Aim To investigate the effects of ginsenoside(GS) on phosphorylation of tau protein, microtubule, cellular apoptosis and its related factors in cellular model of Alzheimer disease(AD) induced by the protein phosphatase 1 and 2A inhibitor okadaic acid(OA).Methods The human neuroblastoma cell line SK-N-SH cells were cultured with GS for 24 h,the culture medium was changed, and then incubated with OA 10 nmol?L-1 for 6 h.The changes of cell morphology were observed by inverted microscope. The laser confocal microscopy was used to observe the microtubule changes.Western blot was applied to determine the expression of phosphorylation of tau protein, and apoptosis-regulating factors Bcl-2,Bax and Caspase-3.The changes of apoptotic cells were observed by TUNEL method. Results The normal SK-N-SH cells spread well.OA-treated cells showed that the cell axons and the microtubules were broken and decreased under the inverted microscope and laser confocal microscope.Preincubation of GS demonstrated the significantly protective effects against the morphologic damage induced by OA.In OA-treated group, the phosphorylation of tau protein at Ser-199/202 and Ser-404 sites was higher than that in normal group, and the non-phosphorylation of tau protein at the same sites was lower;Incubation of GS at the dose of 50 mg?L-1 and 100 mg?L-1 with the cells decreased the phosphorylation of tau protein Ser-199/202 and Ser-404 sites.GS group at the dose of 50 mg?L-1 and 100 mg?L-1 decreased the expression of at non-phosphorylation of tau protein at the Ser202 site. The apoptotic cells were not found in normal group. The number of apoptotic cells were obviously increased.the expression of Bax and caspase-3 significantly enhanced, and Bcl-2 expression decreased in the OA-treated model group.GS significantly decreased the apoptotic cell number of nerve cells, inhibited the expression of Bax and caspase-3.Conclusion GS can protect the nerve cells from pathological change induced by OA.Maybe because it can inhibit the hyperphosphorylation of tau protein and protect the nerve cells from apoptosis, thus GS may have potential to treat Alzheimer disease.

The theory that "the kidney nourishes marrow and brain is the sea of marrow" has been instructing traditional Chinese medical doctors in preventing and treating dementia in aged people for thousands of years. However, the modern biological basis of this theory has not been systemically studied. In this review, we summarized our serial pharmacological studies on the treatment of Alzheimer's disease (AD) with Chinese medicinal herbs, using multiple kinds of AD-like animal and cell models. The results show that "kidney-reinforcing" herbs of traditional Chinese medicine play a very important role in the anti-AD effects; and different "kidney-reinforcing" herbs have different effects in brain functions. The common effects of "kidney-reinforcing" herbs are improving cellular energy metabolism, increasing neurotrophic factors and the number of cholinergic neurons, and decreasing neurotoxin production. Based on above results, we propose that the essence of "brain marrow" is the neurons and neurotrophic factors in the brain; "the deficiency of brain marrow" is induced by the decrease in neurotrophic factors and the atrophy and loss of neurons in the brain, thus resulting in cognitive impairment and dementia. The modern biological basis of "reinforcing kidney to replenish marrow" by traditional Chinese medicine includes improving cellular energy metabolism and utilization, enhancing endogenous neurotrophic effects and decreasing neurotoxin production, thus reducing the cell death and increasing the survival and regeneration of neurons.

Chinese Journal of Pediatrics 2019;57(2):108-112
Objective:
To investigate the application of coronary angiography (CAG) in children with Kawasaki disease complicated with coronary artery lesion (CAL).
Methods:
A total of 139 patients (115 boys and 24 girls, age (48±42) months) with Kawasaki disease complicated with CAL underwent CAG between June 2006 and June 2018 in Children′s Hospital of Fudan University. CAL in all children were at grade Ⅲ and above assessed by echocardiography and underwent CAG under general anesthesia with parental consent. A retrospective analysis was performed to identify the safety and complication of CAG, degree of satisfaction of image, distribution of CAL, and the treatment and follow-up management according to the result of CAG.
Results:
All the 139 patients underwent CAG, and 17 patients received CAG twice. In these cases, there was a total of 309 CAL with an average of 2.22 CAL per patient. After CAG, children were re-graded as grade Ⅲ in 33 cases, grade Ⅳ in 56 cases, grade Ⅴa in 33 cases and grade Ⅴb in 17 cases according to the clinical severity. Ten patients (7.2%) had only aortic root angiography and 129 (92.8%) patients had selective left or right CAG. Among these, 27 cases (19.4%) showed thrombosis in coronary aneurysm, 23 cases (16.5%) showed coronary artery stenosis, 4 cases (2.9%) showed recanalization after occlusion in right coronary artery. All the patients obtained satisfied images, and no complication was found. Eight of the grade Ⅴb patients underwent coronary artery bypass grafting (CABG). One patient died during the surgery due to severe heart failure, and the other seven patients received CABG successfully and recovered well during a long-term follow up (18 to 108 months).
Conclusions
CAG is safe for children with Kawasaki disease with CAL, and the images of CAG are satisfied. Patients who were graded according to CAG and received the corresponding surgical treatment could get satisfied effect in the medium and long-term follow-up.
