1.Zymography identification of the matrix metalloproteinases synthesized by cultured hepatic stellate cells.

AIMTo establish a method for identification of the activities of matrix metalloproteinases (MMPs) synthesized by cultured hepatic stellate cells (HSC).
METHODSSDS-gelatin polyacrylamide gels electrophoresis was used for the identification of MMPs synthesized by cultured HSC.
RESULTSMMP-2 and MMP-9 in culture medium and HSC were sensitively identified, and MMP-2 activity was higher than MMP-9 in culture medium, the changes of MMPs in culture medium and HSC were also observed after 24h medicine treatment.
CONCLUSIONZymography is suitable for the studies on the role of HSC in the regulation of extracellular matrix turnover.

3.Expression and significance of ADAMDEC1 protein in human glioma

Objective To investigate the expression and significance of ADAMDEC1 in glioma.Methods We prospectively observed the expression of ADAMDEC1 in 77 glioma patients,of which 28 cases were grade Ⅳ glioma,26 were grade Ⅲ glioma (high-grade glioma);of which 23 cases were Grade Ⅱ glioma,5 were grade Ⅰ glioma (low-grade glioma).For 10 cases in the control group,brain tissue samples were collected from internal decompression patients with heavy traumatic brain injury.The expression of ADAMDEC1 in glioma tissue was detected immunohistochemistry,RT-PCR,and Western blot.With follow-up survey we explored the relationship between the survival time and the expression of ADAMDEC1 in the patients with high-grade gliomas and low-grade gliomas.Results Compared with the low-grade glioma group and the control group,qPCR,immunohistochemistry and Western blotting all showed that the protein and mRNA levels of ADAMDEC1 in high-grade glioma group were significantly increased,while ADAMDEC1 was expressed little in low-grade glioma group and not expressed in control group (P < 0.05).The survey results showed that the survival time of high-grade glioma patients was significantly shorter than low-grade glioma patients;however,the expression level of ADAMDEC1 in the highgrade glioma tissue was significantly higher than in low-grade glioma tissue (x2=5.031,P<0.05).Conclusion The expression of ADAMDEC1 is closely related to the malignant degree of glioma cells and the prognosis of glioma patients.Therefore,ADAMDEC1 can be used as an index to evaluate the malignance degree of glioma and the prognosis of glioma.
4.Risk factors for prolonged mechanical ventilation after adult double valve replacement surgery

Objective During last decades,mechanical ventilation has been an important support in the postoperative management of patients undergoing cardiac surgery.However,a considerable number of patients need mechanical ventilation for a prolonged period after cardiac surgery,and this is associated with increased mortality and morbidity.The study was designed to determine the pre-and perioperative predictors of prolonged mechanical ventilation (PMV) in adult patients undergoing double valve surgery.Methods The retrospective study considered of 2026 adult patients who underwent double valve replacement surgery at Changhai Heart Center from January 1990 to December 2010.PMV is considered as mechanical ventilation period of > 48 hours at postoperative hospital stay here.Results PMV occurred in 11.35 % of patients.The total hospital mortality was 4%.The hospital mortality of patients who had undergone double valve replacement surgery required PMV was significantly higher than control (33.5% vs 0.2%,P < 0.01).And so as comorbidities.Age more than 60 years (OR =1.943),diabetes(OR =1.757),preoperative active endocarditis (OR =3.167),New York Heart Association class higher than 3 (OR =2.464),preoperative critical state(OR =2.556),ejection fraction less than 0.50 (OR =2.026),creatinine greater than 110 μmol/L (OR =2.740),cardiopulmonary bypass time longer than 180 min (OR =3.529) and perioperative intra-aortic balloon pump(OR =18.350) were independent predictors of PMV in our patients.Conclusion PMV is associated with significant comorbidities and increased hospital mortality.Strategies to delineate the patients at risk and to modify these risk factors by prophylactic measures should probably lead to a lower incidence of prolonged mechanical ventilation for adult patients undergoing double valve replacement surgery.
5.Cloning, Sequencing of Suaeda heteroptera kitag CMO cDNA and Construction of its Recombinant Plant Expression Vector

