1.Retrospective analysis of quality data of different blood components in Chongqing

【Objective】 To provide reference for formulating relatively unified quality control strategies and meeting the requirements of homogenization construction of blood banks across Chongqing area by retrospectively analyzing sampling results of different blood components during the past two years in all levels of blood banks in Chongqing area. 【Methods】 The key quality data of blood components prepared by 6 blood banks in Chongqing were analyzed retrospectively. According to the issuing units to the clinical during the past two years, the research objects were selected as leukocyte-depleted suspended RBCs, cryoprecipitate, pathogen inactivated fresh frozen plasma(FFP) and apheresis platelets. The quality data of the above-mentioned blood components from January 2019 to June 2021 were collected and analyzed. 【Results】 For leukocyte-depleted suspended RBCs(1U)prepared by 5 blood banks, statistically significant differences in Hb, residual white blood cells and hemolysis rate at the end of storage, except for Hct, were noticed(P<0.05). For cryoprecipitate, the content of blood coagulation factor Ⅷ and fibrinogen were statistically different among 3 blood banks in 1U specification(P<0.05) and among 5 blood banks in 2U specification(P<0.05). For pathogen inactivated FFP, the content of blood coagulation factor Ⅷ, plasma proteins, and residual methylene blue were statistically different among 3 blood banks(P<0.05). For apheresis platelets, Plt, white/red blood cells contamination and pH at the end of storage were statistically different among 3 blood banks(P<0.05). 【Conclusion】 The quality data of blood components, prepared by different blood banks, meet the requirements of national standard, however, certain differences are existing among blood banks.Key points during the process of collection, preparation, storage and transportation need to be cleared and unified, so as to reduce the differences between each other, and determine the direction and basis for homogeneity construction in the next step.
2. Applications and challenges of paper-based microfluidic in laboratory medicine

Paper-based microfluidic, as a rapid, low-cost and portable detection technology, has been widely used in many fields such as disease diagnosis, food safety and environmental monitoring, especially in laboratory medicine. Compared with conventional methods, paper-based microfluidic provides reliable support for rapid and immediate diagnosis of diseases due to its unique physical and chemical properties of test strips. Based on the construction scheme and result analysis strategy of paper-based microfluidic, this paper reviews its application in clinical detection of nucleic acid and protein, and looks forward to its potential opportunities and challenges in laboratory medicine.
3.Status quo of and challenges for research on rust disease in medicinal plants.

Medicinal plants are beneficial to human health. However,most of the major producing regions of medicinal plants suffer from rust disease,which threatens the yield and quality of Chinese medicinal materials,thus causes huge economic loss,and hinders the sustainable development of the Chinese medicine industry. By the end of 2020,rust disease had been reported in medicinal plants of 76 species and 33 families. In the 76 species,79 rust pathogens were detected. The majority of these pathogens belonged to Puccinia( 33,39. 24%),Coleosporium( 14,15. 19%),and Aecidium( 11,13. 92%). Of these 79 rust pathogens,10 were autoecious and 13 were heteroecious. Through literature research,this study reviewed the symptoms,pathogen species,severity and distribution,prevalence and occurrence conditions,and control measures of rust disease in medicinal plants,and thereby summarized the research status of rust disease in medicinal plants and the gap with other plants,which is expected to serve as a reference for further research on rust disease in medicinal plants.

4.Evaluation of Percutaneous Internal Fixation with Hollow Screw in the Treatment of Femoral Neck Fracture

