1. Expression of GATA 4 and alpha fetoprotein in fetal mouse liver and hepatocarcinoma cells and their relationship

Objective: To explore the expression of GATA4 and alpha fetoprotein (AFP) in fetal liver and hepatocarcinoma cells and their correlation. Methods: Semi-quantitive RT-PCR was used to detect the expression of GATA4 and AFP in liver tissues from mouse of E 10.5 d to P 0.5 d, normal liver cells, and hepatocarcinoma cells. Eukaryotic expression vector of mouse GATA4 was used to transfect normal liver cell line and the product was identified. Doxorubicine was used to treat hepatocarcinoma cells; then semi-quantitive RT-PCR was used to determine the mRNA of GATA4, FTF and AFP; Western blot was used to examine the expression of GATA4 and AFP; and ELISA was used to determine AFP in supernatants. Results: The expression of GATA4 and AFP decreased with the maturation of fetal liver cells and reached adult level 0.5 d after birth. GATA4, FTF and AFP were not detected in normal liver cells but were overexpressed in hepatocarcinoma cells. The expression of FTF and AFP was increased in normal liver cells after transfection of GATA4, and the level of AFP was also increased in the supernatant. GATA4 was sharply decreased in hepatocarcinoma cells within 6 h after doxorubicine treatment; the expression of FTF and AFP was also decreased in supernatants 12 h after doxorubicine treatment (P<0.05). Conclusion: GATA4 is highly expressed in fetal liver and hepatocarcinoma cells,which may contribute to the elevation of AFP expression.
2. Diagnostic value of serum Cys-C, hs-CRP, and urine mALB for early renal injury in gonty patients

Objective To explore the diagnostic value of serum Cys-C, hs-CRP, and urine mALB for early renal injury in gonty patients. Methods A total of 100 gonty patients and 50 healthy volunteers were divided into 3 groups, acute phase group (n = 50), intermittent group (n= 50), and healthy control group (n = 50) . The serum Cys C,hs-CRP,and urine creatinine were determined by automatic chemistry analyzer and urine mALB by immunofluorometer equipment, and the results were analyzed in each group. Results The results of mALB/Cr(mg/[g • Cr]), hs-CRP(mg/L), and Cys-C(mg/L)in acute phase group were 11.45±3.55, 5.15±1.98, and 2.62±0.83, in intermittent group were 8.96±1.78, 3.28±1.23, and 0.93±0.26, and in healthy volunteer group were 3.89± 0.76, 1.91±0.57, and 0.78±0.22, respectively. The values of the above three parameters in the acute phase and intermittent groups were significantly higher than those in the healthy control group (P< 0.05) ; and the values in the acute phase group were significantly higher than those in the intermittent group (P<0.05). The abnormal rate of hs-CRP+ Cys-C+ mALB/Cr was 87% in gonty patients. Conclusion Simultaneous determination of mALB/ Cr, hs-CRP, and Cys-C can help to assess the early renal injury in gonty patients.
3.Changes of Hemorheology of Children with Essential Hypertension

Objective To explore the hemorheological changes in children with essential hyepertension,and explore their clinical significance.Methods The hemorheological parameters,blood lipid,blood glucose in 37 children with essential hyepertension [19 boys and 18 girls,median age(12.49?3.20) years] were measured and analyzed.The parameters in 30 healthy children were measured as controls.SPSS 11.5 software was used to analyze the data.Results The whole blood viscosity under high,middle and low shear rate,plasma viscosity,hematocrit,rigidity index of erythrocyte,platelet aggregation M were significantly higher in hypertension group than those in healthy control group(Pa0.05).Conclusions There have been developed significant hemorheological changs in children with essential hypertension.The hemorheological changes might play an important role in pathogenesis of childhood hypertension.
4.Protective effect of pulsed magnetic field against cerebral ischemia-reperfusion injury in rats

Objective To examine the neuroprotective effect of pulsed magnetic field in a animal focal cere-bral ischemica-reperfusion injury model. Methods Forty-eight Sprague-Dawley (SD) rats were randomized into 3 groups, a sham-operation group, a model group and a pulsed magnetic field group, with rats 16 in each group. Mid-dle cerebral artery occlusion (MCAO) method was employed to establish the focal cerebral ischemia-reperfusion inju-ry model in rats of model group and pulsed magnetic field group. Rats in sham-operation group was subject to the same operation procedure but not underwent ischemia-reperfusion. The infarction volume, histopathological damage and expressions of IGF-1 in ischemic brain tissue were investigated to evaluate the effect of pulsed magnetic field. Results The infarction volume was reduced, histopathological damage alleviated and expressions of IGF-1 in ische-mic brain tissue elevated in the pulsed magnetic field group as compared against the model group (P<0.05). Con-clusions Pulsed magnetic field might provide neuro-protection against cerebral ischemia-reperfusion injury.
6.Tumor necrosis factor-αupregulates MMP9 expression through site-specific DNA demethylation

