Chinese Journal of Rehabilitation Theory and Practice 2013;19(10):984-987
Objective To summarize the clinical characteristics of foreign accent syndrome (FAS) among Chinese. Methods The journal articles published in Chinese between 1980 and 2011 were retrieved with three Chinese translated versions of FAS as the key words, and 38 articles in line with the inclusion criteria were systematically analyzed. Results and Conclusion Each year, about (3.06±1.65) cases were published on FAS ever since 1996. The age of FAS patients was averagely around 38.10 years, and of the reported cases, there were 26 males and 23 females, there is no significant difference in gender (χ2=0.184, P=0.668). The etiological factors of FAS were identified mainly as injury (59.19%), stroke (32.65%) and psychogenic disorder (8.16%), and 65.31% of the focus were found at the left side of brain.85.72 % of the FAS patients were seen to be speaking a new dialect used by a larger population. 80% of the patients who didn't suffer from a coma changed their accent in 72 h following the injury, while 72.73% of those once in a coma changed their accent as they gained consciousness. The prognosis of FAS was fine, with an improvement rate of 70.83%, and 73.53 % of the patients began to recover less than 3 months later following the injury.

OBJECTIVETo investigate the apoptosis inducing effects of ponicidin (PON) on leukemic K562 cells and its mechanisms of action.
METHODK562 cells in culture medium in vitro were given different concentrations of PON (10-50 micromol x L(-1)) for 24, 48 and 72 h. The inhibitory rate of the cells was measured by MTT assay, cell apoptotic rates were detected by flow cytometry (FCM) using Annexin V staining after K562 cells were treated with different concentrations of PON for 72 hours, and cell morphology was observed by Wright-Giemsa staining. Western blot was used to detect caspase-3 and poly(ADP-ribose) polymerase (PARP) expression, and the protein levels in mitogen-activated protein kinase signaling pathways (MAPKs, p-P38, p-ERK and p-JNK) as well as p-AKT and p-P85 in PI3K/AKT signaling pathways were also detected.
RESULTPON (over 30 micromol x L(-1)) could inhibit the growth of K562 cells in both time- and dose-dependent manner. FCM analysis revealed that apoptotic cells were gradually increased in a dose-dependent manner after treatment for 72 hours, and that marked morphological changes of cell apoptosis such as condensation of chromatin was clearly observed by Wright-Giemsa staining after treatment by 50 micromol x L(-1) PON. Western blot showed cleavage of the caspase-3 zymogen protein (32 kD), with the appearance of its 17 kD subunit, and a cleaved 89 kD fragment of 116 kD PARP was also found. Furthermore, Western blotting also showed that expression of p-AKT and p-P85 in PI3K/AKT signaling pathways was downregulated dramatically whereas the expression of p-P38 as well as p-ERK and p-JNK remained unchanged after the cells were treated by PON for 48 h.
CONCLUSIONThe results demonstrate that PON exhibits in vitro anti-leukemia effect by induction of apoptosis in K562 cells, and that PON induced apoptosis in K562 cells mainly related to activation of caspase-3 as well as inactivation of PI3K/AKT signaling pathway via down regulation of the expression of p-AKT and p-P85 protein levels. These results provide strong laboratory evidence for further anti-leukemia trials of PON.

Hepatic stellate cells(HSCs)are essential drivers of fibrogenesis.Inducing activated-HSC apoptosis is a promising strategy for treating hepatic fibrosis.18beta-glycyrrhetinic acid(18β-GA)is a natural com-pound that exists widely in herbal medicines, such as Glycyrrhiza uralensis Fisch, which is used for treating multiple liver diseases, especially in Asia.In the present study,we demonstrated that 18β-GA decreased hepatic fibrosis by inducing the apoptosis in activated HSCs.18β-GA inhibited the expression of α-smooth muscle actin and collagen type Ⅰ alpha-1.Using a chemoproteomic approach derived from activity-based protein profiling, together with cellular thermal shift assay and surface plasmon reso-nance,we found that 18β-GA covalently targeted peroxiredoxin 1(PRDX1)and peroxiredoxin 2(PRDX2)proteins via binding to active cysteine residues and thereby inhibited their enzymatic activities.18β-GA induced the elevation of reactive oxygen species(ROS),resulting in the apoptosis of activated HSCs.PRDX1 knockdown also led to ROS-mediated apoptosis in activated HSCs.Collectively, our findings revealed the target proteins and molecular mechanisms of 18β-GA in ameliorating hepatic fibrosis, highlighting the future development of 18β-GA as a novel therapeutic drug for hepatic fibrosis.

OBJECTIVETo establish a three- dimensional virtual deformable mandible model used for individual reconstruction design of large mandibular defect.
METHODSA virtual deformable mandible model has been established by a 3D animation software. The model could be used for preoperative reconstruction design of large mandibular defects cases. According to the temporomandibular joint fossa position, maxillary dental arch, the normal relationship of cranio-maxillofacial profile, and the morphology of the residual segments of mandible, the virtual mandible model could be scaled and adjusted and a virtual mandible with individual features was obtained. Three normal skulls have been used to validate the adjustment ability of the virtual deformable mandible model. The preoperative reconstruction design process of 1 typical large mandibular defect case was demonstrated.
RESULTSThe deformation matching ability of the virtual deformable mandible model was very good. The registration between the design model and the original mandible was over 90%. The design effect of the large mandiblar defect case was satisfied.
CONCLUSIONSVirtual deformable mandible model is a new feasible method to aid preoperative reconstruction design of large mandibular defects.
