1.Gene expression differences between murine intestine intraepithelial lymphocytes and splenic T lymphocytes

Objective: To compare the differences in gene expression between murine intestine intraepithelial lymphocytes (iIELs) and splenic T lymphocytes, establishing the cDNA subtractive library of iIELs, analyzing the iIELs special genes. Methods: The iIELs and splenic T lymphocytes were isolated by density gradient in Percoll and nylon column respectively. cDNA subtractive library of iIELs was established by improved subtractive hybridization. The cDNA was ligated with a different adaptor. After 2 rounds of hybridization and suppression PCR amplification, the PCR products were directly inserted into the plasmid vectors. The ESTs special for iIELs were screened by reverse Northern blot. Results: Fifty ESTs with obvious difference in the cDNA subtractive library of iIELs were screened. These ESTs were squenced and submitted to Genebank. The ESTs were analyzed through BLAST, and 14 genes special for iIELs were obtained. These genes were associated with the pattern recognition of pathogen, cytokine signal transduction and proteinase inhibitors. Conclusion: iIELs are important components of intestine natural immunity, it can recognize the conservative antigens of bacteria and can be affected by cytokines and proteinase inhibitors, leading to unique cellular behavior--more resting and less activation.
2.Optimization of the Formula of Ginkgo Flavone Tablets

OBJECTIVE:To optimize the formula of Ginkgo flavone tablets.METHODS:The effects of dextrin,CMS-Na,microcrystalline cellulose and L-HPC on the quality of Ginkgo flavone tablets were studied.RESULTS:The results showed that the optimum formula was 5% L-HPC Ginkgo flavone.Tablet prepared with this optimum formula was much better in terms of disintegrantion.CONCLUSION:The formula is reasonable and the preparation technic simple.The prepared tablets are suitable for clinical use.
3.The giant mucous cyst of frontoethmomaxillary sinus: one case report.

One case of rare giant mucous cyst of nasal sinus that implicated frontal, ethmoid and maxillary sinus was in our hospital. The patient manifested mainly slowly developing swelling and pain of cheek, exophthalmos and visual impairment. The image studies indicated cyst of nasal sinus, including frontal, ethmoid, and maxillary sinuses. The clinical diagnosis was cyst of frontoethmomaxillary sinus.

4.Differentiation of Candida Species by PCR-SSCP Fingerprinting Analysis

Thirty strains of seven Candida species from CICC(China Center of Industrial Culture Collection)were studied. The strains were differentiated by ITS1 region PCR-SSCP fingerprinting analysis and ITS2 region PCR-SSCP fingerprinting analysis. Results showed that both ITS1 region and ITS2 region were able to differentiate the seven species of Candida clearly. Contrasting the maps and effects on the identification of Candida species of ITS1 region with that of ITS2 region, result indicated that on the identification of Candida species the application of ITS2 region was better than ITS1 region.
5.Expression of gap junction connexin 32 and connexin 43 and inhibition of carbenoxolone in cultured hippocampal neurons of rats after epileptiform discharges

Objective To study the role of gap junctions in epileptiform activity. Methods The epileptiform activity was induced by zero-Mg 2+ medium in cultured hippocampal neurons of newborn rats. Immunocytochemistry and real time RT-PCR were introduced to evaluate the expression of gap junction Cx32 and Cx43. Results The level of Cx32 mRNA increased quickly one hour after the neurons were treated with zero-Mg 2+ medium and was raised by 10 times 5 hours later, while Cx32 protein began to develop at the 2nd hour (21.80?1.74) and was raised by 5 times at the 8th hour (47.30?5.75). The expression of Cx43 mRNA went up obviously 5 hours later, and Cx43 protein developed visibly 8 hours later. Carbenoxolone depressed the expressions of Cx32 and Cx43. Conclusions The expression of Cx32 and Cx43 increases dramatically after epileptic discharges and carbenoxolone inhibits both the discharges and the expression of gap junctions, which indicates that gap junction could contribute to epileptogenesis.
6.Primary culture of hippocampal neurons of new-born rats

