1.The Relationship Between Hyperferritinemia And Early Metabolism Disturbance Of Sugar And Lipid

Objective To study the relationship between hyperferritinemia and early metabolism disturbance of sugar and Lipid.Methods 382 people in health examination from Hangzhou First People's Hospital were divided into hyperferritin group(A group)and hypoferritin group(B group)according to the level of ferritin.Blood fat,fasting blood glucose(FBG),uric acid(UA),hematoglobin(Hb),red blood cell count(RBC),hematocrit(HCT),mean corpuscular-hemoglobin concentration(MCHC)were determined,and adiposis hepatica was judged by ultrasonic inspection,then we analyzed by meams of statistics.Results The result showed that TG,LDL,HDL,ApoA1,ApoB,FBG,UA,Hb,RBC,HCT,MCHC had significant deviation between two groups.Ferritin was positive correlated with TG,TC,LDL,ApoB,Hb,RBC,HCT,and negative correlated with HDL,ApoA1.The incidence of adiposis hepatica in A group was obviously higher than B group.Conclusions Probably,ferritin was one of the markers of early metabolism disturbance of sugar and Lipid.
2.The expression changes of MMP-9 and TIMP-1 in the brain of neonatal rats with hypoxic-ischemic brain damage(HIBD)

Objective To explore the mechanism of hypoxic-ischemic brain damage(HIBD) and provide new ideas for clinical treatment of hypoxic-iscbemic encephalopathy.Methods Neonatal 7-day-old Wistar rats were randomly divided into sham-operation control group,HIBD 6 h,12 h,24 h,48 h and 72 h groups(n =6 per group).The model of HIBD was induced by unilateral carotid ligation followed by timed exposure to 8% oxygen.The expression of MMP-9 mRNA and TIMP-1 mRNA in brain tissue of neonatal rats was measured by Real-time Q-PCR method.Results (1) The ligatod brainhemisphere of HIBD groups showed obvious edema from 12 h to 48 h after hypoxia -ischemia in the neonatal rats.(2) The expression of MMP-9 mRNA was very low in the sham-operation control group,but in HIBD groups,it began to increase at 6 h,and reached a peak at 24 h,then gradually decreased,but still maintained at high level at 72 h(P<0.01).(3) The expression of TIMP-1 mRNA was aslo very low in the sham-operation control group.But in HIBD group,it increased slightly at 6 h,12 h and 24 h,compared to the sham-operation control group,each group was statistically significant(P<0.05),with no significant difference among the three groups(P>0.05),then decreased at 48 h and 72 h,but with no significant difference from sham-operation control group (P> 0.05).(4) The ratio of MMP-9 mRNA/TIMP-1 mRNA was normal in the sham-operation control group and HIBD 6 h group,it began to increase at 12 h,and reached a peak at 48 h,then gradually decreased,but still maintained at high level at 72 h(P <0.01).Conclusion Hypoxia-ischemia increases the expression of MMP-9 mRNA in brain tissue of nenatal rats,and the imbalance in the expression of MMP-9 mRNA and TIMP-1 mRNA possibly is one cause of brain edema induced by HIBD.

Objective To study the expression of RASSF1A gene in gastric cancer tissues and cell lines and to investigate its clinical roles. Methods Immunohistochemistry was employed to detect the expression of RASSF1A gene in 39 cases of human gastric cancer tissues and 18 cases of normal human gastric tissues, and RT-PCR was used to examine the expression of RASSF1A mRNA in 4 gastric cancer cell lines, normal gastric cell lines GES-1 and positive control Hela cells. Correlations between expression of RASSF1A gene and clinicopathological characteristics of gastric cancer were also analyzed. Objective To investigate the correlation between serum S100B, IL-6 and intracranial pressure in patients with severe craniocerebral injury.Methods 81 cases of patients with severe brain injury in our hospital from August 2012 to April 2016 were selected,intracranial pressure was monitored immediately after admission to calculate the average daily ICP level ,and venous blood were collected after craniocerebral injury 6,12,24,48, 72 hours.Detection of serum S100B protein and IL-6 levels,and the correlation analysis with the level of intracranial pressure.Results Patients with severe craniocerebral injury S100B levels increased gradually after injury, reached the peak at 24 hours, then decreased gradually; while patients with IL-6 and intracranial pressure after injury gradually increased, the difference was statistically significant in different time points among the S100B,IL-6 and intracranial pressure levels (P<0.05).Conclusion The changes of intracranial pressure after severe craniocerebral injury were proportional to the levels of serum S100B and IL-6,S100B and IL-6 can reflect the changes of intracranial pressure,intracranial pressure changes predicted by S100B plasma concentration in 48 hours were more sensitive than those in the same concentration of IL-6.

