1.Reversal of Multidrug Resistance in NPC HNE-1(200) Cell Line by Oxymatrine

OBJECTIVE:To study the reversal mechanism of oxymatrine on multidrug resistance in the NPC HNE-1(200) cell line. METHODS: The multidrug resistant HNE-1(200) subline was derived from an HNE-1 cell line after exposure to fractionated X-irradiation. The expression of P-gp was tested by immunocytochemical method and western blot and that of mdr1 mRNA was measured by RT-PCR. RESULTS: After 24-hour treatment with oxymatrine, both the immunocytochemical method and western blot showed a decrease in the expression of P-gp. However, RT-PCR measurement showed that the expression of mdr1 mRNA of HNE-1 and HNE-1(200) was nonsignficant. CONCLUSION: Oxymatrine can reverse the multidrug resistance of NPC HNE-1(200) cell line to some degree by down-regulating the expression of P-gp. However, this medicine did no effect on the expression of mdr1 mRNA, suggesting the up-regulation of the expression of P-gp is under the control of multiple mechanism.
2.Inhibitory Effect of Tea Polyphenols in Combination with Anti-tumor Drugs on Cell Proliferation of the Multiresistant HNE-1 Cell Line

OBJECTIVE: To study the joint action of tea polyphenols and common anti-tumor drugs(vincristine,pingyangmycin,5-Fu,cisplatin) in inhibiting cell proliferation of the multiresistant HNE-1(200) cell line in patients with nasopharyngeal carcinoma after radiographic exposure.METHODS: The low toxic dosage of tea polyphenols and 4 kinds of anti-tumor drugs were optimized by MTT method.Then the inhibition ratio of multiresistant HNE-1(200) cell line treated by tea polyphenols and anti-tumor drugs waere detected.RESULTS: The low toxic dosage of tea polyphenols was 0.50 mg?mL-1.When the anti-tumor drugs were used in combination with tea polyphenols(0.50 mg?mL-1) as compared without,the inhibition ratio on HNE-1(200) cell line was increased by 100%~300%.CONCLUSIONS: Addition of tea polyphenols to anti-tumor drugs showed remarkable inhibitory effect on cell proliferation of HNE-1(200) cell line in patients with nasopharyngeal carcinoma,suggesting the reverse effect of tea polyphenols on resistant HNE-1(200) cell line.
3.Effects of oxymartine on expression of P-glycoprotein in HNE-1 cell line radiation therapy

Objective:To investigate the effects of oxymartine on cell growth and the changes of P-glycoprotein in HNE-1 cell line after exposed to fractionated X-irradiation.Methods:By MTT,the cell proliferation was observed both before X-irradiation and after X-irradiation conditions with the extent of vincristine,oxymartine and verapamil respectively.P-glycoprotein expression was tested by immunohistochemistory method SP.Results:(1)Vincristine,oxymartine and verapamil were capable of inhibiting cell growth on HNE-1 cell line and fractionated X-irradiated HNE-1(200)cell subline;(2)Vincristine was less inhibitory on cell growth in HNE-1(200)cell subline than HNE-1 cell line,and the difference had statistic significance;(3)P-glycoprotein expression in HNE-1 cell line was negative while HNE-1(200)cell subline positive;oxymartine with low inhibitory concentration could increase the expression of P-glycoprotein in HNE-1(200) cell subline,which is similar to that of verapamil.Conclusion:(1)Oxymartine is capable of inhibiting cell growth on HNE-1 cell line and fractionated X-irradiated HNE-1(200)cell subline;(2)HNE-I(200)cell subline is resistant to vincristine;(3)Oxymartine with low inhibitory concentration could increase the expression of P-glycoprotein in HNE-1(200)cell subline.which is similar to that of verapamil.
4.LncRNAs and drug resistance in tumors

Chemotherapy is one of the primary treatments for cancer patients,while tumor multidrug re-sistance( MDR) represents a major obstacle in the treatments of patients,limiting the efficacy of chemotherapy.To date,the mechanism of drug resistance has not been fully elucidated.lncRNAs are a family of long noncoding RNAs that regulate gene expression by epigenetic modification,transcription and post transcriptional processing. Recent studies have showed that the aberrant regulation of target genes by lncRNAs may be involved in the acqui-sition of tumor MDR.Therefore,targeting lncRNAs may be an attractive strategy for improving the chemosentivity and reversing chemotherapeutic resistance.This paper reviews the relationship between lncRNAs and tumor drug resistance as well as the underlying possible mechanism.
5.RNA interference of silent mating type information regulation 2 homolog 1 SIRT1 arrests cell cycle progress of prostate cancer PC3 cells

