1.Study on p53 gene construction and functional status of PA-1 cells derived from human teratocarcinoma

Objective:To understand the construction,functional status and the meaning of p53 gene of PA-1 cells derived from human teratocarcinoma.Methods:DNA basic sequence analysis,FASAY(functional and sequencing analysis of separated alleles in yeast)and Western blot analysis were used.Results:Direct sequence analysis RT-PCR products showed both wild and mutated bands(p53 colon 239 mutation), FASAY showed that one allele of the p53 gene was active(wild) .while the other was inactive( mutant) .In Western blot analysis haven't defect the gene protein of p53, while the PA-1 cells expressed the p21 protein to a lesser extent than normal human tibroblaste. Conclusion:One type of the p53 allele gene of PA-1 cells of human teratocarcinoma was detected as missense mutation after cultured for more than twenty years, and this mutation is not sufficient to induce the instabih'ty of the chromosome of cell line. So that the study for the maintenance contruction of the stability nuclein type of PA-1 cells is very important to determin whether the genes is stability or instability of chromosomes.
2.Characteristics of chromosome of teratocarcinoma and its influence elements

AIM: To investigate the characteristics of chromosome of teratocarcinoma and its influence elements. METHODS: We used the methods of G-banded karyotype analysis, DNA basic sequence analysis and Western blot and the others, and studied the chromosome karyotype and the status of p53 gene of teratocarcinoma PA-1 cell line which had been cultured for 407-445 passages for 20 years. RESULTS: More than 80% PA-1 cells still maintained the near-diploid karyotype ,after passage 30 the cells appeared with M1 and M2 chromosomal markers because of a balanced translocation between chromosomes 15 and 20. DNA directional sequence analysis of RP-PCR products revealed that there were both wild and mutated band (p53 codon 239 mutation), Western blot did not detect mutational p53 gene protein,while p21 protein expression lower than that in normal human fibroblasts. CONCLUSION:Missense mutation of one of the p53 allele gene of PA-1 cells in human teratocarcinoma was detected after cultured for more than twenty years, which was not sufficient to induce the instability of the chromosome of cell line.
3.The effect of MOTOmed movement therapy on balance and ability in the activities of daily living in patients with hemiplegia after stroke

Objective To investigate the effect of MOTOmed movement therapy on balance and ability in the activities of daily living (ADL) of convalescing stroke patients with hemiplegia. Methods Forty convalescent stroke patients exhibiting hemiplegia were randomly divided into a treatment group and a control group with 20 cases in each group. The control group received routine rehabilitation training; the treatment group received MOTOmed training in addition. Balance function of all the patients was assessed using Berg's balance scale (BBS) , and the Barthel Index ( BI) was used to assess ADL ability at the beginning of the program and after 6 weeks of treatment. Results Balance and ADL ability improved significantly in both groups after 6 weeks of treatment. The effect in the treatment group was significantly better than in the control group. Balance and ADL ability were positively correlated. Conclusions Applying MOTOmed therapy along with routine rehabilitation training can distinctly improve balance and the ADL ability of hemiplegics after stroke.
5.Multi-disciplinary and comprehensive treatment of colorectal cancer

The curative effects of multi-disciplinary and comprehensive treatments for colorectsl cancer have been widely recognized in recent years.Because of the diversification of colorectal cancer treatment and the relatively mature technology,the current treatment of colorectal cancer could embody the multi-disciplinary and comprehensive treatment.The curative effect of neo-adjuvant chemoradiotherapy has been validated by several recent clinical trials.Other treatment methods such as Chinese medicine,radiofrequency ablation and hepatic artery infusion therapy are also involved in the comprehensive treatment,which provide considerable curative effects.
6.Three kinds of periodontal stabilization splints in severe periodontitis of the mandibular anterior teeth:which one is preferred?

BACKGROUND:It is believed that the splintage can promote periodontal tissue repair and improve the prognosis of loose teeth. However, the force of the splint is associated with the rigidity of the splint and adaptation between the teeth and the splint.
OBJECTIVE: To evaluate the therapeutic effects of the nylon composite resin periodontal splint, the
fiber-reinforced composite periodontal splint and the bracket periodontal splint on the treatment of severe periodontitis of the mandibular anterior teeth.
METHODS:Seventy-two patients with severe periodontitis of the mandibular anterior teeth were randomized into 3 groups, with 24 cases in each group: nylon composite resin periodontal splint, fiber-reinforced composite
periodontal splint and bracket periodontal splint groups. The clinical effects of the three kinds of periodontal splints and periodontal indexes were compared at 1, 6 and 12 months after splinting.
RESULTS AND CONCLUSION: The clinical effects in the group of the fiber-reinforced composite periodontal
splint were significantly better than those in the other two groups at 1, 6, 12 months, which were 100%, 100% and 95.8%, respectively. The plaque index, bleeding index, and pocket depth were worse in the group of the nylon
composite resin periodontal splint than the other two groups (P < 0.05). Results suggest that the fiber-reinforced composite periodontal splint is preferred to preserve the loosened teeth with severe periodontitis.
7.The relationship between the fracture of the clavicle which was fixed by torsional-shaped plate and nonunion

