ObjectiveTo explore the molecular mechanism that LRIG1 inhibits signal transduction system of epidermal growth factor receptor(EGFR)by investigating the role of LRIG1 in glioma.MethodsThe plasmid pcDNA3.1-LRIG1 was transfected into primary glioma cells by Lipofectamine.Then,the changes of LRJG1 and EGFR in the transfected glioma cells were measured by RT-PCR and Western blot,and the cell proliferation and apoptosis were analyzed by MTT and flow cytometry.ResultsThe expression levels of LRIG1 mRNA and protein in the glioma cells transfected with pcDNA-LRIG1 were significantly higher than those of control group and pcDNA3.1 transfected glioma cells,while those of EGFR mRNA and protein were significantly lower.The expression of PKC? and Bax was up-regulated,while the expression of bcl-2 was down-regulated.The growth of glioma cells was inhibited and their apoptosis was obviously enhanced.ConclusionBy participating the construction of the negative feedback loop of EGFR,LRIG1 inhibits the occurrence and growth of tumor through several pathways.

Objective To investigate the expression of apoptosis-related proteins in rat cerebral cortex following traumatic brain injuries(TBI)and discuss the role of oxygen free radical-mitochondria signal pathway in Edaravone treating TBI.Methods A total of 180 male adult Sprague-Dawley rats were randomly divided into TBI group,Edaravone treatment group and control group.Each group was divided into six subgroups at 1,3,6,24,48 and 72 hours after TBI.Edaravone treatment group was injected with Edaravone(10 mg/kg)and the other two groups injected with the same volume of 0.9%normal saline.The pathological change in the rat cortex following TBI was observed with HE staining.At different time points,the expressions of Cytc,Bcl-2 and Bax in rat cortex as well as cell apoptosis and MDA change were observed by means of immunohistechemistry,TUNEL and TAB.Results HE staining showed scattered degenerated and necrotic neurous in cerebral cortex six hours after neuron injury,which peaked at 24 hours.Compared with control group,intermediate product MDA of free radical was increased six hours after TBI and peaked at 48 hours in Edaravone treatment group,which was lower than TBI group especially at 24,48 and 72 hours(P＜0.05).Compared with control group,the immunity reaction of Cytc positive cells inereased at six hours and peaked at 24 hours in TBI group,with statistical difference at 3,6,24,48 and 72 hours(P＜0.05).Compared with TBI group,the immunity reaction of Cyte positive cells was decreased obviously at 24,48 and 72 hours in Edaravone treatment group.Hyperexcitability of Bcl-2 after TBI reached peak at 3 hours and decreased gradually.But the expression of Bax was increased gradually after TBI and peaked at 48 hours,when Bax/Bcl-2 reached peak too.Folowing TBI,TUNEL positive cells increased gradually and reached peak at 48 hours,with mainly type Ⅰ TUNEL cells before 24 hours and typeⅡTUNEL cells after 24 hours.Conclusions There exist necrosis and apoptosis of nerve cells in cortex after TBI,especially apoptosis.Oxygen free radical mitochondria is one of the signal transduction pathways of nerve cell apoptosis following TBI.Edaravone exerts certain therapeutic effect on TBI.

Objective To explore the effect of Mycobacterium tuberculosis antigen (Mtb-Ag) on neutrophils apoptosis.Methods The fresh isolated neutrophils from healthy adults blood were cultured with Mtb-Ag for 24 h,with or without pretreatment of nuclear factor -?B (NF-?B) inhibitor N-tosyl-L-phenylanyl chloromethyl ketone (TPCK) for 30 minutes.Annexin V staining and Flow cytometry were used to measure cell apoptosis of neutrophils.NF-?B DNA binding was measured by gelelectrophorestic mobility shift assay (EMSA) in neutrophils after incubated with Mtb-Ag for 0,1,2,4,6,24 hours.Results Comparing to the spontaneous apoptosis (55%?6%) of neutrophils after culture in vitro for 24 h,treatment of Mtb-Ag (1.125 mg/ml) decreased the cell apoptosis of neutrophils (32%?3%).The NF-?B shift bands were detected at 1 h in neutrophils after stimulated by Mtb-Ag,and reached maximum peak at 2 hours,and then returned to basal levels within 24 h.Pretreatment of TPCK inhibited the anti-apoptosis role of Mtb-Ag in neutrophils.Conclusion Mtb-Ag prevents neutrophils apoptosis and its inhibitory role concerns NF-?B pathway.

