1.Observations on the Therapeutic Effect of Acupuncture on Biased Constitution-based Subhealth

Objective To investigate the clinical efficacy of acupuncture in treating subhealth.Methods One hundred and fifty-four patients with subhealth were allocated, using a random number table, to observation and control groups, 77 cases each. The control group received basic treatment by diet, exercise, rest and rest and psychological intervention and the observation group, acupuncture therapy in addition. Subhealth scale subscores and the 36-Item Short-Form Health Survey (SF-36) subscore were recorded and blood biochemical indicators were measured in the two groups before and after treatment. the clinical therapeutic effects were compared between the two groups.Results The total efficacy rate was 96.1% in the observation group and 83.1% in the control group; there was a statistically significant difference between the two groups (P<0.05). There were statistically significant pre-/post-treatment differences in subhealth scale subscores in the two groups (P<0.05). After treatment, sleep, digestion, urination, positive emotion, ability and social relation subscores were higher, and fatigue, discomfort and negative emotion subscores were lower in the observation group than in the control group (both P<0.05). There were statistically significant pre-/post-treatment differences in blood biochemical indicators in the two groups (P<0.05). After treatment, blood uric acid (UA), total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) were significantly lower and high-density lipoprotein cholesterol (HDL-C) was significantly higher in the observation group than in the control group (both P<0.05). The 36-Item Short-Form Health Survey (SF-36) subscore were higher in thetwo groups after treatment than before treatment (P<0.05). After treatment, physiological function, role-physical, role-emotional, body pain and general health subscore were significantly higher in the observation group than in the control group (P<0.05).Conclusions Acupuncture helps to improve subhealth status in population, regulate blood biochemical indicators and enhance the clinical therapeutic effect to raise the quality of life in the patients.
2.Kyoto global consensus report for treatment of Helicobacter pylori and its implications for China.

Kyoto global consensus report on Helicobacter pylori gastritis (Gut, July 2015) is another important international consensus since the European Maastricht Ⅳ consensus was published. Kyoto consensus will improve the etiology-based classification, the diagnostic assessment of gastritis, and the treatment of H. pylori-associated dyspepsia and H. pylori gastritis. However, because of high rate of H. pylori infection and antibiotic resistance as well as limited health resources in China, we need to develop our own strategies of H. pylori infection control with the reference of the Kyoto global consensus.

3.Manipulative reduction and external fixation with cardboard splint for the treatment of calcaneal fractures: a 60-case report.

OBJECTIVETo investigate the therapeutic effects of manipulative reduction for calcaneal fractures.
METHODSFrom January 2009 to June 2012, 53 cases (60 affected feet) of Sanders type I-IV calcaneal fractures were treated by manipulative reduction and external fixation with cardboard splint,including 45 males and 8 females with an average age of (33.5 +/- 1.54) years old ranging from 18 to 65. The course of disease ranged 0.5 h to 7 d. Before treatment the feet were swelling, ache and activity limitation in evidence, some feet were wide flat deformity, the cortical bone was broken in the imaging examination. All selected cases were evaluated using Creighton-Nebraska health foundation assessment scale for fractures of calcaneus.
RESULTSAll cases were followed up at 12 months after treatment, 13 feet got excellent curative effect, 34 good, 11 moderate and 2 poor.
CONCLUSIONManipulative reduction for Sanders type I-IV calcaneal fractures could get excellent curative effect. Manipulative reduction for calcaneal fractures could avoid surgical trauma while assure high curative effects. Manipulative reduction is not only economical and easy therapy, but also can restore maximum function of the calcaneus with few complications and facilitate early rehabilitation of ankle and joint function.

4.Role of HCN channels in the nervous system: membrane excitability and various modulations.