Objective To establish a primary culture method of hippocampal neurons of new-born rats,and to observe the morphological characteristics at different developmental and differential stages.Methods The hippocampus was digested and the cells were seeded in a flask.The neurons were then transferred and re-seeded on cover glasses coated with poly-L-lysine.The neurons were identified by polyclonal antibody against neuron specific enolase(NSE).The morphology was observed under phase-contrast microscope.Results A large number of hippocampal neurons began to adhere to the cover glasses after 12~24 hours.They showed different shapes-shuttle,triangle,pyramidal,or nonregular after clinging to the plate.Their processes connected into nets and different in length and thickness.They were well developed on the 7th~10th day and survived as long as 28 days after seeding.Immunocytochemistry of NSE proved high purity.Conclusion The culture method of new-born rat hippocampal neurons in vitro is successful and can be used as an in vitro model of research.

9.The influence of Gardiner bacteria infection on premature birth and changes of endocrine hormone levels

Objective To investigate the effect of Gardiner bacteria infection on preterm delivery and changes of endocrine hor-mone levels.Methods Application of Gardiner bacteria fluorescent marker detection kit in 120 cases of suspected Gardiner bacteria infection pregnant women with immunofluorescence assay,and the levels of serum follicle stimulating hormone (FSH),luteinizing hormone (LH),estradiol (E2),progesterone (P)were measured at the same time,90 cases of non maternal Gardiner bacteria infec-tion were enrolled as control.Results After the treatment,98 cases of pregnant women with Gardiner bacteria infection should ex-perienced premature delivery (24-37 week),accounted for 81.67%,compared with the normal pregnant women were statistically significant (P <0.05);and 22 cases of Gardiner bacteria infection pregnant women after treatment no premature birth,accounted for 18.33%,and to all pregnant women infected Gardiner bacteria and experiencing preterm delivery and the control group levels of endocrine hormone were determined,compared the results of FSH,LH,found no significant differences(P >0.05),E2 increased,P decreased,changed significantly (P <0.05 ).Conclusion The probability of premature delivery in women infected with Gardiner bacteria is significantly higher than that of normal pregnant women,at the same time the secretion of hormone changes,so dealing with pre pregnancy and early pregnancy should carry owt Gardiner bacteria examination.
10.Clinical study of Guillain-Barre syndrome combined with demyelinating lesion in central nervous system

Objective To investigate Guillain Barre syndrome (GBS) combined with demyelinating neuropathy in central nervous system(CNS), and explore the possible mechanism and the relationship between the two.Methods 3 cases GBS combined with demyelinating neuropathy in CNS were observed clinically and the datum of laboratory were analysed.Results Case 1, a 28 year old man had symptoms of general flaccid paralysis and coma.The result of blood gas analysis was normal. CSF showed an albuminocytological dissociation, delayed nerve conduction velocity and missed F waves. Brain MRI showed multifocal T 2 Wight Image high signs in white matter of bilateral brain and cervical spinal cord. The patient is getting recovery by treatment with plasma and immunoglobulin. Case 2 , a 5 year old girl with progressive weakness of her limbs and respiratory arrest, appeared confusion,dully light reflex and absent corneal reflex, at last she died because of rejecting treatment.Case 3,a 12 year old boy with progressive weakness of his limbs and the difficult of relieving the bowels.Brain MRI was normal.Spinal MRI showed multifocal T 2 weight imagine hight signs from T 5 to L 4.CSF showed an albuminocytological dissociation.EMG showed a delayed nerve conduction velocity.Conclusion GBS combined with disorders of consciousness are mostly severe, the pathological mechanism is unclear. It is suggested that auto immunoreaction caused by P 1 myelin basic protein can relate to around and CNS demyelination.
