Objective To investigate the effects of sex hormone-binding globulin (SHBG)gene in the apoptosis of human trophoblastic cells.Methods The siRNA specific-targeting SHBG gene was transfected into human trophoblastic cells and they were divided into six groups:trophoblasts without transfection in normal control groups(group Ⅰ);transfect liposome in blank control groups(group Ⅱ);transfect nonspecific siRNA in negative control groups(group Ⅲ);transfect SHBG siRNA-Ⅰ,SHBG siRNA-Ⅱ,SHBG siRNA-Ⅲ respectively in trans-fection group(group Ⅳ,Ⅴ,Ⅵ).Hoechst 33258 dying method was used to detect cell apoptosis.SHBG and Caspase-3 mRNA profiling and the level of SHBG and caspase-3 protein were detected by real-time PCR and Western blot.Results There was no statistical significant difference in the gene expression and protein level of SHBG and caspase-3 in group Ⅰ,Ⅱ and Ⅲ (P >0.05).In Ⅳ,Ⅴ and Ⅵ group,there was no statistical significant difference in the expression level of SHBG and caspase 3 (P >0.05).Compared with group Ⅰ,Ⅱ and Ⅲ,the a-mount of SHBG gene expression decreased obviously,the caspase-3 mRNA and protein level increased obviously and the trophoblast cell ap-optosis increased markedly (P <0.05).Conclusion Through siRNA interference technology can reduce SHBG gene expression in human trophoblastic cells,and it can lead to excessive apoptosis of human trophoblasts cells.

3.Observation on clinical therapeutic effect of acupuncture on upper limb spasticity in the patient of poststroke.

OBJECTIVETo observe therapeutic effect of acupuncture at acupoints selected according to rehabilitation medical theory on upper limb spasticity in the patient of poststroke.
METHODSSixty cases were randomly divided into an acupuncture group and an electro-stimulation group, 30 cases in each group. The acupuncture group were treated by acupuncture at the contralateral scalp motor region of the affected limb, Jiquan (HT 1), Chize (LU 5), Daling (PC 7) on the flexor side and Jianyu (LI 15), Tianjing (TE 10), Yangchi (TE 4) on the extensor muscle side of the affected limb; the electro-stimulation group were treated by electric stimulation. The two groups also were treated with necessary medical treatment and anti-spasm rehabilitation motor training. The course was 3 weeks. Modified Ashworth Scale for muscle spasm (MAS), modified Fugl-Meyer Assessment (FMA) for upper limb motor function, and Modified Barthel Index (MBI) for ability of daily living were used for assessment of the therapeutic effect.
RESULTSAfter treatment, the spasm was significantly alleviated, the motor function of the upper limb and daily living ability were significantly increased (P<0.01) in the two groups; after treatment, BMI scores in the acupuncture group was very significantly superior to that in the electro-stimulation group. The total effective rate was 93.3% in the acupuncture group and 86.7% in the electro-stimulation group, with no significant difference between the two groups.
CONCLUSIONProper acupuncture is an effective method for upper limb spasm in the patient of poststroke, and the therapeutic effect is better for mild-moderate spasm of the upper limb.

4.An association study between transforming growth factor-β1 receptor 2 gene polymorphisms and essential hypertension.

OBJECTIVETo evaluate the association between two single nucleotide polymorphisms located in the promoter of transforming growth factor-β1 receptor 2 (TGFBR2) gene and hypertension in Han Chinese population.
METHODSThe subjects were recruited from the population of cluster sampling survey for essential hypertension (EH) in two townships of Yixing city, Jiangsu province in 2009. Overall, 2012 patients with hypertension and 2116 age (± 2 years) and sex-matched unrelated controls were selected. Epidemiological data, physical measurements results and serum glucose and lipid biomarker were collected and detected. Linkage disequilibrium (LD) analysis were applied and two tagging single nucleotide polymorphisms (tagSNP) in 5' upstream of TGFBR2 gene (rs6785358, -3779A/G; rs764522, -1444C/G) were selected for genotyping and analyzing for the association with hypertension.
RESULTSThe frequencies of AA, AG, GG in case and control of rs6785358 were 1455 (72.3%), 517 (25.7%), 40 (2.0%) and 1582 (74.8%), 490 (23.2%), 43 (2.0%) respectively, and CC, CG, GG of rs764522 were 1524 (75.7%), 464 (23.1%), 24 (1.2%) and 1654 (78.2%), 436 (20.6%), 26 (1.2%) respectively. SNP rs764522 was significantly associated with EH and OR (95%CI) were 1.17 (1.01 - 1.36) (P < 0.05) in dominant model after adjustment for confounding factors such as age, sex, glucose, lipids, smoking and alcohol drinking. Further stratification analysis by age, sex, smoking and alcohol drinking indicated that individuals carrying G allele (CG/GG genotype) of SNP rs764522 had higher susceptibility to EH than CC genotype (OR = 1.21, 95%CI: 1.01 - 1.45) (P < 0.05) in ≥ 55 years group. No statistical significance was detected in the distribution of genotypes and allele frequencies for SNP rs6785358 between cases and controls (P > 0.05). Haplotype analysis showed that no significant frequency difference of haplotype structured by rs6785358 and rs764522 was found between cases and controls (P > 0.05), and no significant blood pressure change was found between genotype variations of rs6785358 and rs764522 (P > 0.05).
CONCLUSIONSNP rs764522 of TGFBR2 gene is associated with increased risk of EH in elderly Han Chinese population.