Hyperpolarization-activated and cyclic nucleotide-gated (HCN) channels, distributing in a variety of tissues, especially in excitable cells such as heart cells and many kinds of neurons, have an important role in the modulation of heart rate and neuronal excitability. Different from typical voltage-gated sodium channels and potassium channels, HCN channels were evoked inward currents when the cell was hyperpolarized. More and more recent studies have disclosed that HCN channels play important roles in the nervous system, which were linked with its special electrophysiological features as well as its regulatory effect on the cellular membrane excitability. HCN channels could be modulated by many factors including both extracellular molecules and intracellular signaling cascades, which made its functions complicated in the different condition. Based on its role, HCN channels are presumed to be a promising target for chronic pain and brain disorders. In this paper, we will focus on the advancement of roles of HCN channels in the neural system as well as its complex modulator factors.

It is of capital importance to establish a reliable experimental animal model of intracranial aneurysm which should be extremely similar to human intracranial aneurysm in histological and hemodynamic aspects.For recent years,the elastase-induced aneurysm model by means of endovascular balloon occlusion has been widely used abroad.This paper aims to review the preparation of animal mode,the advantages and disadvantages of such a model as well as the recent progress in its applications.
7.Mouse model of dry eye induced by hyperosmolar saline and its evaluation

Objective To evaluate the murine model of dry eye induced by hyperosmolar saline. Methods Sixty female BALB/c mice at the age of 6 -8 weeks were randomly divided into blank group,control group and experimental group,20 in each group. Mice in control and experimental groups were treated with 308 mOsmol/L and 500 mOsmol/L sodium chloride solution,respectively,5 times a day. Mice in blank group were not treated with sodium chloride solution. Schirmer test,fluorescein staining,corneal scoring,rose bengal staining,tear ferns experiment,corneal epithelial HE staining and thickness measurement,conjunctival epithelial PAS staining and Goblet cell counting were conducted on days 0,7,14,28,and 42,respectively. Corneal surface was observed by scanning electron microscopy on day 42. Results No significant difference was found in the above parameters on day 0 between the two groups. 8.Effect of sodium ferulate on transdifferentiation and extracellular matrix of rat renal tubular epithelial cells induced by transforming growth factor beta 1

BACKGROUND: Renal tubulointerstitial fibrosis is mainly featured as the accumulation of extracellular matrix (ECM) in renal interstitium. The tubular epithelial-myofibroblast transdifferentiation (TEMT) is important to the pathogenesy of renal tubulointerstitial fibrosis. OBJECTIVE: To examine the effects of sodium ferulate (SF) on TEMT, and ECM main components such as collagen Ⅰ, collagen Ⅲ and fibronectin, in rat renal tubular epithelial cellsinduced by transforming growth factor-beta 1 (TGF- β1)- DESIGN: Randomized and controlled experimental study based on cells. SETTING: Department of Kidney in West China Hospital of Sichuan University. MATERIALS: Rat renal tubular epithelial cells (NRK-52E) originated from American Type Culture Collection (ATCC), were offered by the laboratory of Department of Nephrology in Australian Monash Medical Center. Cell strain used in this study was cultured at the 36th passage. SF white crystal with water solubility and more than 98.0% purify, was from Chengdu Hengda Pharmaceutical Co., Ltd. Different concentrations of SF (125,250, 500μreel/L) were designed in this study. Rabbit anti-rat α-smooth muscle actin (α -SMA) was produced by Wuhan Boster Company. Enzyme-linked immunosorbent assay (ELISA) kit was the produced of Shanghai Senxiong Science and Technology Co.,Ltd. Human recombinant TGF- β1 was produced by R&D Company. DNA Engine OpticonTM real-time fluorescence quantitative polymerase chain reaction apparatus was the product of MJ Research Company. METHODS: Rat renal tubular epithelial cells (NRK-52E) cultured in vitro were divided into five groups. Control group was added with serum-contained DMEM; TGF-β1-induced group was added with TGF-β1 at final concentration of 5 ng/L; SF at different concentrations groups were added with 125, 250, 500 μ mol/L SF and TGF- β1 at final concentration of 5 ng/L,respectively. MAIN OUTCOME MEASURES: The contrast phase microscope, real-time fluorescence quantitative polymerase chain reaction and ELISA method were used to detect TEMT of NRK52E cells induced by TGF-β1 and levels of collagen Ⅰ, collagen Ⅲ and fibronectin in the supernatant. RESULTS: Morphology of NRK52E cells: Compared with control group, TGF-β1 could induce the transdifferentiation of NRK52E cells, showing fibroblast-like in morphology after 3 days, which were previously the typical road stone-like epithelial cells. In three different concentration SF groups, the morphologic transformation stimulated by TGF-β1 could be partly ameliorated in a dose-dependent manner. Expression of α-SMA mRNA: Compared with control group, 5 ng/L TGF- β1 enhanced expression of α-SMA at 6 hours, and reached a peak at 72 hours; SF depressed the expression in a dose-dependent manner at 72 hours (P < 0.05). Changes of ECM: After induced by 5 ng/L TGF- β1 for 72 hours, the levels of collagen Ⅰ, collagen Ⅲ and fibronectin in the supernatant increased significantly (P < 0.05), whereas SF decreased these levels in a dose-dependent manner (P < 0.05). CONCLUSION: TGF- β1 induces the TEMT, and promotes the secretion of collagen Ⅰ, collagen Ⅲ and fibronectin. SF can inhibit TGF- β1-induced TEMT In a dose-dependent manner.
9.Effects of Helicobacter pylori on the expression of catalytic subunit of the DNA-dependent protein kinase and Ku70/Ku80 heterodimer in gastric mucosa epithelial cells in vivo and in vitro

