1.The clinical application of contrast-enhanced ultrasound-guided radiofrequency ablation in treating iso-echo liver cancer

Objective To investigate the detection rate of contrast-enhanced ultrasound (CEUS) for iso-echo liver cancers, and to evaluate the application of CEUS in treating iso-echo liver cancers with radiofrequency ablation (RFA). Methods A total of 27 patients with liver cancers (31 lesions in total) were enrolled in this study. Of the 27 patients, hepatocellular carcinoma (HCC) was seen in 11 (11 lesions) and hepatic metastasis in 16 (20 lesions). In all patients the diagnosis was detected by contrast-enhanced CT or contrast-enhanced MRI and was clinically confirmed. Under real-time CEUS guidance, RFA was carried out for all the 31 lesions. The ablated extent was evaluated by CEUS, and CEUS findings were used to guide the further treatment. Results All 31 lesions were detected by CEUS and were treated with RFA. No serious complications occurred after RFA. Both CEUS and CECT performed one month after RFA showed that complete tumor necrosis was seen in all the 31 lesions. After RFA the serum AFP levels in all 11 HCC patients fell to normal range, and the serum CEA, CA-199 as well as CA-153 levels in patients with hepatic metastasis decreased significantly(P<0.01). Conclusion CEUS can markedly increase the detection rate of iso-echo liver cancers. Besides, CEUS plays an important role in guiding the performance of RFA for iso - echo liver lesions.
2.Screening result analysis of diabetes in high-risk population in Songjiang District of Shanghai, 2016-2017

Objective To investigate the prevalence of diabetes and impaired glucose regulation in high risks community residents in Songjiang District of Shanghai, and to explore the risk factors affecting their blood glucose metabolism, providing effective suggestions for improving community diabetes prevention and treatment. Methods Questionnaire, physical examination and laboratory testing were used to collect information on the basic characteristics and blood glucose levels of 21 035 residents in Songjiang District who were assessed to be at high risk of diabetes. Results A total of 3 008 people with impaired regulation and 2 241 patients with diabetes were detected.The detection rates were 14.3% and 10.6%, respectively.It was found that as the residents′ age was higher, their education level was lower, high-risk factors for them were numerous, and their detection rates of diabetes and impaired sugar regulation were higher.It was found that age, gender, history of impaired glucose regulation, relatives with type 2 diabetes, hypertension, dyslipidemia and overweight/obesity were all factors influencing blood glucose metabolism (
3.Analysis of influencing factors for pregnancyinducedhypertension retinopathy and its influence on pregnancy outcome of mothers and infants

AIM: To investigate the risk factors for pregnancy induced hypertension retinopathy, and analyze its influence on maternal and child pregnancy outcomes. METHODS: A total of 100 patients with gestational hypertension who met the requirements of screening were selected, they were divided into two groups, without retinal lesion group (n = 29) and retinopathy group (n=71). The age, course of disease, gestational age, blood pressure, proteinuria and hematocrit in two groups were investigated. The risk factors were detected by single factor analysis, and then the study on independent risk factors were analyzed by Logistic regression analysis. At the same time,their influence on maternal and child pregnancy outcomes were investigated. RESULTS: The single factor analysis showed that except for age, the differences in the course of disease, gestational age, blood pressure, proteinuria and hematocrit were statistically significant (P < 0. 01). Logistic regression analysis showed that the course of disease, blood pressure, proteinuria and hematocrit were the risk factors of retinopathy, gestational age was a protective factor. Compared with the group without retinopathy, the incidence of preterm birth, placental abruption, postpartum hemorrhage, perinatal death in the patients with retinopathy significantly increased, and the difference were statistically significant(P<0.01),and there was a certain correlation between the extent of increase and the grade of retinopathy. CONCLUSION: Course of disease, blood pressure, proteinuria and hematocrit of patients with the hypertension of pregnancy are the risk factors of retinopathy,retinopathy has a serious adverse effect on maternal and child pregnancy outcomes.
4.Transgenic 4-1-BB ligand therapy induces tumor specific immune response in oral squamous cell carcinoma