Objective To discuss the relationship between the fracture of the clavicle which was fixed by tor-sional-shaped plate and nonunion after operation.Methods Retrospective analysis was conducted in 426 patients with midshaft clavicle fractures.Among the patients,the A group used torsional-shaped plate to meet the anatomical structure of the clavicle:the medial plate matched under the front face of the clavicle,and the lateral plate matched the up face of the clavicle.And the traditional method was used in the B group.Patients were followed up for at least 12 months,the patients were judged by X-ray criteria for nonunion.Results A total of 28 patients with nonunion, including 4 cases in A group,24 cases in B group.After statistics processing,the difference between A group and B group was significant (χ2 =6.679,P=0.010).Conclusion We find that the treatment of the fracture of the middle part of the clavicle with torsional-shaped plate can effectively reduce the incidence of nonunion.
8.Progress on prevention for anterior knee pain after primary total knee arthroplasty.

Total knee arthroplasty (TKA) identified as an effective treatment for ultimate knee joint disease can effectively relieve pain, correct deformity, improve knee function and enhance the quality of life of patients. Patient satisfaction has been increasingly considered as an important factor in evaluating the success of primary TKA. Anterior knee pain that usually appears in the region of the anterior knee is a recognized complaint for primary TKA and has a strong impact on the improvement of knee function and patient satisfaction of primary TKA. Accordingly, the relief of anterior knee pain has become one of the primary goals of primary TKA. At present, soft tissue lesions around the patellar caused by patellar maltracking and the elevation of internal pressure in subchondral bone because of the high contact stress of patellofemoral joint are both considered as the mechanism of anterior knee pain. For the past few years,on increasing number of studies have focused on the prevention of anterior knee pain following primary TKA. However, none of the past treatment such as patellar resurfacing, patellar denervation without patellar resurfacing or a mobile-bearing prosthesis has a good and affirmative effect on it. The prevention and treatment of anterior knee pain following primary TKA still is a difficult solved problem. To address this problem, we need further researches about the cause of anterior knee pain, knee joint prosthesis and biomechanics of patellofemoral joint, as well as lots of randomized controlled trials.

9.Mutagenicity evaluation of the hydrolysate of Meretrix meretrix Linnaeus soft tissue

Objective To evaluate the mutagenicity of hydrolysate of Meretrix meretrix Linnaeus soft tissue, so as to provide experimental basis for its exploitation.Methods Three mutagenicity tests were used to evaluate the mutagenic effects, including Ames test, CHL chromosome aberration assay and bone marrow micronucleus assay in mice.Results In Ames test, the revertant colonies numbers in each group were twice less than the numbers of spontaneous revertant colo-nies, five bacterial strains showed negative results with or without S9 activation, and the result of Ames test was negative. The CHL chromosome aberration assay and bone marrow micronucleus assay showed that the chromosome aberration rate and micronucleus rate of each dose group showed no significant difference compared with the negative control group, respec-tively ( P>0.05) .Conclusions Under this condition, the results show that all of the Ames test, chromosome aberration assay and bone marrow micronucleus assay are negative, and no mutagenicity is observed in the hydrolysate of Meretrix mer-etrix Linnaeus soft tissue.
10.Pediatric liver transplantation from split livers: a single-center experience

Objective To analyze and evaluate the efficay of split liver transplantation in children.Method From September 2006 to December 2014,210 children were treated with liver transplantation in Tianjin First Central Hospital.The clinical data were retrospectively analyzed and the difference in postoperative survival was compared between the groups.The 210 childrens were categorized into living donor liver transplantation group (183 cases) and split liver transplantation group (27 cases) based on their operation styles.In living group,all donors to recipients were immediate relatives within three generations.In split group,all donors were men,and livers were obtained from no heartbeat donors.Postoperatively,tacrolimus combined a duplex of prednisolone served as immunosuppression scheme.The survival and incidence of complications were observed.Result There was significant difference in the sex ratio between two groups (P<0.05).The donor liver cold ischemia time was significantly longer in split group than in living group (P<0.05).The 1-month,6-month,1-year and 2-year overall survival rate in 210 recipients was 99.5%,98.1%,96.2% and 94.2% respectively.The median follow-up time in living group and split group was 15.2months and 26.1 months,respectively.The 1-mont,6-month,1-year and 2-year survival rate was 99.5%,96.7%,92.6% and 74.1 % in living group,and 97.8%,96.2%,77.8% and 74.0% in split group,respectively (P<0.05).During the follow-up period,8 cases died (29.6%) in split group (5deaths due to infection and sepsis,and 3 deaths due to multiple organ failure),and 10 cases died (5.5%) in living group (6 deaths due to infection and sepsis,and 4 deaths due to multiple organ failure).Conclusion In the case of strict selection of donors,split liver transplantation can obtain good effect,but the incidence of complications is higher than living donor liver transplantation.Especially,the biliary complications should be prevented and managed actively.
