1.Combined effects of fluoride and arsenite on the expression of Runx-related transcription 2 mRNA in bone of rats

Objective To explore the combined effects of fluoride and arsenite on the expression of Runx-related transcription 2 (Runx2) mRNA in bone of Sprague Dawley (SD) rats.Methods Fifty four SD rats were selected[body mass(109.71 ± 10.52)g,half male and half female].3 × 3 Factorial experimental design was used to evaluate the combined effects of fluoride and arsenite on the expression of Runx2 mRNA by random number talbe.Rats were exposed to NaF,NaAsO2 and NaF plus NaAsO2 for 6 months by oral perfusion at gradient doses,respectively:the control group(0 mg/kg NaF + 0.0 mg/kg NaAsO2),the low fluoride group(5 mg/kg NaF),the high fluoride group(20 mg/kg NaF),the low arsenite group(2.5 mg/kg NaAsO2),the high arsenite group(10.0 mg/kg NaAsO2),the low fluoride low arsenite group(5 mg/kg NaF + 2.5 mg/kg NaAsO2),the high fluoride low arsenite group(20 mg/kg NaF + 25 mg/kg NaAsO2),the low fluoride high arsenite group(5 mg/kg NaF + 10.0 mg/kg NaAsO2) and the high fluoride high arsenite group(20 mg/kg NaF + 10.0 mg/kg NaAsO2).The expression of Runx2 mRNA was determined by quantitative real-time RT-PCR.Results The expressions of Runx2 mRNA in the control,low fluoride,high fluoride,low arsenite,high arsenite,low fluoride low arsenite,low fluoride high arsenite,high fluoride low arsenite and high fluoride high arsenite groups were 1.024 ± 0.015,1.377 + 0.014,1.587 ± 0.012,1.182 ± 0.015,1.343 ± 0.010,1.444 ± 0.019,1.504 ± 0.013,1.608 ± 0.013 and 1.714 + 0.009,respectively.The expressions of Runx2 mRNA in experimental groups were higher than those in control group (all P < 0.05),fluoride and arsenite were positively correlated with the expression of Runx2 mRNA(all P < 0.01),and there was a dose-response relationship between Runx2 mRNA and fluoride-arsenite levels.Factorial analysis showed that fluorine or arsenic alone could affect the expression level of Runx2(F =46.967,8.317,all P < 0.05),and there was a interaction between fluorine and arsenic to the expression of Runx2 mRNA (F =105.271,P < 0.01).Conclusion Fluoride or arsenic could promote the expression of Runx2 mRNA in bone of rats; there is an interaction between fluorine and arsenic to the expression of Runx2 mRNA.
2.Status quo and research of medical students′professional perception and humanistic education

Objective To better understand students' professional perception,humanistic education status quo and their preference to different humanistic education methods.Methods Questionnaire was conducted for 990 undergraduate students (2006 grade- 2010 grade) at medical school of Shanghai Jiao Tong university.SPSS 17.0 statistical software was used to analyze results of the questionnaire.Results Nearly 45% students chose to major in clinical medicine out of personal interest.The identification of students for humanistic quality was lower than that for clinical knowledge and skills.More than 50% students believed doctors were with high humanistic quality and there was a gap between students′ action and their understanding of the medical career identity and medical humanistic cultivation.More than 50% students preferred activities that train medical and auxiliary skills.Nearly 67% students also showed a shallow reading habit from regular internet surfing.Conclusion The professional perception of medical students are influenced by both social and historical factors influence.The gap between understanding and action is a consequence of current humanistic educational system.The humanistic educational methodology should conform to the psychological and s~ial characteristics of contemporary students.
3.Changes of choroidal thickness after ranibizumab treatment for non-proliferative diabetic retinopathy with macular edema