Objective To explore the clinical efficacy of single-port Da Vinci robotic surgical system in the radical resection of rectal cancer.Methods The retrospective descriptive study was conducted.The clinical data of one patient with rectal cancer who was admitted to the Southwest Hospital of the Third Military Medical University in June 2017 was collected.The patient underwent the first case of radical resection of rectal cancer with the single-port la Vinci robotic surgical system,and intraoperatively received laparoscopy-assisted digestive tract reconstruction.Observation indicators:(1) intra-and post-operative situations;(2) follow-up and survival.Follow-up using outpatient examination and telephone interview was performed to detect patients' survival up to July 2017.Results (1) Intra-and post-operative situations:the patient underwent successful single-port Da Vinci robotic radical resection of rectal cancer.The operation time and volume of intraoperative blood loss were 145minutes and 20 mL,without blood transfusion.The using time of nonsteroidal analgesic drug,time for out-of-bed activity,time to anal exsufflation,time of fluid diet intake and time of pelvic drainage tube removal were 1 day,24hours,48 hours,72 hours and 4 days after operation,respectively.No postoperative complication was detected.Results of postoperative pathological examination:moderate-differentiated rectal adenocarcinoma invaded serosal layer,with negative circumferential margin,negative distal margin of rectum and mesenteric lymph node metastasis (1 of 16 harvested lymph nodes was positive).Pathologic staging was T4aN1M0 (stage Ⅲb).The patient was discharged from hospital at postoperative day 6.(2) Follow-up and survival:the patient was followed up for 1month,with a good survival.Conclusion Single-port Da Vinci robotic surgical system in the radical resection of rectal cancer is safe and feasible,with good short-term outcomes.

For the current problems of the development of clinical pathways, a solution of clinical pathway information based on data mining is put forward in the paper. Using data mining techniques, the clinical pathway adapt to the conditions of the hospital is mined from the large amount of data available from the hospital, which reduces the development cycle and cost. The solutions of clinical path information based on data mining engine links seamlessly with existing hospital systems, which will provide a new way of thinking to the development of clinical pathways.

OBJECTIVETo investigate the clinical outcomes of cannulated screw combined with buttress plate in treating transarticular shear fractures of the distal humerus.

METHODSFrom July 2008 to December 2011, 17 patients with shear fractures of the distal humerus were treated with cannulated screw combined with buttress plate. Among them, included 10 males and 7 females aged from 20 to 59 years old (mean 36.5). All the fractures were classified into type I (8 cases), type II (1 case), type III (3 cases) and type IV (5 cases) according to Bryan-Morrey and McKee classification. According to AO/ASIF classification, 10 cases were type 13B3.1, 7 cases were type 13B3.3; according to Dubberley classification, 5 cases with type 1A, 2 cases with type 1B, 2 cases with type 2A, 2 cases with type 2B, 4 cases with type 3A, 2 cases with type 3B. There were 12 cases with fresh injuries, 4 cases with old injuries. All fractures were closed injuries. Complications and range of motion of elbow were observed, and clinical efficacy were evaluated by Broberg-Morrey standard.

RESULTSAll patients were followed up with a mean time of 21.3 months (ranged, 18 to 24). All fractures obtained bone healing from 10 to 15 weeks with a mean of 12.5 weeks. No malunion or delayed healing occurred. Two cases occurred ossification in lateral collateral ligament attachment. 2 cases occurred heterotopic ossification in the capitellum in front of capsule joint. The range of elbow extension was (16 +/- 7) degrees, flexion was (115 +/- 9) degrees, the average are of pronation and supination was (65 +/- 5) degrees and (60 +/- 5) degrees respectively. According to Broberg-Morrey standard, there were excellent in 7 cases, good in 8 cases, fair in 2 cases, and average score was 92.44 +/- 4.64.

CONCLUSIONFor transarticular shear fractures of the distal humerus, classification should be performed according to preoperative X-ray and CT, cannulated screw and buttress plate combined and early function rehabilitation could recover elbow function.