Total RNA was extracted from leaf of Suaeda hetroptera kitag, then the CMO ( choline monooxygenase) cDNA was amplified using the reverse transcriptase polymerase chain reaction ( RT-PCR) method and cloned into pMD-T-simple vector. The positive clones from the Blue/White Screen were sequenced. After confirming its validity, the CMO gene fragment was cloned into pBI121 vector. Double enzyme restriction and PCR analysis indicated that the pBI121/CMO recombinant plasmid was successfully constructed.
6.Effects of genistein on contractility of isolated right ventricular muscles in guinea pig.

AIMTo study the effect of genistein (GEN) on contractility of isolated right ventricular muscles in guinea pig and its mechanisms.
METHODSIsolated guinea pig ventricular muscles were suspended in organ baths containing K-H solution.After an equilibration period, the effect of GEN on contraction of myocardium was observed.
RESULTSGEN and isoprenaline hydrochloride had the positive inotropic effects on contractity of myocardium. Meanwhile, the effect of GEN (1-100 micromol x L(-1)) was in dose-dependent manner. Propranolol (1 micromol x L(-1)) and verapamil hydrochloride (0.5 micromol x L(-1)) attenuated the positive inotropic effect of isoprenaline hydrochloride (1 micromol x L(-1)), but did not change the effect of GEN (50 micromol x L(-1)). Further more, the enhancement of the contraction induced by elevation of extracellular Ca2+ concentration in ventricular muscles had no change after pretreatment with GEN (1.10 micromol x L(-1)). In addition,the positive inotropic effect of GEN was inhibited partially by tamoxifen (1 micromol x L(-1)) and SQ22536 (1 micromol x L(-1)), also, could be attenuated by bpV (1 micromol x L(-1)).
CONCLUSIONGEN has the positive inotropic effect on guinea pig ventricular muscles, which is not related to the activation of beta adrenoceptor, Ca2+ channel on cell membrane,but may involve in cAMP of intracellular signal transduction and tyrosine kinase pathway.

7.A control study on the clinical outcome of tension-free vaginal tape-obturator and modified tension-free vaginal tape-obturator for female stress urinary incontinence

ABSTRACT:Objective To evaluate the curative effect of tension-free vaginal tape-obturator (TVT-O)and modified TVT-O in treating female stress urinary incontinence (SUI).Methods We selected 1 1 3 female patients diagnosed with SUI from January to December 2013 in our department and divided them into standard TVT-O group (group A,5 6 cases)and modified TVT-O group (group B,5 7 cases).We evaluated prospectively the safety,short-term efficacy and complications of operation in the two groups.Results The length from bilateral obturator membrane to the puncture point in the skin was greater in group A than in group B.The average intraoperative blood loss was more in group A than in group B.The median NRS score of postoperatie thigh pain in group A was higher than in group B (P<0 .0 5 ).The two groups did not differ significantly in operation duration,postoperative catheterization,mean hospital stay duration,the length and position from bilateral obturator membrane to the puncture point,or surgical efficacy (P>0.05).Conclusion Both TVT-O and modified TVT-O techniques are effective in treating female SUI.Modified TVT-O has a short path to get through the adductor muscles and less intraoperative blood loss;therefore,it can significantly reduce the complications such as postoperative pain to enhance the quality of life after operation. 
8.Effects of benzo(a)pyrene exposure on oxidative stress and ATPase in the hippocampus of rats.

