1.Properties of artificial joint materials and exercise-induced articular cartilage injury

BACKGROUND:The artificial joint has gone through nearly a century of history from the beginning of design to the clinical application. The forerunner of artificial joints is artificial hip joint. With the skiled technological application in clinic, the design, application and improvement of wrist, shoulder, knee and other artificial joints have been promoted. OBJECTIVE: To summarize the application progress and performance characteristics of artificial joint materials in exercise-induced articular cartilage injury. METHODS: PubMed (http://www.ncbi.nlm.nih.gov/PubMed), Wanfang database (http://wanfangdata.com.cn/) and China National Knowledge Infrastructure (http://www.cnki.net/) during 1980 to 2015 were retrieved by the first author. “Sports; Cartilage Injury; Artificial Joint” were taken as the key words in English and Chinese, respectively. Retrieved data were analyzed and summarized. The artificial joint materials commonly used at home and abroad were introduced and analyzed one by one. The clinical advantages and disadvantages of various materials were analyzed from the view of materials science. The surface treatment methods commonly used for artificial joints were described. The direction of the artificial joint research was pointed out from a development perspective. RESULTS AND CONCLUSION:The surface modification of titanium aloys can obtain surface ceramic layer with good performance, and can effectively improve the anti-wear properties of titanium aloys. The filing and modification of ultra-high molecular weight polyethylene can obtain joint composites with good anti-wear properties that can effectively reduce the production of ultra-high molecular weight polyethylene wear particles and the biological response induced by ultra-high molecular weight polyethylene wear particles. Further research and improvement of ceramic artificial joints wil be the future direction of development of artificial joints.
2.Expression of MMP-9 and TIMP-1 in lesions of systemic sclerosis and its implications.

In order to investigate the role of MMP-9 and TIMP-1 in the pathogenesis of systemic sclerosis, the expression of MMP-9 and TIMP-1 was immunohistochemically detected in skin lesions of the patients with diffuse cutaneous systemic sclerosis, skin lesions of the patients with limited cutaneous systemic sclerosis, and skin tissues of normal subjects. The results showed that the expression of MMP-9 in lesions of diffuse cutaneous systemic sclerosis was significantly lower than that of normal skins (P<0.05). However, no significant difference in the level of MMP-9 in the limited cutaneous systemic sclerosis and normal skin was found. Meanwhile, the expression of TIMP-1 in lesions of diffuse cutaneous systemic sclerosis and limited cutaneous systemic sclerosis were significantly higher than that of normal skins (both P<0.05). It was suggested that the expression of MMP-9 and TIMP-1 might play an important role in the development of systemic sclerosis.

Objective: To study the effect of XRK on mice's acute inflammation. Methods:The three XRL groups were fed on XRK of high, middle and low dosage, the positive group was fed on mezolin, and the control group was fed on distilled water. All groups were fed for three days, once per day. 30 minutes after the last treatment, Xylene (0.05 ml) was spreaded on mice's right ear in two sides, 15 minutes later, mice were put to death and its two ears were cut in same area, then weighed, calculated the swelling degree of its ears. 5.The effect of sling exercise therapy on the balance and walking ability of persons with incomplete paraplegia

Objective To explore the effect of sling exercise therapy (SET) for improving the balance and walking ability of patients with incomplete paraplegia.Methods The 37 patients were randomly divided into two groups:19 patients in the observation group and 18 cases in the control group.Both groups were given conventional rehabilitation training,including bridge training on the mat,kneeling position training,sit-stand training and orthosis walking training.The observation group was additionally provided with sling exercise therapy with multi-point,muhi axis sling suspension supine,prone with a neutral lumbar spine,supine with pelvic elevation and in a lateral position like an arch.Before the treatment and 6 weeks after the treatment,trunk control tests (TCTs) were administered.The Berg balance scale (BBS) and Holden's walking function classification were used to assess the subjects' balance and walking ability.Results There were no significant differences in the two groups' average TCT and BBS scores before the treatment.After 6 weeks of treatment the average scores of both groups had increased significantly,with a significantly bigger increase observed in the observation group.After the treatment,the average Holden classification of the observation group was significantly superior to that of the control group.Conclusion SET in addition to conventional rehabilitation training can significantly improve the balance and walking ability of patients with incomplete paraplegia.It is worthy of application in clinical practice.
6.Nerve growth factor promotes proliferation of human pancreatic cancer cell line MIA PaCa-2

