1. Chemical constituents of Opuntia milpa (I)

Objective: To investigate the chemical constituents of Opuntia milpa. Methods: The compounds were isolated by repeated silica gel chromatography and were elucidated by chemical and spectral analyses. Results: Ten compounds were isolated from the ethyl acelate extraction of O. milpa and identified as β-sitosterol (1), 1-heptadecanol (2), 5-hyadroxymemyl-2-furancarboxaldehyde (3), syringaldehyde (4), vanillin (5), p-hydroxybenzaldehyde (6), coniferyl aldehyde (7), p-hydroxyl-cinnamaldehyde (8), quercetin-3-methyl ether (9), and 2,6-dimethoxyphenol (10). Conclusion Compounds 3-10 are isolated from O. milpa for the first time.

Objective To study transforming growth factor ?_1 (TGF-?_1) expression in the lung of neonatal premature rats exposed to hyperoxia at different time. Methods 99 neonatal premature rats were assigned to hyperoxic group and air group randomly They were sacrificed at the postnatal 3 rd ,7 th and 14 th day,then their lungs were achieved for H.E staining and immunohistochemistry to measure the expression of TGF-?_1. The difference of TGF ?_1 expression were tested by rank-sum test. Objective To study the effects of Nitric oxide(NO),Nitric oxide synthases (NOS)on calcium-activated potassium channels (KCa) of mesenteric artery smooth muscle(SMC)between patients with essential hypertension (EH) and normotensive patients.Methods (1)Mesenteric artery branch from 21 EH and 18 normotensive patients respectively was digested by enzyme.Patch clamp technique was used to pull cell-attached and inside-out patches on mesenteric artery SMC from EH.The signal channel open number probability (Po),open dwell-time(To) and close dwell- time(Tc),were recorded.(2)The levels of plasma NO?NOS were measured by colorimetry analysis method.(3)The analysis of linear correlation was performed to estimate the relationship between the levels of plasma NO and Po?To?Tc of KCa of mesenteric artery SMC in the subjects of the two groups.Results (1)Compared to that of normotensive patients,the activities of KCa channels of patients with EH was higher.After adding Ca 2+ to cytoplasm,the Po of KCa channels in normotensive patients increased significantly.But there were few changes in EH group.(2)The levels of plasma NO?NOS were lower in EH group than in control group.(3)Linear correlation analysis showed that the levels of plasma NO were correlated positively with Po?To in normotensive patients,wherese the positive correlation were decreased in EH group.Conclusion The activities of KCa channels of patients with EH increase significantly.but the sensitivity to Ca 2+ decreased.NO?NOS may stimulate the KCa channels of normotensive and EH patients,but they may not be main factors in EH patients.

8.Effect of Conditioned Medium from Endothelial Cells on Cancer Stem Cell Phenotype of Hepatoma Cells.

In this study, we aimed to investigate the influences of conditioned medium from human umbilical vein endothelial cells (HUVEC) on cancer stem cell phenotype of human hepatoma cells. HUVEC and human hepatoma cells (MHCC97H) were cultured, respectively, and then the MHCC97H cells were co-cultured with conditioned medium from HUVEC (EC-CM) with Transwell system. Anti-cancer drug sensitivity, colony-formation, migration/invasion ability, expression of cancer stem cell marker and sphere formation were performed to determine the cancer stem cell phenotype in MHCC97H cells. We found that MHCC97H cells co-cultured with EC-CM exhibited significantly higher colony-formation ability and lower sensitivity of anti-cancer drugs 5-FU and Cis. Transwell assay showed that treatment with EC-CM obviously increased migration and invasion of MHCC97H cells. Moreover, increased sphere forming capability and expression of CD133 in MHCC97H cells were observed after co-cultured with EC-CM. These results suggested that EC-CM could promote cancer stem cell phenotype of hepatoma cells.

9.Risk factors of aggressive posterior retinopathy of prematurity:a retrospective,longitudinal study

Objective To analyze the risk factors of aggressive posterior retinopathy of prematurity (AP-ROP). Methods A retrospective case-control study was applied. Questionnaire was designed through literature research to collect data of AP-ROP and ordinary ROP(except for AP-ROP)from March,2012 to September, 2014 in the department of neonatology in Guangdong Women and Children′s Hospital. Based on the ratio of 1:2 , the two groups were selected with the matching conditions:the closest date of admission in hospital,ordinary ROP children,collection and analysis of the data. Results Univariate analysis:gestational age(P<0.05),birth weight(P<0.05),oxygen therapy days(P<0.05),days of mechanical ventilation(P<0.05),gestational age<32 w(P<0.05),birth weight<1250 g(P<0.05),oxygen(P<0.05),oxygen≥14 d(P<0.05),mechanical ven-tilation(P < 0.05),mechanical ventilation ≥ 14 d(P < 0.05),PS(P < 0.05),neonatal anemia(P < 0.05), blood transfusion≥2 times(P<0.05),RDS(P<0.05),neonatal pneumonia(P<0.05),apnea(P<0.05), BPD(P < 0.05),intracranial hemorrhage(P < 0.05),metabolic acidosis(P < 0.05),and retinal hemorrhage (P < 0.05) were related to AP-ROP. Multivariate analysis with conditional logistic regression analysis indicated that blood transfusion ≥2 times(OR:14.956,95%CI:4.328~51.684)and apnea(OR:7.590,95%CI:2.507~22.978)were regarded as the independent risk factors for AP-ROP. Conclusions AP-ROP is associated with low gestational age,low birth weight,oxygen therapy and related complications including RDS and retinal hemorrhage. Blood transfusion and apnea are important independent risk factors of AP-ROP.
10.Efficacy of chitosan on rehabilitation treatment after sinuses open surgery

Objective To study the efficacy of chitosan on rehabilitation treatment after sinuses open surgery .Methods Fifty-six cases patients after sinuses open surgery were collected during June 2013 to October 2014 and divided into experimental group and control group by random sampling method, 28 cases in each group.The experimental group were smeared with chitosan on surface after cleaning by normal saline, while control group cleaned by normal saline alone.The efficacy, adhesion of nasal mucosa and time of mucosa complete epithelialization in two groups at three and six months were analysed while out-patient review.ResuIts After three and six months, the total effective rate of experimental group was significantly higher than that of control group[85.71% vs.60.71%(χ2 =4.46,P<0.05);92.86% vs.67.86%(χ2 =5.54,P<0.05)].The time of mucosa complete epithelialization in experimental group was (50.4 ±6.5)d, which was shorter than (62.3 ±7.5)d of control group (t=6.34,P<0.05).After three and six months, the nasal mucosa without adhesion in experimental group was better than that in control group (χ2 =5.60, χ2 =5.49;P<0.05). ConcIusion Chitosan can promote nasal mucosa epithelialization, reduce adhesion of nasal mucosa, improve the postoperative curative effect of chronic nasal-sinusitis.