5.Reperfusion arrhythmias in acute myocardial infarction do not enhance myocardial injury.

OBJECTIVETo investigate the clinical implications of reperfusion arrhythmias during primary percutaneous coronary intervention (PCI) for patients with acute myocardial infarction (AMI).
METHODSData from 228 AMI patients in whom the infarct-related artery (IRA) were successfully recanalized by primary PCI were retrospectively analyzed. The 228 patients were divided into 2 groups: myocardial ischemia-reperfusion injury (MIRI) group (n=119) in whom MIRI events occurred within minutes after successful recanalization of IRA, and non-MIRI group (n=109). The 119 patients in MIRI group were further divided into 3 subgroups: severe bradycardia with hypotension (brady-arrhythmia subgroup), lethal ventricular arrhythmias requiring electrical cardioversion (tachy-arrhythmia subgroup), and IRA antegrade flow less than or equal to TIMI 2 grade without angiographic evidence of abrupt closure (no-reflow subgroup).
RESULTS(1) Clinical and angiographic data: Compared with non-MIRI group, MIRI group was characterized by more inferior infarct location, shorter ischemic duration, more frequently right coronary artery as IRA, more diseased vessels, more often TIMI 0 grade of initial antegrade flow in IRA, less pre-infarction angina, more renal insufficiency, and higher in-hospital mortality (13.4% vs. 4.6%, P=0.021). (2) The peak CK level was remarkably lower in brady-arrhythmia subgroup than that in non-MIRI group (2010 IU/L vs. 2521 IU/L, P=0.039). The peak CK or CK-MB level was notably higher in no-reflow subgroup than in non-MIRI group (4573 IU/L, 338 IU/L, respectively, P=0.000). (3) Left ventricular ejection fraction in no-reflow subgroup was significantly lower than in non-MIRI group (38.7% +/- 8.3% vs. 51.2% +/- 8.1%, P=0.000), left ventricular end-diastolic volume in no-reflow subgroup was greater than that in tachy-arrhythmia subgroup [(135 +/- 32) ml vs. (105 +/- 19) ml, P=0.029].
CONCLUSIONReperfusion arrhythmias may imply the existence of much survived myocardium and do not enhance myocardial damage, while no-reflow increases myocardial injury and induces permanent impairment of cardiac function.

6.Expression of Chk1 in human sperm and its significance.

OBJECTIVETo investigate the expression of the Chk1 gene in human sperm and its clinical significance.
METHODSWe collected 80 semen samples and divided them into 4 groups of equal number: normal, oligospermia, asthenozoospermia and oligoasthenozoospermia. The Chk1 expression and its relative level were detected by Western blot and RT-PCR, sperm DNA damage and gradient changes assessed by DNA ladder analysis, and sperm apoptosis determined by Annexin V/PI double staining in each group.
RESULTSThe Chk1 gene was expressed in all the four groups, but with significant differences (P < 0.01); the relative levels of CHK1 protein were similar to those of Chk1 mRNA in the normal, oligospermia, asthenozoospermia and oligoasthenozoospermia groups, which were 1.00 +/- 0.22, 0.76 +/- 0.10, 0.45 +/- 0.08 and 0.37 +/- 0.07, respectively. DNA ladder analysis showed a marked DNA ladder in the asthenozoospermia and oligoasthenozoospermia groups. Sperm apoptosis was markedly increased in the oligospermia, asthenozoospermia, oligoasthenozoospermia and 100% graded sperm groups ([ 8.3 +/- 0.60]%, [11.6 +/- 0.92]%, [12.5 +/- 1.43]% and [17.0 +/- 1.98]%), as compared with the normal group ([7.6 +/- 0.34]%) (P < 0.05).
CONCLUSIONChk1 is expressed in human sperm, but differently in different semen quality groups. And its expression is correlated with sperm DNA damage and apoptosis; its reduction may lead to declined sperm repair and increased sperm apoptosis and thus affect semen quality.