6.Determination of methaqualone and 2-methyl-3[2-(hydroxymethyl) phenyl]-4 (3H)quinazolinone concentrations in human serum by RP-HPLC

Methaqualone and 2-methyl-3 [2- (hydroxymethyl) phenyl]- 4 (3H) quinazolinone (metabolite I) concentrations in human serum wereanalyzed quantitatively by reverse high performance liquid chromatography (RP-HPLC). The concentrations of methaqualone ranging from 1-35?g/ml were in good linear relationship(Correlation coefficient r = 0. 9980, regression line y = 0. 06324x-0. 1029), The average recovery of methaqualone reached as high as 102.08?9. 987(SD)% (n= 5). The detective limit was 1 ng. The relative quantity of metabolite I was determined according to the linear concentration of methaqulone. RP-HPLC assay is proved to be useful in determening blood methaqualone and metabolite I concentrations in forensic toxicology.
7.Determination of the fingerprint chromatogram of Hongjingtian Injection by HPLC

AIM: To establish the fingerprint chromatogram of Hongjingtian Injection ( Rhodiola kirilowii (Regel.) Maxim. METHODS: HPLC with YWG C 18 column was used, the (a)MeOH ACN(1∶1)、(b) 0.07% H 3PO 4 H 2O gradient elution as a mobile phase and detection wavelength at 278nm . RESULTS: 17 peaks were indicated on the HPLC fingerprint of Hongjingtian Injection. The relative retention time and relative peak area were obtained with itself peak. CONCLUSION: The method is simple and accurate with good reproducibility and can be used as a quality control method of Hongjingtian Injection.
9. The value of combined detection of p16INK4A, Brn3a, telomerase and other biomarkers in cervical cytology for predicting the progression of cervical neoplasia

Objective: To study the expressions of p16INK4A, Brn3a, telomerase and human cervical cancer oncogene (HCCR) in cervical intraepithelial neoplasia (CIN), and to estimate their values for predicting the progression of cervical lesions. Methods: Paraffin specimens of 262 cases were all from the residue of patients' liquid-based cytology test (LCT) samples, including 30 normal cases, 80 cases of atypical squamous cells of undetermined significance (ASCUS), 72 cases of low-grade squamous intraepithelial lesions (LSIL), 50 cases of high-grade squamous epithelial lesions (HSIL) and 30 cases of invasive squamous cell carcinoma. The expression levels of p16INK4A, Brn3a, telomerase and HCCR in these samples were detected using immunocytochemistry. Results: The expression levels of p16INK4A and Brn3a in ASCUS and LSIL specimens were significantly higher than those in normal cells (P < 0.05). Compared to ASCUS and LSIL, the expression levels of p16INK4A, Brn3a, telomerase and HCCR in HSIL and invasive squamous cell carcinoma were significantly higher (P < 0.05). Among ASCUS, both the expression of p16 INK4A in cases that had pathological diagnosis of CIN1 and the expressions of p16INK4A and Brn3a in cases that had pathological diagnosis of CIN2+ were significantly higher than those had pathological diagnosis of chronic cervicitis or cervical condyloma (P < 0.05). Among LSIL, the expressions of p16INK4A and Brn3a in cases that had pathological diagnosis of CIN2+ were significantly higher than those had pathological diagnosis of chronic cervicitis or cervical condyloma (P < 0.05). Moreover, the expressions of p16INK4A and Brn3a in ASCUS and LSIL had a strong relationship (r = 0.580, P < 0.001). Conclusion: The expression levels of p16INK4A, Brn3a, telomerase and HCCR can help distinguish HSIL and invasive squamous cell carcinoma from ASCUS and LSIL, and combined detection of p16INK4A and Brn3a maybe assist in predicting the progression of ASCUS and LSIL. 10.Clinical Observation on Treatment of Post-operative Gastrointestinal Syndrome with Combined Acupuncture and Herbs

Objective: To observe the therapeutic effect of treating postoperative gastrointestinal syndrome with combined needles and herbs. Method: Altogether 117 in-patients were randomly allocated into the treatment group of 86 cases and control group of 31 cases. Point injection was mainly adopted in the treatment group plus Chinese herbal decoction, while Western medicine was adopted in the control group. And then the therapeutic effect of the two groups was compared. Result: The total effective rate of the treatment group and control group were 96.5% and 80.6% respectively, and the treatment course of the treatment group was significantly shorter than the control group (P < 0.01). Conclusion: Combined needles and herbs have excellent effect in treating postoperative gastrointestinal syndrome and were superior to Western medicine.