OBJECTIVETo investigate the effects of benzo[a]pyrene (B[a]P) exposure on the behaviors and hippocampal oxidative stress and ATPase in rats and the molecular mechanism of neurobehavioral toxicity of B[a]P.
METHODSA total of 120 male SD rats (21 days old) were randomly and equally assigned to five groups: blank control group, vegetable oil (solvent control) group, and 2.5, 5, and 10 mg/kg B[a]P exposure groups. The rats in B[a]P exposure groups were injected intraperitoneally with B[a]P once a day for 4 consecutive weeks. Then, Morris water maze and shuttle box were used to evaluate the learning and memory abilities of rats; colorimetric assay was used to measure the activities of superoxide dismutase (SOD), Na(+)/K(+)-ATPase, and Ca(2+)/Mg(2+)-ATPase and the content of malonaldehyde (MDA) in the hippocampus; the concentration of Ca(2+) in the hippocampus was measured by fluorescent labeling.
RESULTSCompared with the blank control group and solvent control group, the B[a]P exposure groups exhibited significant increases in escape latency, active avoidance response latency, and passive avoidance response latency and significant decreases in number of platform crossings and active avoidance response frequency in the last test (P < 0.05 for all comparisons), with a dose-effect relationship. In addition, the B[a]P exposure groups had significantly lower activities of SOD, Na(+)/K(+)-AT-Pase, and Ca(2+)/Mg(2+)-ATPase and significantly higher MDA level and Ca(2+) concentration than the blank control group and solvent control group (P < 0.05 for all comparisons), with a dose-effect relationship.
CONCLUSIONThe neurobehavioral toxicity of B[a]P may be related to increased oxidative stress and decreased activities of Na(+)/K(+)-ATPase and Ca(2+)/Mg(2+)-ATPase in the hippocampus of rats.

9.Application value of cardiovascular MR T 1 mapping in patients recovered from COVID-19

Objective:To explore the value of cardiovascular MR (CMR) T 1 mapping in evaluating myocardial injury in patients recovered from COVID-19. Methods:The clinical and image data of 15 patients with COVID-19 (9 with moderate clinical manifestation, 6 with severe clinical manifestation) who underwent CMR screening at 3 months after being discharged from the Second People′s Hospital of Fuyang City during May 2020 to June 2020 were prospective collected. Fifteen COVID-19 patients were selected as case group, and another 11 healthy volunteers were selected as control group. A standardized CMR protocol included cine, native and enhanced T 1 mapping, late gadolinium enhancement (LGE). Cardiac functional parameters, native T 1 value of left ventricular and extracellular volume fraction (ECV) were measured. One way ANOVA was used to assess the difference between CMR parameters among moderate and severe manifestation groups and control group, and LSD- t was used to assess the difference between the three groups. Results:LGE value was negative in all subjects. ECV values were higher in recovered COVID-19 patients with either moderate (27.9%±2.7%) or severe manifestation (30.0%±3.7%) than control group (23.2±1.9%) ( P<0.05); there was no significant difference of ECV values between recovered COVID-19 patients with moderate and severe manifestation ( P=0.100). There was no difference of native T 1 values and other functional and morphologic parameters of left ventricular and right ventricular among recovered COVID-19 patients with moderate and severe manifestation and control group ( P>0.05). Conclusion:CMR myocardial tissue ECV increase in patients who recovered from COVID-19, suggesting subclinical myocardial injury.
10.Microsurgical one-stage repair of hand flexor tendon injuries and rehabilitation: a report of 97 cases.

OBJECTIVETo investigate the therapeutic effects of microsurgical one-stage repair of hand flexor tendon injuries.
METHODSAmong 97 patients with (182 flexor tendons) hand injuries, 59 patients were male and 38 patients were female, ranging in age from 6 to 65 years, with an average of 32 years. Twenty-two patients got injuries by glasses, 32 patients got injuries by knife, 29 patients got injuries by saw, and 14 patients got crush injuries. The tendon injuries in this study consisted of 12 cases of I zone, 35 cases of II zone, 28 cases of III zone, 8 cases of IV zone and 14 cases of V zone. Sixty-eight patients complicated with injuries of blood vessel and nerve, and 53 patients also had fingers fractures. All the patients were treated with modified Kessler method to repair tendon at one-stage, and were given early rehabilitation step by step.
RESULTSAfter the treatment, 97 patients were followed up from 3 to 24 months. According to TAM standard, 48 patients got an excellent result, 39 good, 8 fair and 2 bad.
CONCLUSIONMicrosurgical one-stage tendon repair should be applied. Early rehabilitation and microsurgery repair are important for preventing tendon adhesion.
