1.Effect of acupuncture plus exercise on pain and motor function in patients with knee osteoarthritis

Objective: To observe the effects of acupuncture combined with isokinetic eccentric exercise on pain and motor function in patients with knee osteoarthritis (KOA). Methods: Sixty-four patients with KOA were randomly divided into an observation group and a control group, with 32 cases in each group. The observation group received acupuncture and isokinetic eccentric exercise, and the control group received isokinetic eccentric exercise and oral intake of meloxicam tablets. Both groups were treated for 4 weeks. Before and after treatment, the visual analog scale (VAS) score, the lysholm knee scale (LKS) score, and the quadriceps peak torque (PT) were evaluated. Results: The total effective rate was 93.8% in the observation group, versus 81.3% in the control group. The difference between the two groups was statistically significant (P<0.05). After treatment, the VAS scores of the two groups were lower than those before the treatment; both LKS score and quadriceps PT were higher than those before treatment, and the differences were statistically significant (allP<0.05). The VAS score, LKS score and quadriceps PT (angular velocity was 60 °/s) of the observation group were significantly different from those of the control group (all P<0.05). Conclusion: Acupuncture plus isokinetic eccentric exercise is effective in treating KOA. The combined treatment can reduce the pain of KOA patients, improve joint function, strengthen quadriceps, improve joint stability, and its curative effect is better than isokinetic eccentric exercise plus oral intake of meloxicam tablets.
2.Application of asymmetrical Z-plasty in correction of epicanthus

Objective To investigate a new method in correction of epicanthus.Methods Asymmetrical Z-plasty was used in correcting epicanthus and lengthening palpebral fissure in 42 cases with 84 eyes.It could be combined with double-eyelid-blepharoplasty to create beautiful eyes.Results All 42 cases with 84 eyes had satisfactory outcome after half a year to 2 years follow-up.The epicanthus scar was conspicuous in the first 6 weeks and it subsided gradually.In the three cases.the hypertrophic scar occurred in the epicanthus,but it softened and became inconspieuous 6 mouths after operation.Conclusion Asymmetrical Z-plasty is a new and reliable method for correction of epicanthus with inconspicuous scar and little chance of recurrence that can be widely used.

Vascular morphology and function changes exist in degenerative retinopathy,which have an important influence on the development and prognosis of the disease.The literature has been reported that retinal vascular structure and function change can be found early by using the retinal oximeter,color Doppler flow imaging,ocular pulse amplitude and so on,as well as exploring the primary and secondary factors affecting the development of retinal blood vessels.This article reviews the vascular behavior characteristics in degenerative retinopathy and their influence factors.
4.Factors affecting the prognosis of ruptured intracranial aneurysm

Objective To investigate factors related to the prognosis of patients with ruptured intracranial aneurysm.Methods Clinical data of 165 patients with ruptured intracranial aneurysm treated with surgery were retrospectively analyzed.The age,gender,PAASH scale on admission,smoking,hypertension,hyperlipemia,cerebral vasospasm,timing of surgery and mode of surgery,etc.were analyzed by the simple and multiple Logistic regression models.Results Simple Logistic regression analysis indicated that age,PAASH scale on admission,smoking,hypertension,timing of surgery and mode of surgery were influencing factors for the prognosis of patients with ruptured intracranial aneurysm(all P<0.05).Multiple Logistic regression analysis showed that age,PAASH scale on admission(OR=2.542),smoking(OR=5.772),hypertension(OR=7.150),timing of surgery(OR =2.908) and mode of surgery(OR =0.161) were independent risk factors for the prognosis of patients with ruptured intracranial aneurysm(all P<0.05).Conclusions Accuratel and systemsatic assessment of patient conditions and the optimal timing and mode of surgery are the key to improving the prognosis of patients with ruptured intracranial aneurysm.
5.The proposals and analysis of relevant factors of TCM clinical pathway