Objective To determine the involvement of DNA demethylation in tumor necrosis factor-α(TNF-α)-induced matrix metalloproteinase 9 ( MMP9) expression in human epidermal keratinocytes. Methods Real-time RT-PCR, Western blot, and enzyme-linked immuno sorbent assay (ELISA) were performed to determine the mRNA and protein levels of MMP9 after human keratinocyte cell line (HaCaT) cells were treated with 10 ng/ ml TNF-α or 2. 5 μmol/ L DAC/ 300 nmol/ L TSA. Bisulfite sequencing PCR ( BSP) and Methylation-sensitive high-resolution melt analysis ( Ms-HRM) were used to detect significantly differentially demethylated CpG sites in the human MMP9 promoter region in cells exposed to TNF-α. Different sites methylation constructs of promoter-luciferase reporter gene were made to detect the influences of site-specific DNA demethylation on transcription activity of MMP9 promoter. Results Compared with PBS-treated control, TNF-α significantly increased the expression of MMP9 in HaCaT cells for indicated culture duration ( P < 0. 05 ). Real time PCR, Western blot, and ELISA analysis demonstrated that the mRNA and protein levels of MMP9 were increased initially, followed by a decline with prolonged incubation time. After TNF-α treatment, varied degrees of DNA demethylation occurred at 10 CpG sites in the promoter of MMP9, and the changes at the -36 bp site were statistically significant (P<0. 05). The demethylation at the -36 bp site greatly increased the transcription activity of MMP9. Conclusion TNF-α promotes MMP9 expression in HaCaT cells through inducing -36 bp site DNA demethylation on the promoter of MMP9.
7.Efficacy of domestic gemcitabine combined with Lobaplatine in salvage treatment of taxanes-refractory advanced lung squamous carcinoma

Objective To evaluate the effiacy and adverse reactions of domestic gemcitabine combined with lobaplatine in salvage treatment of taxanes-refractory advanced lung squamous canceroma.Methods 71 patients with squamous carcinoma of lung,all had been failer to combined chemotherapy with taxanes treatment before,received the treatment with domestic gemcitabine (1 000mg/m2,d1,8) combined with lobaplatine (30mg/m2,d2),21-28 days as a cycle,at least 2 cycles should been recieved.The therapeutic efficacy was evaluated after 2 cycles of chemotherapy,according to RECIST1.0 standard.Results All of 175 cycles had been observed.In terms of the treatment efficacy,68 patients could be evaluated objectively of 71 cases patients,partial response (PR) was 15 cases (22.0%),stable disease(SD) was 18 cases(26.4%),with an overall response rate(RR) was 22.0% (15/68),disease control rate (DCR) was 48.5 % (33/68).The median time to progression (TTP) was 13.6 weeks.the median survival time (OS) was 7.9 months.One year survival rate was 42.6% (29/68).On 71 cases for a total of 175 cycles of chemotherapy,the major toxic reaction was hematological toxicities,including myelosuppression about 38.3% (67/175) for grade Ⅲ and Ⅳ.Conclusion The combination treatment of domestic gemcitabine plus lobaplatin is a feasible and active scheme in salvage treatment of taxanes-refractory advanced quamous carcinoma of lung.
10.Relationship between Diabetic Retinopathy and Peripheral Arterial Disease in Type 2 Diabetes Patients

[Objective]To explore the relationship between peripheral arterial disease(PAD)and diabetic retinopathy(DR)in type 2 diabetes patients.[Methods]A total of 99 patients diagnosed with PAD were classified into grade 1-3 by their total scores of peripheral arterial stenosis assessed by color doppler ultrasound examinations,where the degree of stenosis 30% ~ 49% scored 0, 50%~99%scored 1,lumen occlusion(i.e. degree of stenosis 100%)scored 2,and therefore the total score 0-2 was categorized into Grade 1 ,3~4 into Grade 2 ,5~12 into Grade 3. The bilateral anterior tibial artery ,posterior tibial artery and dorsalis pedis artery of these patients were analyzed. The presence of diabetic retinopathy(DR)was graded from retinal photographs using a standard protocol.[Results]Among 99 cases of type 2 diabetic patients with peripheral arterial disease ,58.6%of them were male with average age of 67.3 ± 7.9 years old. Patients of Grade 1,Grade2,Grade 3 lesion accounted for 45.4%,30.3%,24.2%,respectively. Age, gender,smoking history,SBP,DBP,BMI,FBG,TC,TG,LDL-C,HDL-C,HbA1C among 3 groups were not statistically signifi-cant. The associations of DM duration and HbA1C value were significantly larger in DR than in PAD. The proportion of DR patients increased with the severity degree of PAD(p for trend=0.004). Degree of stenosis Grade 2 and Grade 3 could be predictive for DR.[Conclusions]DR is associated with the severity degree of PAD in type 2 diabetes patients as evaluated by duplex ultrasonography.Degree of stenosis Grade 2 and 3 could be used for screening or finding DR. Strategies for optimum treatment and early prevention are needed.
