4.Immune regulation and repair mechanisms of mesenchymal stem cells on incident thrombosis in systemic lupus erythematosus --- review.
Journal of Experimental Hematology 2011;19(4):1092-1096
Systemic lupus erythematosus (SLE) is an autoimmune disease caused by abnormal immune regulation and excessive production of autoantibodies, which characterized by T and B cell dysfunction and excessive production of pathological cytokines and autoantibodies. Vascular endothelia and subendothelial collagen were injured by harmful antibodies, so that the body was in a thrombophilic state, increasing the multi-system and multi-organ damage of body. Mesenchymal stem cells (MSC) are as multipotent cells, capable of multilineage differentiation, self-renewal, homing, inflammatory chemotaxis, immune regulation and reconstruction. To date, MSC are known to affect not only T cells, but also other cells of the immune system. MSC can inhibit or promote B cell proliferation, suppress NK cell activation and modulate the cytokine secretion profile of dendritic cells and macrophages. Thus decreasing the secretion of harmful cytokines and autoantibodies, can ease the thrombosis-prone state of the body, reducing the incidence of thrombosis. In addition, MSC are able to differentiate into various types of tissue cells, such as hematopoietic cells, endothelial cells, liver cells, nerve cells, bone cells, cartilage cells etc, therefore, MSC can repair the damaged tissues and organs. In this article, the advance of studies on immune regulation and repair mechanisms of MSC on incident thrombosis in SLE is reviewed.

AIMTo explore the change of number working memory ability in healthy young adults, a continuous 3-back number working memory task were performed for an hour and 12 Blocks according to different COMT genotypes of young adults.
METHODS18 different genotype subjects were chosen from 112 healthy young adults, P3 event-related potentials was utilized to observe the relationship between this COMT polymorphism and cortical physiology in a continuous working memory task.
RESULTSSubjects bearing the Val/Val homozygote had significantly higher mean P3 amplitudes than Val/Met heterozygote (P < 0.01), however, no significant differences in comparison to Met/Met homozygote.
CONCLUSIONVal/Met Heterozygote subjects are associated with the poorest performance of working memory. There is a relationship between COMT genotype and P3 visual event-related potentials evoked from 3-back task.

The wild resources of Dendrobium officinale in Anhui province were studied by textural research, data collection, interview survey and regional survey, in order to investigate the resources distribution and ecological characters and provide the reference for Anhui Dendrobium industry. In this paper, a part of producing areas of wild D. officinale in Anhui province was selected to analyze the ecological characters. As a result, we find that the wild resources of D. officinale in Anhui distributed only sporadic and the conditions of growth environment were harsh. Our findings may provide some suggestions on wild resources protection and artificial cultivation in suitable environments because the wild resources of D. officinale in Anhui are decreasing rapidly and facing an endangered situation.

OBJECTIVETo assess the practical value of BIOMED-2 primers in the diagnosis of ocular adnexal lymphoma by PCR.
METHODSDNA was extracted from 63 formalin-fixed paraffin-embedded (FFPE) ocular adnexal lymphoma specimens. The DNA quality was evaluated by PCR-based amplification of housekeeping gene beta-actin. IgH(B) and IgK(B) primers of BIOMED-2 standardized clonality analysis system were used to evaluate the immunoglobin gene rearrangements. PCR products were analyzed using capillary electrophoresis and GeneScan software.
RESULTS76.2% (48/63) of FFPE samples produced amplifiable DNA for detection of Ig gene rearrangements.Positive detection rates by BIOMED-2 IgH(B) and IgK(B) primers were 79.2% (38/48) and 68.8% (33/48), respectively, with a combined positive detection rate of 91.7% (44/48).
CONCLUSIONSIgH(B) and IgK(B) primers of BIOMED-2 are suitable for the detection of clonal rearrangements of Ig gene using FFPE specimens of ocular adnexal lymphomas.

Chinese Journal of Cancer 2010;29(6):596-602
BACKGROUND AND OBJECTIVEIn patients with hepatocellular carcinoma (HCC) receiving potentially curative minimally invasive therapy, autologous cytokine-induced killer (CIK) cells were used to reduce recurrence. In this study we observed the changes in serum alpha-fetoprotein (AFP) after the treatment with CIK cells to explore if AFP could serve as a marker for predicting immunotherapeutic clinical outcome.
METHODSA total of 122 patients with HCC and elevated AFP (>25 ng/mL) received a curative treatment of transcatheter arterial chemoembolization (TACE) plus radiofrequency ablation (RFA) at the Sun Yat-sen University Cancer Center. Of these patients, 83 patients without residual tumor or extrahepatic metastasis and with AFP level less than 1.5 times the normal range (AFP<37.5 ng/mL) were randomly assigned to the study group (n=42) and the control group (n=41). In the study group, CIK cells were transfused intravenously or via common hepatic arteries every week for at least 4 times, and the T-lymphocyte subset data before and after CIK cell infusions was examined by flow cytometry. All the two groups of patients were screened by tomography every 2 months to observe tumor recurrence. Serum AFP was collected at baseline and at different time points after treatment in parallel with radiologic response and clinical outcome.
RESULTSTwo patients in the control group were lost to follow-up after treatment. After CIK cell infusions, the downtrend of the AFP level was observed in the study group and not in the control group. There was a significant difference in the level of AFP between different time points after CIK infusions in both groups. The 1-year recurrence rate was 7.14% for the study group and 23.1% for the control group (P=0.044). In subgroup analysis, for patients with a slightly high level of AFP (25 ng/mL CONCLUSIONSCIK cells transfusion may reduce the level of serum AFP and anti-HBV and decrease the 1-year recurrence rate of patients with HCC after curative TACE plus RFA. Serum AFP decrease after CIK cell treatment may serve as a useful marker for predicting immunotherapy clinical outcome in patients with HCC undergone curative minimally invasive therapy.