Objective To observe the changes of choroidal thickness after ranibizumab treatment for non-proliferative diabetic retinopathy (NPDR) with macular edema,and determine the association between choroidal thickness and visual acuity.Methods Twenty-three eyes from 23 patients diagnosed with NPDR and diabetic macuiar edema were treated with 3 monthly intravitreal injections of ranibizumab.The subfoveal choroidal thickness and central macular thickness were measured,and the best corrected visual acuity was recorded.Changes of subfoveal choroidal thickness,correlation between subfoveal choroidal thickness and best corrected visual acuity were assessed at 3 months follow-up.Results After 3 monthly anti-VEGF treatments,subfoveal choroidal thickness and central macular thickness decreased significantly,there was no statistical difference at 1 month and 2 months compared with pre-treatment (all P > 0.05),but there was statistical differences at 3 months compared with pre-treatment (P =0.04,0.01).In the treatment,the best corrected visual acuity increased gradually,there was no statistical difference at 1 month and 2 months compared with pre-treatment (all P > 0.05),but there was statistical differences at 3 months compared with pre-treatment (P =0.04).Before the treatment,the subfoveal choroidal thickness was positive correlated with macuiar retinal thickness (R2 =0.94,P =0.00);And the best corrected visual acuity before treatment and 3 months after treatment had a positive correlation (R2 =0.93,P =0.00;R2 =0.82,P =0.00).There was a positive correlation between central macular thickness and best corrected visual acuity at 3 months after treatment (R2 =0.83,P =0.00).There was a positive correlation in the best corrected visual acuity between before treatment and 3 months after treatment (R2 =0.84,P =0.00).Conclusion The subfoveal choroidal thickness is a clinical index to evaluate the efficacy of anti-VEGF in the treatment of diabetic macuiar edema.Baseline subfoveal choroidal thickness can be considered as one of the indicators of clinical efficacy in the short term.

This research explored the synergistic effects and the potential mechanisms of RCE-4 and various nonsteroidal anti-inflammatory drugs (NSAIDs) on the proliferation of cervical cancer Ca Ski cells. The MTT assay and CalcuSyn V2.0 software were used to detect cell proliferation and calculate the combination index (CI); the expression levels of various proteins were analyzed using Western blot assay; mitochondrial membrane potential (MMP) was assessed using JC-1 staining; acridine orange/ethidium bromide (AO/EB) double-fluorescence staining was used to detect the apoptosis of Ca Ski cells; a co-immunoprecipitation (Co-IP) assay was used to analyze the relative content of Bcl-2-Beclin 1 complex in Ca Ski cells. The results demonstrate that the combination of RCE-4 and NSAIDs increases the inhibition of Ca Ski cells compared to the single-RCE-4 group, and celecoxib provided the best synergistic effect among the four NSAIDs tested, with a CI of 0.32. 7.An ex vivo study on the vaporization ratio of the prostatic tissue lased by the 2 micron laser.

OBJECTIVESTo observe the vaporesection efficiency of the 2 micron laser to the prostatic gland in benign prostatic hyperplasia, and investigate the method of estimating the amount of the vaporesected prostatic tissues during transurethral vaporesection of the prostate using the 2 micron laser system in the treatment of benign prostatic hyperplasia.
METHODSTotal 9 fresh prostatic gland specimens were obtained from patients with BPH under open surgical procedures, and vaporesected under a simulated transurethral environment with the 2 micron laser system immediately after weighted. Energies and time consumptions were noted, collections of vaporesected tissue specimens and the remnants of the prostatic glands were weighted after the procedures. The ratios of the vaporized tissues and the collected tissues to the whole vaporesected tissues were calculated respectively. The vaporesection efficiency of the 2 micron laser to the prostatic tissues was also calculated.
RESULTSAmong the total lost tissues, about (65.6 +/- 1.5) percent of which were that of vaporized, and nearly (34.5 +/- 1.5) percent were resected. Linear correlation between the weight of collected prostatic tissue(x) and the weight of prostatic gland specimens(y) could be defined as a formula of [y = 3.245x - 6.475 (t = 15.097, P = 0.000)].
CONCLUSIONThe amounts of the whole prostatic tissues removed by the 2 micron laser could be calculated from the collected resected prostatic specimens under a simulated transurethral surgical procedure.

