1.Effect of apolipoprotein E gene polymorphism on early apoptosis of astrocytes after hypoxic injury

Objective To investigate effect of polymorphism of apolipoprotein E (APOE) gene on early apoptosis of astrocytes after hypoxic injury.Methods Astrocytes separated from APOE wild mice and APOE transgenic mice (ε3,ε4) were primarily cultured,and then purified and identified.Models of astrocyte hypoxic injury were set up by hypoxia.Morphological changes of astrocytes and mitochondria were observed by transmission electron microscope.Early apoptosis rate and changes of mitochondrial membrane potential were detected by flow cytometry.Results Cell foot process tumidness and mitochondria with irregular outline,vacuoles and irregular cristae were observed in each group by electron microscopy at six hours after hypoxia.There were no significant differences of cellular form changes among groups.Early apoptosis rate and decreasing degree of mitochondrial membrane potential in APOFε4 group were significantly higher than those in APOEε3 group and APOE wild group (P < 0.05).Conclusion Compared with astrocytes from APOEε3 group and APOE wild group,mitochondrial membrane potential in astrocytes from APOEε4 group at early period after hypoxia declines more significantly,as may be one of causes for more astrocyte apoptosis.
2.MR molecular imaging of pancreatic cancer cell targeted by superparamagnetic iron oxide nanoparticles

Objective To explore the feasibility of MR molecular imaging in human pancreatic cancer cell (BxPC-3 cell) targeted by superparamagnetic iron oxide nanoparticles (SPION).Methods Both MUC1 SPION probes with MUC1 targeted modification (targeted group) and bull serum albumin (BSA)-SPION as the control (non targeted group) were prepared.The cytotoxicity of MUC1 SPION in different concentrations (0,6.25,12.50,25,50,100,200 μg/ml) was verified by MTT (3 [4,5-Dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay.BxPC-3 cells were cultured with MUC1-SPION (targeting group) and BSA-SPION (control group) in different concentration as 50,100,200 μg/ml,respectively for 2 h.Then MRI scans were performed,the transverse relaxation time (T2) value and the enhancement ratio of T2 were recorded and calculated.The combination conditions of targeting probes and cells were observed by prussian blue staining.Results The cell cytotoxicity of MUC1 SPION in different concentrations showed no statistical difference according to MTT assay (F=1.74,P 0.18).There were statistical differences of the T2 value and the enhancement ratio of T2 for the concentration as 50,100,200 μg/ml,respectively (all P<0.05).More blue particles were observed by prussian blue staining in targeted group than in non targeted group.Conclusion MUC1-SPION has favourable targeting ability to BxPC 3 cell,and MRI of BxPC-3 cell targeted by SPION is satisfied security and feasibility.
3.Emergent reconstruction and repair of thumb and finger defect by toes and combined toes transplantation

Objective To explore the method and effect of thumb and finger reconstruction and repair by emergent primary toes transplantation. Methods Three hundred and fourteen fingers of 282 cases including finger thumb defect and complex injury with hyperthena palm radid soft tissue of forearm were reconstructed and repaired by the second toes, wrap-around flap and the second third toes wrap-around flap combined with distal flap of leg or other complex tissue flap transplantation. Results Three hundred and eight reconstructed fingers survived, the survival rate was 98%. After a follow up to 6 months to 6 years, the reconstructed thumbs and fingers have accepted nip, grasp, opponent function. The result was graded as excellent in 214 fingers, good in 63, fair in 26, poor in 5. The excellent and good rate was 88%according to the upper limb functional evaluation criteria issued by the Hand Surgery Society of the Chinese Medical Association. Conclusion Different type of toes and combined toes transplantation can obtain preferable clinical effect on emergent primary thumb and finger defect.
4.Effect of manual therapy based on surface electromyography on knee osteoarthritis for older people:a random-ized controlled trial

Objective To investigate the effect of manual therapy based on surface electromyography on knee osteoarthritis(KOA)in the older people. Methods A total of 106 outpatient with unilateral KOA were selected from Shuguang Hospital Affiliated to Shanghai University of Traditional Chinese Medicine,from August,2023 to June,2024,and were randomly divided into control group(n=53)and experimental group(n=53).The control group accepted routine manual therapy,and the experimental group accepted manual therapy based on the analysis of average electromyography(AEMG).They were assessed with Western Ontario and McMaster Universities Osteoarthritis Index(WOMAC),Visual An-alogue Scale(VAS)for pain,Tinetti Balance and Gait Score,and 6-minute walk test(6-MWT)distances before and after treatment. Results One case dropped down in each group.Before treatment,AEMG decreased in the rectus femoris,medial femoris and medial head of gastrocnemius on the affected side in the experimental group(|Z|>8.647,P<0.001),and it increased in the lateral femoris,semitendinosus and biceps femoris(|Z|>4.808,P<0.001).The scores of WOMAC,VAS,Tinetti Balance and Gait Score,and distances of 6-MWT improved in both groups after treat-ment(|t|>3.987,P<0.001),and improved more in the experimental group than in the control group,except the VAS score(|t|>2.213,P<0.05). Conclusion Manual therapy focusing on activation of rectus femoris,medial femoris and medial head of gastrocnemius,inhibition of the lateral femoris,semitendinosus and biceps femoris,and releasing the tension of the medial and lateral collateral ligaments,according to the results of surface electromyography,can alleviate the pain of the KOA in the older people and improve the mobility of the knee.