Objective:To observe the effects of double-stranded small interfering RNA (siRNA) of the silent mating-type infor-mation regulation 2 homolog 1 (SIRT1) on the cell proliferation, cell cycle progression, and expression levels of the cell cycle negative regulators. These regulators include P21, P27, and phosphorylated retinoblastoma (PRb) proteins present in prostate cancer PC3 cells. This work further aims to explore the possible underlying mechanism for such effects. Methods:PC3 cells were cultured in vitro and then randomly divided into the mock group, scramble siRNA transfected group, and SIRT1 siRNA-transfected group. SIRT1 siRNA ef-ficiency was examined through reverse transcription polymerase chain reaction and Western blot analysis. The inhibitory rate of PC3 cell growth was determined through a methyl thiazolyl tetrazolium assay, and the cell cycle was investigated with the use of flow cytom-etry. The P21 and P27 protein expression levels and PRb status were determined by Western blot assay. Results:Compared with those of the mock and scramble siRNA groups, the expression levels of SIRT1 mRNA and protein significantly decreased in SIRT1 siR-NA-transfected cells. In addition, the inhibitory rate of PC3 cell growth was markedly increased, and the cell cycle of the PC3 cells was arrested at the G1 stage. The expression levels of negative cell cycle regulators, including P21 and P27 protein levels increased, whereas Rb protein phosphorylation was inhibited in SIRT1 siRNA-transfected PC3 cells. Conclusion: SIRT1 RNA interference inhibits PC3 cell growth and arrests cell cycle progression through the upregulation of the P21 and P27 proteins and the inhibition of Rb protein phosphorylation.

To investigate the effect of herbal drugs for clearing away heat, removing dampness and killing parasites,which are used for washing on tinea pedis. Two hundred and eight cases confirmed as tinea pedis were randomized to two groups: group A (n=108) and group B (n=100). Group A was treated with Xiaoxuan Lotion (mainly composed of Radix et Rhizoma Rhei, Herba Artemisiae Scopariae, Cortex Phellodendri, Radix Sophorae Flavescentis, Radix Stemonae, etc.) and group B with miconazole nitrate cream. 7.STUDIES ON THE TRICHLOROISOCYANURIC ACID(TCCA)IN THE DISINFECTION OF DRINKING WATER

TCCA containing a high amount of effective chlorine is an organic disinfectant for drinking water. A small amount of it has satisfactory effect with less odour and color, It is stable in storage.The factors which effect disinfection are less.It is suitable for the disinfection of various kinds of water sources. The laboratory experiment and field investigation showed that the rate of killing bacteria in water was 99.99% when its concentration was 1.4-3.4 mg/1.the temperature was 5?to 30℃, the pH value was 5-9, the disinfecting time was 10 to 30 minutes.There was an exception of the experimental group number 7 and 9 which showed the rate of killing bacteria was 99. 94%. The toxicity of TCCA is low, the disinfectant effect is much better when it is used with SM-92 coagulant.
8.Deconvolution Reconstruction of Thermoacoustic Imaging for Small-scale Objects

Objective To increase the speed of thermoacoustic imaging, a deconvolution reconstruction (DR) algorithm is proposed for small-scale need detecting living beings, on the basis of deconvolution of thermoacustic imaging algorithm. Methods A new function was firstly constructed from the detected acoustic pressure function. Then, the distribution of electromagnetic wave absorption coefficient of the need detecting living beings could be reconstructed from this new function based on the deconvolution method. Computer simulation studies were carried out to compare the DR algorithm with the two popular algorithms: the time-domain reconstruction (TDR) and the filtered back projection (FBP). Results For small-scale need detecting living beings, the accuracy of DR was nearly equivalent to TDR and slightly better than FBP. However, DR was about 4 to 6 times faster than TDR and about 25 to 100 times faster than FBP under the simulation experiment condition. Conclusion DR is a fast and effective algorithm of thermoacoustic imaging for small-scale living beings.

However, because outcomes of clinical interventions only can be studied in human subjects, this practice must not only meet scientific criteria. it also must meet strict ethical criteria.