Objective To evaluate the efficacy of percutaneous internal fixation with hollow screw for femoral neck fractures.Methods From June 2001 to June 2004,48 cases of femoral neck fractures,including 22 cases of fracture without displacement(type Garden I and II) and 26 cases of fracture displacement(type Garden III and IV),were treated by close reduction and percutaneous internal fixation with 3 hollow screws forming inverse triangle.Results The intraoperative blood loss was less than 10 ml,and the average operation time was 32.5 min(range,18-54 min).48 cases were followed up for 24-61 months(mean,38.2 months),union of fracture found in 97.9%(47/48) of cases,union time being 4-12 months(mean,6.2 months).According to Harris score of hip joint function,32 cases were excellent,11 cases good,3 cases fair,2 cases bad,the rate of excellent and good results being 89.6%(43/48).In 22 cases of fracture without displacement,there was no nonunion and ischemic necrosis of head of femur.In 26 cases of fracture displacement,the rate of nonunion and ischemic necrosis of head of femur were 3.8%(1/26) and 11.5%(3/26) respectively.Conclusions Percutaneous internal fixation with hollow screw has the advantages of ideal curative effects,simple procedure,reliable fixation and less complications.
5.Effects of internet plus continuous rehabilitation nursing on older patients after lumbar fusion

ObjectiveTo observe the effects of Internet plus continuous rehabilitation nursing on older patients after lumbar fusion. MethodsFrom January, 2018 to August, 2021, 157 older patients after transforaminal lumbar interbody fusion (TLIF) in Department of Orthopedics, Chongqing General Hospital were retrospectively studied. A total of 66 patients accepting routine continuous rehabilitation nursing care were as control group, and 91 patients accepting internet plus continuous rehabilitation nursing care were as intervention group. They were assessed with Visual Analog Scale for pain (VAS), Oswestry Disability Index (ODI), Huaxi Emotional-distress Index (HEI) and Adult Health Self-management Skill Rating Scale (AHSMSRS) before, and one and three months after intervention. The compliance and nursing satisfaction were investigated with self-made questionnaire three months after intervention, and the postoperative complications were recorded. ResultsA total of 148 patients finished follow-up of three months, including 61 patients for the control group and 87 patients for the intervention group. The VAS, ODI, HEI and AHSMSRS scores improved in both groups one and three months after intervention (F > 85.4, P < 0.001), and improved more in the intervention group than in the control group (|t| > 3.645, P < 0.001). Both the compliance and nursing satisfaction were better in the intervention group than in the control group (χ2 > 5.478, P < 0.05), and no postoperative complication was observed in both groups. ConclusionInternet plus continuous rehabilitation nursing can promote the recovery of function, pain and psychological emotion, ability of self-management, compliance, and nursing satisfaction.
6.Analysis on the current monitoring and management of blood donation adverse reaction in Chongqing

【Objective】 To investigate the management of adverse reactions to blood donation(ARBD) in blood services, so as to promote the surveillance of ARBD and improve the quality of blood donation service in Chongqing. 【Methods】 A questionnaire, involving the staff and facilities in blood donation sites as well as the prevention and treatment, the record and report, the following up and data related to ARBD was developed by Chongqing Society of Blood Transfusion in February 2019, and was issued to 18 blood services(1 blood center and its sub-center, 6 central blood stations and 11 hospital blood banks) in the Chongqing via email. The questionnaire was filled in and submitted before March 31 by management personnel participating in the investigation, and the data was collected, collated, revised and analyzed by Excel 2011. 【Results】 A total 19 questionnaires were collected, with the valid rate at 100%(19/19). 78.95%(15/19) of the blood services met the requirements of medical personnel allocation(>6 medical staff) when the number of daily blood collection was more than 60, and 100%(19/19)met the requirements of medical personnel allocation(2 to 6 medical staff) when the number of daily blood collection was less than 60. 89.47%(17/19) of the blood services were equipped with epinephrine hydrochloride, and 84.21%(16/19) with dexamethasone(an anti-allergic drug). There were significant differences in the allocation of other types of drugs. 100.00%(19/19) of the blood services formulated prevention and treatment measures concerning ARBD. In 2019, the incidence of ARBD in Chongqing was reported to be 0.54%(1 958 / 359 871), with the highestas [1.35%(223/16 543)] in subcenters and the lowest [0.32%(179/56 299)] in central blood centers (P<0.05). There was statistical significances in the incidences of ARBD reported by different blood stations(P<0.05). 【Conclusion】 The monitoring and management of ARBD among blood services in Chongqing should be further standardized in terms of staffing allocation, emergency drugs allocation and reporting, so as to gradually realize regional homogenization and ensure blood safety.
7.Aerobic exercise improves renal fibrosis in spontaneously hypertensive rats.

