4.Effect of zhimu and huangqi on cardiac hypertrophy and response to stimulation in mice.

OBJECTIVETo observe the effect of Zhimu and Huangqi on increasing heart rate and cardiac hypertrophy, and its bradycardia effect, to evaluate the exercise capacity and response to stimulation and to discuss the use of Zhimu in treating heart failure.
METHOD1. Rats were treated with isoproterenol 1.25 micrograms.kg-1 i.p., and then the increasing of heart rate and the bradycardia effect were measured after administrated with Zhimu or Huangqi for 3 days. 2. Mice cardiac hypertrophy model was established by s.c. ISO 1 mg.kg-1, 2 times per day for 14 days and heart-weight-index was measured after Zhimu and Huangqi was given orally alone or jointly for 14 days. 3. Abdominal aorta banding operation was done in mice and 3 weeks after operation, they were administrated for 2 weeks, and then run-time(exercise capacity), quiet heart rate, ISO response heart rate after i.p. isoproterenol and heart-weight-index were measured. 4. Cardiac hypertrophy model (inject ISO 14 days) mice were administrated for 12 days, and the mortality and dying time of mice in cold (-20 degrees C) and heat(45 degrees C) stimulative condition were observed.
RESULTZhimu could cut down the increasing of heart rate induced by ISO, decrease significantly heart-weight-index in cardiac hypertrophy mice, reduce the quiet heart rate and prolong the run time in abdominal banding model. Zhimu combined with Huangqi could also improve the ISO response in abdominal banding model mice, reduce the mortality and delay dying time of mice in stimulative condition.
CONCLUSIONZhimu combined with Huangqi can slow down heart rate, enhance the reserve force of the heart, and improve the response capacity of cardiac hypertrophy mice in stimulative condition.

5.Study on the effect of zhimu combined huangqi on improving renal hypertension rat's cardiac dysfunction.

OBJECTIVETo observe the effect of Zhimu and Huangqi used singly or combinatively on improving experimental cardiac dysfunction, and mainly to observe zhimu's effect on restraining sympathetic nerve and blocking beta-adrenergic-recepter and huangqi's effect of improving hemodynamics on heart failure.
METHODTwo-clib one kidney operation was done to make renal hypertension rat model, 8 weeks after operation, rats were divided into groups and medicated for 6 weeks, and then their heart rate and blood pressure were measured, left ventricle was cannulated to estimate heart function, and heart-weight-index and left-ventricle-weight-index were measured.
RESULTZhimu could slow rats heart rate obviously, prevent cardiac remodeling, but did not affect cardiac function remarkably; Huangqi could reduce blood pressure, heighten +dp/dtmax and -dp/dtmax remarkably; the combined use of the two drugs could decrease plasma catecholamine concentration, adjust myocardium cAMP content, and improve heart function obviously.
CONCLUSIONZhimu and Huangqi can protect experimental cardiac dysfunction, and the combined use is better than the single use, which shows it better to use the two drugs combinatively in treating heart dysfunction.

Chinese Medical Association (CMA) is an academic society and the Chinese Medical Doctor Association (CMDA) is an administrative organization with academic character. The main functions of CMDA are setting up the system for specialty training, training for residents of different disciplines, establishing training bases, and safeguarding interests of doctors. The author also analyzes the drawbacks in this regard in field of burn surgery in China at the present time, and venture to put forward new suggestions for strengthening and perfecting the frame of CMDA.

8.Experimental study on the inhibition of fibroblast contraction by alpha smooth muscle actin fusion protein.

OBJECTIVETo investigate the influence of alpha smooth muscle actin fusion protein (alpha-SMA-FP) on fibroblast contraction, in order to find a new way to control scar contracture.
METHODSThree dimensional gel culture model of fibroblasts populated collagen lattices (FPCLs) was employed in the study. The fibroblasts were cultured in gel for 5 days. The cells in experimental group were processed by alpha-SMA-FP in dose of 5, 10, 50, 100 and 250 mg/L, respectively. The cells were therefore divided into E(1), E(2), E(3), E(4) and E(5) groups. Blank control was set to be C(1) group, and the cells processed by 250 mg/L of alpha-SMA-FP be C(2) group. The contraction rate was calculated by measuring the diameters of the gel before and after the procession. The change of contraction rate in E5 group was observed after the alpha-SMA-FP being rinsed out. Immunofluorescent staining of alpha-SMA-FP was carried out in fibroblasts.
RESULTSThe contraction rate in C1 and C2 groups showed no difference, being (58.6 +/- 3.1)% and (56.2 +/- 4.9)% respectively, while that in E1 to E5 groups was (45.56 +/- 4.1)%, (42.3 +/- 4.2)%, (41.8 +/- 3.6)%, (37.6 +/- 5.8)% and (26.4 +/- 4.7)%, respectively. However, the contraction rate in E5 was (53.3 +/- 5.6)% after the alpha-SMA-FP had been rinsed out. The difference of the rates among control group and experimental groups, especially in E5 after alpha-SMA-FP being rinsed out, was significant (P < 0.05 or 0.01). alpha-SMA-FP was located on the fibers of the fibroblasts as shown by staining, while the alpha-SMA was not stained. Nevertheless, the staining was obvious in control group.
CONCLUSIONalpha-SMA-FP could inhibit the contraction of fibroblasts specifically with dose dependent effect.

9.Role of GABAAα3 and GABAB receptors in ventrolateral periaqueductal gray in rats with acute pain

Objective To investigate the role of GABAAα3and GABAB receptors in the ventrolateral periaqueductal gray in the development of paw acute pain in rats.Methods Twelve male SD rats, weighing 280~320 g, were randomly divided into two groups: normal saline group (group NS), formaldehyde-induced pain group (group F), 6 rats in each group.In group F, rats were subcutaneously injected with 2% formaldehyde 50 μl into the ventral surface of right hind paw to induce periphery inflammatory pain.In group NS, rats were subcutaneously injected with normal saline into the ventral surface of right hind paw.Mechanical threshold was assessed using von Frey hairs for every ten minutes.The rat pain behavior scores were recorded for every five minutes.The thickness of skin and skin temperature were recorded for every fifteen minutes.Results Mechanical hyperalgesia were induced in group F after formalin injection into right hind paw.Compared with group NS, rat pain behavior scores were increased significantly in group F at all time points after injection, mechanical threshold were decreased significantly in group F at 10-60 min after injection, the temperature of the skin and the skin thickness were increased significantly in group F at 15-60 min after injection (P<0.05), the levels of the expression of GABAAα3 and GABAB were significantly increased in group F (P<0.05).Conclusion GABAAα3and GABAB receptors mediates formalin-induced hyperalgesia at ventrolateral portion of the PAG (vlPAG) of rats.
10.Clinical study of nutritional support in patients with acute peritonitis caused by colonic perforation

Objective:To investigate the effect of different postoperative nutritional support on the gastrointestinal function and nutritional status in acute colon peforation patients.Methods:60 cases of acute emergency operation patients with perforation of the colon,according to the given nutritional support treatment of the different ways,were divided into enteral parenteral joint nutrition (EN + PN) group and total parenteral nutrition (TPN) group.Clinical therapeutic effects of two groups were compared.Results:The results of EN + PN group were significantly better than the those in TPN group (P < 0.05).Conclusion:EN + PN model can improve the postoperative nutritional status and accelerate the recovery of patients with acute colonic perforation.
