4.Differentiation of Candida Species by PCR-SSCP Fingerprinting Analysis

Thirty strains of seven Candida species from CICC(China Center of Industrial Culture Collection)were studied. The strains were differentiated by ITS1 region PCR-SSCP fingerprinting analysis and ITS2 region PCR-SSCP fingerprinting analysis. Results showed that both ITS1 region and ITS2 region were able to differentiate the seven species of Candida clearly. Contrasting the maps and effects on the identification of Candida species of ITS1 region with that of ITS2 region, result indicated that on the identification of Candida species the application of ITS2 region was better than ITS1 region.

7.Single nucleotide polymorphism of human APM1 detected by using allelic 3′-terminal specific primers real-time PCR

Objective To develop an analyzing technique of single nucleotide polymorphism(SNP) based on non-probe real-time quantitative PCR, and to explore the relationship between SNP in human adiponectin (APM1) gene and type 2 diabetes (T2DM).Methods Design primers with specific 3′-terminal nucleotides and perform Real-time PCR using SYBR Green dye. Genotypes were determined by comparing the amplification efficiency of each pair of primer and verified by sequencing. SNPs 45 and 276 in APM1 of 20 T2DM, 24 obesity and 28 healthy persons were analyzed. Results This technique could discriminate genotype as efficient as sequencing. 8.Spiral free perforator flaps for completely degloved thumb reconstruction

Objective To determine the effect of resurfacing completely degloved thumbs by free perforator flaps in a spiral fashion.Methods A retrospective case series study was made on 22 patients with completely degloving injury of the thumb treated by different free perforator flaps in a spiral fashion from June 2009 to December 2014,including 19 male and 3 female patients at the age of 18-46 years (mean,27.8 years).A total of 13 patients were reconstructed using the medial arm perforator flaps ranging in size from 8.0 cm × 3.0 cm-14.5 cm × 3.5 cm,and 9 patients were reconstructed using the peroneal artery perforator flaps ranging in size from 6.0 cm × 3.5 cm-21.0 cm × 10.0 cm.Follow-up was made to evaluate the sensation,appearance and function of the thumbs and the recovery of flap donor sites.Results All flaps survived completely,with no occurrence of postoperative vascular crisis and infection.Only one medial arm flap and one soleus perforator flap distal flap showed blood flow disorder in the distal sections and were healed uneventfully after conservative therapy.Function and cosmetic recovery of the reconstructed thumbs were good.Follow-up ranged from 9 to 22 months (mean,14.5 months).Twopoint discrimination test of the flaps was recovered to 6-13 mm.No linear scar contracture was left.Flap donor site wound healed primarily.All patients were satisfied with the overall results.Conclusion The free perforator flap in a spiral fashion can avoid linear scar contracture,maximally decrease donor site morbidity and achieve satisfactory functional recovery and good aesthetic restoration,indicating a good candidate for reconstruction of thumb degloving injury.
9.The effect of an enhanced load sharing dynamic pedicle screw fixation device on the bone mineral density and bone structure of vertebrae

Objective To compare the influence of the dynamic device(DD) of pedicle screw fixation and its equivalent rigid device (RD) on bone mineral density and bone structure of the vertebrae. Methods 24 goats were randomly divided into 4 groups: control group, single posterolateral fusion group, dynamic device with posterolateral fusion group and rigid device with posterolateral fusion group. After 24 weeks postoperative observation, the animals were sacrificed and the fixed lumbar spines were harvested. The bone mineral density (BMD) of these harvested specimens was measured by quantitative computed tomography (QCT). Then the L3 segment was embedded in methyl methacrylate and sectioned after alcoholic dehydration. Undecalcified slides (50 ?m in thickness) were stained with toluidine blue. Quantitative histologic analysis was performed in a semiautomatic way. The static parameters consisted of trabecular bone volume (TBV), the mean trabecular plate thickness (MTPT) and mean trabecular spacing (Tb.Sp). The main dynamic histomorphometric parameter, bone mineralization rate (MAR), was calculated by measuring the mean distance between two flurochrome labelings. 10.Distribution and significance of NGF in cochleae of neonate rats

Objective To investigate the distribution of NGF in the cochleae of neonate rats and to explore the mechanism of NGF in the auditory path. Methods Twenty healthy neonate SD rats aged 5-7 d were selected and their cochlear paraffin sections were labeled with a rabbit anti-NGF polyclonal antibody by SP method. Results The membranous labyrinth of neonate rats appeared similar to triangle and NGF expression was observed in immature corti’s organ, in spiral ganglion cells and in nerve fibers throughout the cochlea. The distribution located in both cytoplasm and nucleus. Conclusion The distribution of NGF was closely related to survival and maturation of spiral ganglion cells and hair cells in rats aged 5-7 d.
