1.RNA interference of silent mating type information regulation 2 homolog 1 SIRT1 arrests cell cycle progress of prostate cancer PC3 cells

Objective:To observe the effects of double-stranded small interfering RNA (siRNA) of the silent mating-type infor-mation regulation 2 homolog 1 (SIRT1) on the cell proliferation, cell cycle progression, and expression levels of the cell cycle negative regulators. These regulators include P21, P27, and phosphorylated retinoblastoma (PRb) proteins present in prostate cancer PC3 cells. This work further aims to explore the possible underlying mechanism for such effects. Methods:PC3 cells were cultured in vitro and then randomly divided into the mock group, scramble siRNA transfected group, and SIRT1 siRNA-transfected group. SIRT1 siRNA ef-ficiency was examined through reverse transcription polymerase chain reaction and Western blot analysis. The inhibitory rate of PC3 cell growth was determined through a methyl thiazolyl tetrazolium assay, and the cell cycle was investigated with the use of flow cytom-etry. The P21 and P27 protein expression levels and PRb status were determined by Western blot assay. Results:Compared with those of the mock and scramble siRNA groups, the expression levels of SIRT1 mRNA and protein significantly decreased in SIRT1 siR-NA-transfected cells. In addition, the inhibitory rate of PC3 cell growth was markedly increased, and the cell cycle of the PC3 cells was arrested at the G1 stage. The expression levels of negative cell cycle regulators, including P21 and P27 protein levels increased, whereas Rb protein phosphorylation was inhibited in SIRT1 siRNA-transfected PC3 cells. Conclusion: SIRT1 RNA interference inhibits PC3 cell growth and arrests cell cycle progression through the upregulation of the P21 and P27 proteins and the inhibition of Rb protein phosphorylation.

7.Research Progress of Invasive Pneumococcal Disease

Streptococcus pneumoniae is one of the most common cause of invasive disease,such as bacteremia,meningitis,and empyema,et al.But there has significant difference on the incidence of invasive pneumococcal disease among different regions and differernt groups of people.In addition the severity and mortality of invasive pneumococcal disease are closely related to the changes of serotypes,virulence of streptococcus pneumoniae and also the human immune response.The pneumococcal vaccination is an important measure to prevent streptococcus pneumoniae infection,providing good protection to vaccinees and createing herd immunity effect.This article briefly describes the pathogenesis,risk factors and preventive strategies of invasive pneumococcal disease.
8.Analysis on laws of tradition Chinese medicine treating diabetic retinopathy

Objective To investigate the laws of the treatment of diabetic retinopathy with traditional Chinese medicine.Methods By searching domestic literature of treating DR with traditional Chinese medicine in CNKI from 2000 to 2010, made primary summary and analyzed. Results 85 eligible published documents were found involving 147 medicinal herbs. Herbs with the functions of tonifying, activating blood circulation and eliminating stasis and heat were mostly seen.Cases reported in literature were mainly non-proliferative lesions,occupying a ratio of 58.82%;drug observation time was mainly 12 weeks, occupying a ratio of 44.17%. Conclusion Asthenia,statis and heat were pathological factors existing throughout the whole process of DR. Invigorating, romoting blood circulation, and,cooling blood should be the basic therapeutic methods.
9.Total auricular reconstruction using nitinol shape memory alloy stent: Scanning electron microscope verification of surrounding tissue structure changes at 6 and 12 months after implantation

BACKGROUND: Plenty of materials have been used as stent in total auricular reconstruction with varying advantages and disadvantages. The in vitro prepared tissue engineered complex cartilage stent has been not applied in clinic.OBJECTIVE: To observe the ultrastructure changes of surrounding tissues following total auricular reconstruction with nickel-titanium (NiTi) shape memory alloy stent in vivo.DESIGN, TIME AND SETTING: The randomized, controlled, animal experiment was performed in the Central Laboratory of EENT Hospital of Fudan University, Anhui Provincial Hospital and Department of Nuclear Medicine, Huashan Hospital of Fudan University from April 2004 to October 2005.MATERIALS: A total of 12 New Zealand rabbits were randomized divided into the skin-expanded and unexpanded groups, with 6 animals in each group.METHODS: Each animal underwent subcutaneous implantation with a NiTi alloy stent like human auricle. In the skin-expanded group, the stent was implanted after skin expansion, while no treatment in the unexpanded group.MAIN OUTCOME MEASURES: The ultrastructure changes of surrounding tissues were detected by scanning electron microscope at months 6 and 12 after implantation.RESULTS: Structure of nickel-titanium alloy implant stent make fibrovascular ingrowth more easily. The surface of nickel-titanium alloy stent was smooth without corrosion. There was no evidence of infection in the host tissue. Microstructure of Ni-Ti stent wires showed no morphologic and functional changes after subcutaneously implanted in vivo, and the difference between 2 groups had no significance.CONCLUSION: NiTi shape memory alloy stent can smoothly pass the process of fibrovascularization with less complication.According to the examination by SEM, the nickel-titanium stent showed good biocompatibility.
10.Spatial Point Pattern Analysis of Birth Defects

Objective To detect the spatial point pattern distribution rules of neural tube defects.Methods The kernel density estimation and Ripley's K-function were used to analyze the spatial point pattern of the neural tube birth defects in Heshun county in 1998-2001.Results The kernel density estimation result showed that there was two clusters' distribution in central area and southeastern area respectively.In addition,the result by the Ripley's K-function presented that the location of neural tube birth defects had a significant cluster tendency in the spatial distance from 3.17 to 10.41 kilometers in the investigated area.Conclusion These results can provide an important clue for identifying the relations between environment risk factors and birth defects in this area in the future.