This paper analyzes the explore the factors influencing the TCM clinical pathway using the fishbone dragram. We found that the main impacting factors of TCM clinical pathway were government policy factors, hospital management factors and personal factors. It was suggested to promote Chinese medicine clinical pathway pilot job, not only need to cooperate with the administrative departments of health and medical institutions, also need to strengthen the change of the idea of both doctors and patients.
6.Specific stimulation of B cell epitope peptides from Mycobacterium tuberculosis antigen on human peripheral γδT cells

Objective:To investigate the specific stimulation of the B cell epitope peptides of Mycobacterium tuberculosis antigen (Mtb-Ag) on human peripheral γδ T cell proliferation.Methods: We selected the sequences of B cell epitope peptide from Mtb-Ag that were reported in literature and T cell epitope peptide that recently identified in this laboratory to synthesize six peptides of B cell epitopes (BP1-BP6) and two peptides of γδ T cell epitopes (TP14,TP15).The 24-well culture plates were coated with these peptides.The PBMCs were isolated from peripheral blood of healthy individuals and stained with CFSE,followed cultured for 12 days in the IL-2 containing medium.Mtb heat resistant antigen ( Mtb-HAg ) group as positive control and IL-2 only group as negative control.The percentages and proliferation index of γδ T cells were determined by flow cytometry.Results: By using Wilcoxon signed rank test for paired comparison of negative control group,the percentages of γδ T cells in cultured PBMCs with BP2,BP4 and TP14, TP15 and Mtb-HAg increased significantly in 14 samples (P<0.05);and the proliferation index of γδT cell in cultured PBMCs with BP2,BP4,BP5,BP6 and TP14,TP15 increased significantly in 7 samples (P<0.05).Conclusion: Taken together,the B cell epitope peptides from Mtb Antigens are capable of stimulating the γδ T cell proliferation specifically in vitro.Although there was individual difference inγδT cell proliferative response to B cell epitope peptides,these results strongly suggest the B cell epitope peptides also can specifically trigger the TCR ofγδT cells.

9.Study on p53 gene construction and functional status of PA-1 cells derived from human teratocarcinoma

Objective:To understand the construction,functional status and the meaning of p53 gene of PA-1 cells derived from human teratocarcinoma.Methods:DNA basic sequence analysis,FASAY(functional and sequencing analysis of separated alleles in yeast)and Western blot analysis were used.Results:Direct sequence analysis RT-PCR products showed both wild and mutated bands(p53 colon 239 mutation), FASAY showed that one allele of the p53 gene was active(wild) .while the other was inactive( mutant) .In Western blot analysis haven't defect the gene protein of p53, while the PA-1 cells expressed the p21 protein to a lesser extent than normal human tibroblaste. Conclusion:One type of the p53 allele gene of PA-1 cells of human teratocarcinoma was detected as missense mutation after cultured for more than twenty years, and this mutation is not sufficient to induce the instabih'ty of the chromosome of cell line. So that the study for the maintenance contruction of the stability nuclein type of PA-1 cells is very important to determin whether the genes is stability or instability of chromosomes.
10.Characteristics of chromosome of teratocarcinoma and its influence elements

AIM: To investigate the characteristics of chromosome of teratocarcinoma and its influence elements. METHODS: We used the methods of G-banded karyotype analysis, DNA basic sequence analysis and Western blot and the others, and studied the chromosome karyotype and the status of p53 gene of teratocarcinoma PA-1 cell line which had been cultured for 407-445 passages for 20 years. RESULTS: More than 80% PA-1 cells still maintained the near-diploid karyotype ,after passage 30 the cells appeared with M1 and M2 chromosomal markers because of a balanced translocation between chromosomes 15 and 20. DNA directional sequence analysis of RP-PCR products revealed that there were both wild and mutated band (p53 codon 239 mutation), Western blot did not detect mutational p53 gene protein,while p21 protein expression lower than that in normal human fibroblasts. CONCLUSION:Missense mutation of one of the p53 allele gene of PA-1 cells in human teratocarcinoma was detected after cultured for more than twenty years, which was not sufficient to induce the instability of the chromosome of cell line.