Objective To examine the activation and cytotoxieity of human peripheral blood T lymphocyte induced in vitro by human 4-1-BB ligand(4-1-BBL) gene transfected into tumor Tca8113 cells.Methods The eukaryotic expression vector pEGFP-h4-1-BBL was transfected into human oral carcinoma cell line Tca8113 by Lipofectamine<'TM>2000. The transfected cells were then selected in medium containing G-418, cloned by limited dilution and named as Tca8113-4-1-BBL Human 4-1-BBL mRNA and protein expression of transfected cells was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and western blotting respectively. The tumor cell vaccines (TCV) were obtained by treatment with mitomycin (MMC). Human peripheral blood mononuclenr cells (PBMC) were prepared from lymphoprep, and then stimulated with anti-CD-3 mAb and incubated with non-transfected or transfected TCV-TcaSI13 cells,respectively. The proliferation of T cells was evaluated by trypan blue exclusion; the CCK-8 was used to detect the cytotoxic effect of T lymphecytes. Meanwhile, the secretion of intefferom-γ (IFN-γ) and interleukin(IL)-2 in culture supernatant was detected by enzyme-linked immunosorbnent assay (ELISA).Results The Tca8113 cells trasfected by pEGFP-h4-1-BBL could express human 4-I-BBL efficiently. As compared with wild type Tca8113 cells, the transfected Tca8113 cells could markedly promote proliferation,IL-2 and IFN-γ production and cytotoxic activity of lymphocytos. Condusions The transfection of human 4-1-BBL gene in Tca8113 cells is effctive in enhancing its immunogenicity and inducing antitumor immune response in vitro.
5.Development and reliability and validity analysis of hospice care attitude scale for medical workers

Objective To develop a hospice care attitude scale for medical workers and to test its reliability and validity. Methods The inital scale was formed by literature review, semi-structured interview and expert consultation. From September to December 2015, a total of 238 medical workers, from different departments from hospitals of different levels in Quanzhou, were selected hierarchically and randomly, and were investigated about their attitude towards hospice care, by which, reliability and validity of the scale were tested, and the final scale was formed. Results In the final scale of hospice care attitude for medi-cal workers, 7 dimensions with 26 items were included. By exploratory factor analysis, 7 common factors were extracted, with cumulative contribution of variance 62.238% . Content validity of the scale was 0.889, with Cronbach's alpha 0.891, split-half reliability 0.863 and test-retest Pearson correlation coefficient 0.867. Conclusions The hospice care attitude scale for medical workers has been proved to be reliable and valid, which can be used to assess hospice care attitude of the medical workers.
6.Effects of Serial Lumbar Puncture on Severe Intraventricular Hemorrhage of Neonates

Objective Hydrocephalus is one of the most common complications of neonatal intraventricular hemorrhage (IVH). In this study, the effect of serial lumbar puncture (LP) on severe neonatal IVH is evaluated. Methods The effects of serial LP on 30 neonates hospitalized with severe IVH at Xinhua hospital since 1993 was evaluated. Another 30 neonates with severe IVH who had not been treated for preventing post-IVH hydrocephalus were used as the control group. Results Of the 30 cases which received serial LP therapy, the cerebral ventricles of 25 cases stopped enlarging and shrank distinctly. The effective rate was 83.3%. The average age when serial LP therapy initiated was (15.0±13.5) days, the average treatment course was (16.9±12.9) days. The serial LP therapy was repeated (6.4±4.7) times. The avarage interval of LP was (2.9±2.7) days and the average CSF volume removed was (6.7±1.6) ml. The average time of LP taking effect was (9.6±5.5) days. Ten cases received diamox treatment during the course of serial LP. After 1-2 weeks' combined treatment, the enlarged ventricles of 8 cases returned to normal or remained steady. Eighteen of 25 ases were followed-up for (8.3±1.9) months. Their physical and mental development were normal. Cranialcerebral ultrasound examination found that the shapes of cerebral ventricles of 14 cases were normal and the cerebral ventricles of another 4 cases enlarged slightly. The treatment with removed CSF >5 ml/time or with a shorter interval of LP (<2 d) acheived better effect (P<0.05 or 0.01). In the control group, the cerebral ventricles of 23 cases enlarged moderately or severely (6 of them complicated with hydrocephalus). Conclusions Serial LP is a safe and effective method for severe neonatal IVH in preventing post-IVH hydrocephalus.
7.Impact of Mild SARS-CoV-2 Infection on Hematological Para-meters and Suitability of Apheresis Platelets Donation in Blood Donors