Objective: To study the effects of aerobic exercise training on renal fibrosis in spontaneously hypertensive rats (SHR), and to explore the protective effect of exercise on renal damage in hypertensive rats. Methods: Eight-week-old male SHR and Wistar Kyoto rats of the same age (WKY) were randomly divided into 4 groups (n=6): sedentary WKY control group (WKY-S), sedentary SHR control group (SHR-S), low-intensity exercise group (SHR-L) and medium-intensity exercise group (SHR-M). SHR-L group and SHR-M group were set at a slope of 0° at 14 m/min (35% of the maximum aerobic speed) and 20 m/min (50% of the maximum aerobic speed), running on a sports treadmill for 14 weeks, 5 times a week, and 60 min each time. WKY-S and SHR-S groups were kept quietly. Blood pressure was measured 72 hours after exercise training. And the serum levels of creatinine (Scr) and BUN were detected. The morphology of renal tissue was observed by hematoxylin and eosin (HE) staining. The collagen deposition of renal tissue was observed by Masson staining, and the renal collagen volume fraction (CVF) was calculated. Results: Compared with WKY-S group, blood pressure, serum Scr and BUN, kidney CVF levels and AngⅡ, AT1R, TGF-β, α-SMA, CTGF expressions in SHR-S group were increased significantly (P<0.05). Compared with SHR-S group, blood pressure, serum Scr and BUN, kidney CVF level and AngⅡ, AT1R, TGF-β, α-SMA, CTGF expressions in SHR-L and SHR-M groups were decreased significantly (P<0.05) and the decreasing trend was more obvious in SHR-M group (P<0.05). Conclusion: Aerobic exercise can improve renal fibrosis and renal function in spontaneously hypertensive rats by inhibiting the AngⅡ-AT1R-TGF-β pathway.

8.Epidemic characteristics and prediction of varicella in Chongqing in 2014-2020

Objective To analyze the epidemic characteristics of varicella in Chongqing from 2014 to 2020, and to provide evidence for the development of scientific and effective varicella control strategies. Methods Data on the outbreak of varicella and vaccination in Chongqing from 2014 to 2020 were collected through the China Disease Prevention and Control Information System, and descriptive epidemiological methods were used for statistical analysis. Results A total of 181 551 cases of varicella were reported in Chongqing from 2014 to 2020, with an average annual incidence rate of 83.79 per 100 000. The incidence rate of varicella increased from 39.95 per 100 000 in 2014 to 81.88 per 100 000 in 2020 (P < 0.001). The incidence of varicella was seasonal, with the peak periods occurring from May to June and from October to December each year. The average annual incidence rate in municipal districts was 88.90/100 000, higher than 67.42/100 000 in counties and 82.50/100 000 in autonomous counties. The average annual incidence rate of varicella in males (87.13/100 000) was higher than that in females (80.38/100 000). The incidence of varicella was mainly distributed in people under 15 years old, with 143 508 cases (79.10%) reported, and the highest incidence age was 5-9 years old (37.00%). Among the affected occupations , 133 733 cases (62.6%) were students , 39 274 cases (18.40%) were children in nursery care, and 17 963 cases (8.4%) were scattered children. The actual number of doses of varicella vaccine from 2014 to 2020 was 2 302 522 doses, with the coverage rates of one-dose and two-dose vaccines being 75.56% and 32.17%, respectively. ARIMA predicted that there would be 2 604, 811, 756, 1 226, 2 405, 3 904, 2 410, 1 211, 2 034, 6 878, 10 887, and 8 955 cases of varicella from January to December 2021. Conclusion The incidence of varicella in Chongqing is on the rise, with obvious seasonal, regional and population distribution characteristics. It is necessary to strengthen the prevention and control of varicella epidemic, strengthen the prevention and control measures of key groups and key institutions in the high incidence season, strengthen the publicity of varicella vaccine, and improve the vaccination rate of two-doses of varicella vaccine for eligible children.
9.Application of binomial distribution-based statistical process control method in blood quality control