Objective To explore the effects of Helicobacter pylori (H.pylori) on the expression of catalytic subunit of the DNA-dependent protein kinase (DNA-PKcs) and Ku70/Ku80 heterodimer in gastric mucosa epithelial cells in vivo and in vitro.Methods After treated with H.pylori for one,three,six,12 and 24 hours,the expressions of DNA-PKcs and Ku70/Ku80 heterodimer in gastric epithelial cells (GES) 1 and gastric adenocarinoma cells (AGS) were detected by Western blot.Mongolian gerbils were gavaged with H.pylori,and were sacrificed after infected for six and 12 months.The gastric mucosa tissues were taken for immunohistochemistry to detect the expressions of DNA-PKcs and Ku70/Ku80 heterodimer at protein level.The data were analyzed by t test and chi-square test.Results After H.pylori infection for one hour,the relative quantity of the expression of DNA-PKcs in GES-1 was 1.16±0.09,which was higher than that of non infected group (1.04±0.31) and the difference was statistically significant (t=4.67,P<0.05).After infected by H.pylori for one,three,six,12 and 24 hours,the relative quantities of the expressions of Ku70/Ku80 heterodimer in GES-1 were 1.58±0.32,1.84±0.40,1.97±0.35,3.72±1.42 and 3.74±1.56,respectively,all were higher than that of non infected group (1.24±0.31) and the differences were statistically significant (t=3.57,4.20,5.03,8.11 and 8.14,all P<0.05).The relative quantities of the expressions of Ku70/Ku80 heterodimer in AGS were 4.69 ± 0.87,3.67 ± 0.67,2.41±0.24,1.35±0.35 and 1.32±0.10 after H.pylori infected for one,three,six,12 and 24 hours,respectively,all were lower than that of no H.pylori infected group (4.84 ± 0.76) and the differences were statistically significant (t=34.13,27.68,19.81,4.47 and 5.69,all P<0.05).In Mongolian gerbil models,DNA-PKcs did not express in H.pylori negative group (0/25),the total positive rate of H.pylori infected group was 98.1% (53/54),the difference between the two groups was statistically significant (x2 =74.55,P<0.01).The total positive rate of Ku70/Ku80 heterodimer in H.pylori negative group was 92.0% (23/25) and in H.pylori infected group was 68.5% (37/54),the difference between the two groups was statistically significant (x2=5.16,P<0.05).Conclusion H.pylori infection affected cellular DNA damage repair through changing the expression of DNA-PKcs and Ku70/Ku80 heterodimer in gastric mucosa in vivo and in vitro,which may cause gastric mucosal lesions.