Objective:To investigate the effects of mild SARS-CoV-2 infection on hematological parameters of adult blood donors and the suitability of apheresis platelet donation,the changes of the hematological parameters in blood donors with mild infection of the SARS-CoV-2 Omicron variant strain were evaluated.Methods:Seventy-two blood donors with mild COVID-19 symptoms who donated consecutive apheresis platelets for 3 times from December 2022 to January 2023,42 cases among which were included in the infection-positive group,and 30 cases in the suspected infection group.Forty-two donors un-vaccinated against SARS-CoV-2,un-infected,and donated three consecutive apheresis platelets from October to November 2022 were included in the control group.The changes of blood routine testing in the positive group and the suspected infection group were retrospectively compared before(Time1)and after(Time2 and Time3)the onset of symptoms,three consecutive times(Time1,Time2,Time3)in the control group by repeated measures analysis of variance.The Bayesian discriminant method was used to establish a discriminant equation to determine whether the recent infection of SARS-CoV-2 occurred or not.Results:Simple effect of the number times of tests in the positive and suspected infection groups was significant(Finfection-positive group=6.98,P<0.001,partial η2=0.79,Fsuspected infection group=4.31,P<0.001,partial η2=0.70).The positive group and the suspected infection group had lower RBC,HCT,and HGB,and higher PLT and PCT at Time2 compared to Time1 and Time3(P<0.05).The positive group and the suspected infection group showes RDW-CV and RDW-SD at Time3 higher than Time1 and Time2(P<0.001).The simple effect of the number times of tests in the control group was not significant(F=0.96,P=0.55,partial η2=0.34).The difference of the whole blood count parameters in the control group for three times was not statistically significant(P>0.05).We established a discriminant equation to determine whether the recent infection of SARS-CoV-2 occurred or not.The equation had an eigenvalue of 0.22,a canonical correlation of 0.43(x2=27.81,P<0.001),and an analysis accuracy of 72.9%.Conclusion:The hematological indicators of RBC,HCT,HGB,PLT,PCT,RDW-CV and RDW-SD in blood donors who had infected with mild COVID-19 showed dynamic changes.The discriminant equation for whether they are infected recently with COVID-19 has a high accuracy rate.
8. Effect of Morinda officinalis on serum metabolic spectrum in ovariectomized osteoporosis rats

Aim To study the effect of Morinda officinalis on serum metabolisms in ovariectomized osteoporosis rats based on metabonomics, and explore the mechanism of Morinda officinalis in the treatment of postmenopausal osteoporosis.Methods Forty-eight SD rats were randomly divided into sham operation group, model group and Morinda officinalis group.The Morinda officinalis group was given Morinda officinalis water extract by gavage.The model group and sham operation group were given normal saline by gavage.The bone mineral density(BMD)of the right femur was measured by dual energy X-ray absorptiometry; the maximum load of the tibia bending at three points and the lumbar compression was measured by universal material testing machine.The endogenous metabolites of ovariectomized osteoporosis rats were identified by serum metabonomics, and the potential differential metabolites were screened and identified..Results The BMD and maximum load of the model group decreased significantly, while the Morinda officinalis group increased significantly compared with the model group.The serum metabolic spectrum of the sham operation group was completely separated from that of the model group, and the Morinda officinalis group was close to the sham operation group, suggesting that the body had a tendency to return to normal after intervention of Morinda officinalis.28 metabolites and 5 metabolic pathways were identified to be related to ovariectomized osteoporosis.Morinda officinalis could regulate the contents of stearic acid, uracil and other metabolites, which were related to the biosynthesis of unsaturated fatty acids, the metabolism of pyrimidine and so on.Conclusion Morinda officinalis can prevent ovariectomized osteoporosis by regulating the lipid metabolism and nucleic acid metabolism.
9.Association between gene polymorphisms and myocardial infarction in Han Chinese of Yunnan province.

OBJECTIVETo determine frequencies of genetic polymorphisms of coagulation factor VII (FVII), coagulation factor FXII (FXII), fibrinogen (FBG) and 9p21 in ethnic Han Chinese from Yunnan province, and to assess the association between such polymorphisms and onset of myocardial infarction (MI).
METHODSOne hundred and forty-two patients with MI and 192 healthy controls were analyzed. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and pyrosequencing were used to determine the genotypes of FVII, FXII, FBG and 9p21.
RESULTSNo significant difference was found in the frequencies of R353Q, 5'F7, C46T, -148C/T, rs1333049 and rs4977574 loci between the two groups (P> 0.05). However, the frequencies of AA of -455G/A, T and TT of rs1333040, T and TT of rs10116277 and G and GG of rs2383207 were significantly higher in MI group compared with the controls (P< 0.05), whilst the frequencies of CT of rs1333040 and GT of rs10116277 were significantly lower in MI group compared with the controls (P<0.05).
CONCLUSIONPolymorphisms of FVII, FXII, -148C/T of FBG and rs1333049 of 9p21 were not associated with myocardial infarction. Polymorphisms of -455G/A of FBG and rs1333040, rs10116277 and rs2383207 of 9p21 may be associated with MI in ethnic Han Chinese from Yunnan province.

10.Quantitative studies of the production phase in rHSA fermentation.

The model equations of the production phase of rHSA fermentation were derived on the base of both elemental balance and metabolic balance, then the unknown parameters of the model were estimated by multivariable optimization. The possible reasons of discrepancy of production rate between different period of fermentation were discussed. The model could preferably described the relations between different macroscopic reaction rates of the process and keys for the high-efficiency expression of HAS were deduced.