7.Clinical efficacy and safety evaluation of cornu cervi degelatinatum in the treatment of leukopenia after chemoradiotherapy of malignant tumor

Objective To evaluate clinical efficacy of cornu cervi dege-latinatum in treating leucopenia after malignant tumor chemoradiotherapy . Methods Sixty cases with leucopenia after malignant tumor chemoradio-therapy and kidney yang deficiency in Chinese medicine were randomly divided into treatment group and control group ,30 cases in each group. Patients in control group were treated with leucogen 40 mg once, 3 times a day.Patients in treatment group were treated with cornu cervi degelati-natum 100 g once a day .Two weeks for a course , and both groups were treated for 3 courses.The leukocyte account before and after treatment and the clinical effect after treatment of two groups were compared , also the security was evaluated . Results The total effective rate was 86.67%in treatment group , much higher than 73.33%in control group ( P<0.05 ) .The leukocyte number apparently rised after treatment in both groups ( P <0.05 ) , while in treatment group , it rised to much higher than that in control group ( P<0.05 ) .The incidence of adverse drug reactions of the two groups was compared , and the difference was not statistically significant ( P >0.05 ) . Conclusion Compared to leucogen , cornu cervi degelatinatum has better clinical effect on the treatment of leucopenia after malignant tumor chemoradiotherapy with good security , which is worth recommended in clinical .
8.The study on the 0, 12 month vaccination schedule' of Healive inactivated hepatitis A vaccine in children.

OBJECTIVETo evaluate the safety, immunogenicity and fit dosage of Healive inactivated hepatitis A vaccine (HAV) in children.
METHODSA total of 85 susceptible aged 4 - 10 years with HAV seronegative children, had been enrolled from two adjacent villages in a county. The volunteers were randomized allocated into two groups and to receive a priming dose of 250 U/0.5 ml/dose or 500 U/1.0 ml/dose of Healive vaccine, produced by Sinovac Biotech Co, Ltd. A booster of the same dose was given at 12th month. Local and systemic side effects were examined and seroconversion rate as well as geometric mean titers of anti-HAV antibody were tested at 3-week, 12-month after the primary dose and at 1 month after the booster dose.
RESULTSThe vaccine was well tolerated in both groups. At 21 days after the primary dose, the seroconversion rates were 94.4%, 100.0% and geometric mean titers (GMT) were 195 mIU/ml and 370 mIU/ml in 250 U and 500 U groups respectively. At 12 months after the primary dose, the seroconversion rate of anti-HAV was 100.0%, and GMT raised to 361 mIU/ml, 456 mIU/ml (P > 0.05) respectively. One month after the booster dose, GMT raised to 14 893 mIU/ml, 21 696 mIU/ml.
CONCLUSIONGMT of the 0, 12 month schedule was higher than other schedule after the booster vaccination. The Healive inactivated vaccine can be used for emergency vaccination. The Healive inactivated vaccine produced by Sinovac Company Ltd was safe and highly immunogenic. Two hundred and fifty U/dose was considered appropriate for children.

9.Effect of Buyang Huanwutang on Protein Expression of MAP-2, NF-M and GAP-43 After Sciatic Nerve Transection Anastomosis in Rats

Objective:To investigate the effects of Buyang Huanwutang on the expression of microtubule-associated protein-2(MAP-2), neurofilament-M(NF-M), and growth associated protein-43(GAP-43)in rat sciatic nerve after sciatic nerve transection and anastomosis. To explore the mechanism of Buyang Huanwutang promoting peripheral nerve regeneration. Method:SD rats were selected as the experimental subjects, and sciatic nerve transection model was selected as the experimental model. They were randomly divided into model group, sham operation group, Buyang Huanwutang group high, medium and low dose (29.6, 14.8, 7.4 g·kg-1)group, and mecobalamin (0.156 mg·kg-1)group, the model group and the sham operation group were given distilled water intragastric administration. After successful modeling, each group was treated with relevant drugs for 4 weeks. After 4 weeks, sciatic nerve function index(SFI), degree of inclined plate test and hematoxylin-eosin(HE)of sciatic nerve in each group were tested. The expression levels of MAP-2, NF-M, and GAP-43 at the sciatic nerve anastomosis site were detected by immunohistochemistry and Western blot. 10.Correlation of An Ultrasonic Scoring System and Intraoperative Blood Loss in Placenta Accreta Spectrum Disorders: A Retrospective Cohort Study.

Objective:
This study aims to investigate the correlation of an ultrasonic scoring system with intraoperative blood loss (IBL) in placenta accreta spectrum (PAS) disorders.
Methods:
A retrospective cohort study was conducted between January 2015 and November 2019. Clinical data for patients with PAS have been obtained from medical records. Generalized additive models were used to explore the nonlinear relationships between ultrasonic scores and IBL. Logistic regressions were used to determine the differences in the risk of IBL ≥ 1,500 mL among groups with different ultrasonic scores.
Results:
A total of 332 patients participated in the analysis. Generalized additive models showed a significant positive correlation between score and blood loss. The amount of IBL was increased due to the rise in the ultrasonic score. All cases were divided into three groups according to the scores (low score group: ≤ 6 points,
Conclusions
The risk of blood loss equal to or greater than 1,500 mL increases further when ultrasonic score greater than or equal to 10 points, the preparation for transfusion and referral mechanism should be considered.
