temperature calcinations, heterogeneous
biological bone becomes a ceramic-like heterologous bone forming a similar structure to the human bone that is a natural network pore structure, which is conducive to seed cel adhesion and proliferation.
OBJECTIVE:To observe the feasibility of constructing tissue-engineered bone through combination of sintered bone and bone marrow mesenchymal stem cel s to repair alveolar defects.
METHODS:Sheep bone marrow mesenchymal stem cel s as seed cel s were combined with the high
temperature sintered bone as scaffold materials to construct tissue-engineered bone. Under general anesthesia, sheep bilateral mandibular first premolars were removed in batches, the alveolar ridge space between the distal root and mesial root of the second premolar to form a bone defect area of 5 mm×5 mm×5 mm. Twelve
experimental sheep were equal y randomized into tissue-engineered bone group and sintered bone group, which were implanted with tissue-engineered bone and sintered bone, respectively, at the left surgical area of the
mandible. The right surgical area was considered as blank control group.
RESULTS AND CONCLUSION:After high-temperature calcinations, the sintered bone was chalk in color, exhibiting a porous structure as the natural cancel ous bone. The porosity was (66.10±1.32)%, and the pore size was between 137.44μm and 538.72μm. After 24 hours of bone marrow mesenchymal stem cel s inoculated to the sintered bone, a large number of cel s are visible adherent to the scaffold;up to day 7, extracel ular matrix was secreted and there was no clear boundary between the cel s and the matrix. X-ray films showed that the tissue-engineered bone and pure sintered bone implants were embedded in the surgical area, and there was a low-density shadow at the edge of the sintered bone. Hematoxylin-eosin staining showed bone trabecular formation at the experimental side, but no obvious bone formation at the control ed side. Tissue-engineered bone prepared by bone marrow mesenchymal stem cel s and sintered bone can better repair sheep alveolar bone defects, which is an ideal seed cel and scaffold material for smal range bone defects.
9.Comparison of anti-inflammatory activity of ethanol extract of coptis chinensis Franch.and Berberine hydrochloride in vitro

Objective:To study the anti-inflammatory effect of the ethanol extract of Coptis chinensis Franch.in vitro.Methods: An inflammatory cell model was established by stimulating the mouse peritoneal macrophages in vitro with lipopolysaccharide(LPS).LPS stimulated of RAW264.7 cells for a long time after administration of intervention.Effect of ethanol extract of Coptis chinensis Franch.on RAW264.7 cell growth activity was analyzed by MTT assay.The production of tumor necrosis factor-α(TNF-α),IL-1β,IL-6,NO,prostaglandin E2(PGE2) was determined by ELISA.The expression of mRNA of TNF-α,induced nitric oxide synthase(iNos) and HO-1 was detected by real-time fluorescent quantitative PCR(RT-PCR).Results: The ethanol extract of Coptis chinensis had no inhibition effect on the scope of RAW 264.7 cells the scope of 5-80 mg/L.Each treatment group concentration of interleukin-6 (IL-6),interleukin-1β (IL-1β),TNF-α,NO,prostaglandin E2 (PGE2) content of LPS stimulation model group were significantly (P<0.01),and content was not related to concentration.Real-time quantitative (RT-PCR) showed,the concentration of each treatment group were significantly lower iNos,HO-1 and TNF-α mRNA expression (P<0.05,P<0.05,P<0.01),and content was not related to concentration.Conclusion: Coptis chinensis Franch.ethanol extract has anti-inflammatory effects in vitro,the mechanism may be related to inhibition of TNF-α,NO and other inflammatory factors and the impact of the activation of arachidonic acid (AA) metabolism.
10.Evaluation of matrix effect of processed materials in serum total glycerol measurements

Objective To evaluate the matrix effect of processed materials in serum total glycerol measurement and to assess the accuracy of routine test systems.Methods With an isotope dilution liquid chromatography tandem mass spectrometry method as the comparative method,matrix effects of 28 processed materials on 8 routine test systems were evaluated ccording to the NCCLS EP 14 protocol.The processed materials and 20 flesh patient specimens were analyzed with both the comparative method and each of the evaluated methods and results obtained with the two methods were plotted.Two-tailed 95% prediction intervals for the mean of the flesh patient specimen were computed and results on the processed aterials were compared with these intervals for evaluation of matrix effect.Results with the two methods on fresh samples were also compared for assessment of the accuracy of the routine test systems.Results Some of the processed samples showed matrix effects on some of the routine test systems.The observed matrix effects were system-specific and aused either positive or negative biases.Calibration biases were also observed on some test systems.Conclusion Matrix effect and calibration bias have been observed in serum total glycerol measurements.Continued efforts are needed for improving the accuracy of serum total glycerol measurements.