8.Experimental study on the microcosmic changes of compressed premaxillary suture with laser scanning confocal microscopy.

OBJECTIVEThe aim of this experiment is to know whether active orthopedic appliance is rationale to do presurgical treatment and seek some regularities.
METHODSLaser scanning confocal microscopy and triple fluorochrome labeling in bone were used to observe and study detailed changes occurred in compressed suture systematically.
RESULTSWidened suture and broken fibres between the adjoining bones was observed at the 3rd week postoperatively. At the 3rd week hyperplasia of organic tissue components was also seen. At the 6 and 9th week there were no apparent differences among all groups no matter whether in control group or experimental groups.
CONCLUSIONRetarding injury of compressed suture and surrounding tissue hyperplasia exists. With growth and development wounded suture will gradually assimilate to control group structurally and functionally.

9.Protective effect of hydrogen against hyperoxia-induced type II alveolar epithelial cell injury.

OBJECTIVETo investigate the protective effect of hydrogen against hyperoxia-induced oxidative stress injury in premature rat type II alveolar epithelial cells (AECs).
METHODSThe type II AECs isolated from premature rats were randomly divided into air (21% oxygen) control group, hyperoxia (95% oxygen) control group, air + hydrogen group, and hyperoxia+ hydrogen group. The cells with hydrogen treatment were cultured in the presence of rich hydrogen. After the corresponding exposure for 24 h, the cell morphology was observed microscopically. MTT assay was used to evaluated the cell proliferation ability, and JC-1 fluorescence probe was used to detect the mitochondrial membrane potential (δφ) changes of the type II AECs. The concentration of maleic dialdehyde (MDA) and superoxide dismutase (SOD) activity in the cell supernatant were detected using colorimetric method.
RESULTSNo significant differences were found in cell growth or measurements between air control and air + hydrogen groups. Compared with air control group, the cells exposed to hyperoxia showed significantly suppressed proliferation, reduced mitochondrial membrane potential, increased MDA content, and decreased SOD activity. Intervention with hydrogen resulted in significantly increased cell proliferation and SOD activity and lowered MDA content, and restored the mitochondrial membrane potential in the cells with hyperoxia exposure (P<0.05).
CONCLUSIONHydrogen can significantly reduce hyperoxia-induced oxidative stress injury in premature rat type II AECs, improve the cellular antioxidant capacity, stabilize the mitochondrial membrane potential, and reduce the inhibitory effect of hyperoxia on cell proliferation.

10.Study on Quality Standard of Yiwei Xiaoyu Granules

Objective To establish the quality standard for Yiwei Xiaoyu Granules.Methods TLC was used for the qualitative analyses of Atractylodis Macrocephalae Rhizoma, Angelicae Sinensis Radix and Dendrobii Caulis; HPLC was applied to determine the contents of notoginsenoside R1, ginsenoside Rg1, ginsenoside Rb1and ginsenoside Re. Results The TLC spots were clear and free from negative interference. Notoginsenoside R1, ginsenoside Rg1, ginsenoside Rb1and ginsenoside Re showed good linear relationships within the ranges of 0.067 2–0.672 μg (r=0.999 6), 0.501 2–5.012 μg (r=0.999 6), 0.326 5–3.265 μg (r=0.999 6), 0.098 3–0.983 μg (r=0.999 7), whose average recoveries were 96.32% (RSD=1.3%), 96.45% (RSD=1.5%), 101.23% (RSD=1.7%), and 97.89% (RSD=1.7%), respectively. Conclusion This method is accurate, reliable, and reproducible, which can be used for the quality control of Yiwei Xiaoyu Granules.