【Objective】 This study endeavors to introduce the statistical process control (SPC) method to analyze the quality control index concerning red blood cells in additive solution with leukocytes reduced, with the aspiration to advance the effective utilization of blood quality control data, thereby providing empirical foundations for the continual enhancement of blood quality. 【Methods】 Between 2020 and 2022, test data pertaining to the quality control index of red blood cells in additive solution with leukocytes reduced were amassed from six blood stations in Chongqing area. Utilizing Minitab software, the SPC analysis was carried out, p-control charts were delineated, the non-conformance rates of each quality control index along with their 95% confidence intervals were computed, as well as the Process Capability Index (Z value). 【Results】 In accordance with the Whole Blood and Blood Components Quality Requirements, the appraisal of the quality control indexes for red blood cells in additive solution with leukocytes reduced manifested a conformity rate of 100% for appearance, end-of-storage hemolysis rate and sterility test. Nonetheless, the conformity rates for volume, hemoglobin, hematocrit and residual leukocytes did not attain 100%, albeit all were ≥75%. Through the employment of binomial distribution-based p-control charts, the controlled state of the production process was discerned. Although the overarching conformity rate satisfied the national standard stipulations, it was discerned that there were out-of-control points concerning volume, hemoglobin, hematocrit, and residual leukocytes across different institutions, exhibiting palpable trends. The non-conformance rates of all quality control indexes were less than 25%, yet at a 95% confidence level, the residual leukocyte counts from institutions B, C, E, and F did not adhere to the stipulations (exceeding 25%). By architecting the ability evaluation index Z value for count data process capability analysis, it was unveiled that the volume of institution E, the hematocrit of institutions B, C, and F, and the residual leukocytes Z values of all six blood collection and supply institutions were below 2, hinting at avenues for amelioration. 【Conclusion】 The SPC method anchored in binomial distribution exhibits substantial application merit in blood component quality management, facilitating real-time surveillance of blood collection, preparation, and storage procedures.
10.Regional hemovigilance system for adverse reactions to blood donation

【Objective】 To establish a regional hemovigilance system of adverse reactions to blood donation(ARBD) and to evaluate its operation effect. 【Methods】 A regional monitoring working group was established to improve the ARBD monitoring system in two phases [phase-ⅠJanuary-June, 2019 and phase-Ⅱ July-December, 2019). The suitability of monitoring standards and reporting approaches, as well as the running effect of this system were evaluated by comparing the accuracy of ARBD classification and severity grading, timely report processing and the contents of reports submitted in two phases. The incidence of ARBD and the number of different types of ARBD cases in the two phases were analyzed to compare the monitoring effect. 【Results】 7 and 17 blood banks joined the regional working groups in phase-Ⅰand phase-Ⅱ, respectively. In phase-Ⅰ, 565 cases of ARBD were reported, 72.04%(407/565)of them was accurately sorted and 80.95%(34/42)were timely reported. Over 99% of reports covered such monitoring items as blood donors′ gender, type of blood donation, place of blood donation, organization mode of blood donation, symptom of ARBD, type, severity grading and other related factors, and less than 20% of reports recorded the details concerning the treatment following ARBD. In phase-Ⅱ, 1098 cases of ARBD were reported, 94.54%(1 038/1 098)of them was accurately sorted and 96.08%(98/102)were timely reported. Over 97% of reports covered the monitoring items mentioned above, still less than 30%, though elevated in comparison with phase-Ⅰ, of reports recorded the details concerning the treatment following ARBD, and less than 20% tracked the ARBD. The overall incidence of ARBD reported was 0.87%, while 0.93%(1 098/118 565)in phase-Ⅱ and 0.77%(565/73 267)in phase-Ⅰ, with statistically significant differences (P<0.01). 【Conclusion】 The regional monitoring system for ARBD in Chongqing, with Blood Safety Hemovigilance Guidelines(T/CSBT 001-2019)as the standard and the Blood Safety Monitoring Management Information System as the reporting approach, has been basically established through two phases of development, and the reporting contents need to be further managed at specific levels.