Objective: To investigate the effect of nerve growth factor-β(NGF-β) on the proliferation and cell cycle of human pancreatic cancer MIA PaCa-2 cells. Methods: MIA PaCa-2 cells were treated with different concentrations of NGF-β and K252a (inhibitor of NGF-β receptor TrKA) alone or in combination. Clone forming rate, proliferation, and cell cycle of MIA PaCa-2 cells treated with different strategies were examined by clone formation assay, MTT, and flow cytometry, respectively. Results: NGF-β significantly increased the clone formation and proliferation of MIA PaCa-2 cells (P<0.05, P<0.01). K252a significantly inhibited the proliferation of MIA PaCa-2 cells (P<0.05), while NGF-β combined with K252a had no significant effect on the proliferation of MIA PaCa-2 cells. NGF-β arrested MIA PaCa-2 cell cycle in S phase, K252a arrested cell cycle in G_0/ G_1 phase, and NGF-β combined with K252a arrested cell cycle in S phase. Conclusion: NGF-β can enhance the proliferation of pancreatic carcinoma MIA PaCa-2 cells.
7.Study on transplantation of induced bone marrow mesenchymal stem cells via various route for the treatment of chronic liver injury

Objective To investigate the efficacy of transplantation of induced bone marrow mesenchymal stem cells(MSCs)via various route for treatment of chronic liver injury.Methods MSCs were isolated and expanded by density gradient centrifugation combined with adherent culture.MSCs were induced to differentiate into hepatocyte-like cells in vitro.The markers of hepatocyte lineage were examined by RT-PCR and immunocytochemistry.The changes of ultramicrostructure of MSCs were observed by transmission electron microscope.One hundred and fourteen Wistar rats were treated with mixture of 40%CCl4 and peanut oil to establish chronic liver injury model and another 6 rats were served as control.Twelve rats in model group were died within 8 weeks,and the rest rats were divided into portal vein,spleen and tail vein transplantation groups.The rats were transplanted with DAPI-labelled MSCs which were induced for 14 days.The distribution of the DAPI-labelled cells were observed by fluorescence microscopy.The liver function and pathology were examined.Results AFP mRNA expression were detected at 7th and 14th day after induction,expressions of ALB mRNA,CK18 and hepatocyte antigen were found at 14th and 2lth day.The ultrastructure examination revealed that MSCs were enlarged with a lot of endoplasmic reticulum,mitochondrion,lysosome and glycogen granule.DAPI labelled cells were found in the spleen and liver transplantion groups.The pathological changes of liver were alleviated in all treated groups,especially in intraspleenic transplantion group(P<0.05).Conclusions Induced bone marrow MSCs can ameliorate liver function of chronic hepatic injury after transplantation.The intraspleenic transplantation is better than others.
8.Observation on therapeutic effects of acupuncture plus Chinese herbs for sudden deafness

Objective:To observe the clinical effect of wind-expelling method of acupuncture and Chinese herbs for sudden deafness.
Methods:A total of 90 cases with sudden deafness were randomly divided into three groups based upon their visit order and random digital table, an acupuncture plus Chinese herbs (APCH) group, an acupuncture group, and a Western medication (WM) group, 30 cases in each group. The APCH group was treated by puncturing Fengchi (GB 20), Fengfu (GV 16) and Fengmen (BL 12), without retaining the needles, and then by puncturing Zhongwan (CV 12), Guanyuan (CV 4), Qihai (CV 6) and Tianshu (ST 25) with reinforcing technique after arrival of the needling sensation, and the needles were retained for 30 min, plus oral administration of wind-expelling Chinese herbs. The acupuncture group was treated with the acupoints selected by syndrome differentiation in reference to the textbook of Acupuncture and Moxibustion Science. The WM group was treated by intravenous infusion of Alprostadil injection, Adenosine Disodium Triphosphate and Coenzyme A and muscular injection of Vitamin B12 and Vitamin B1. The decibel values of hearing loss before and after treatment were respectively recorded and analyzed comparatively.
Results:In the intra-group comparison after treatment among the three groups, the decibel values of hearing loss all declined, with statistically significant differences (allP<0.05). After treatment, the decibel value of hearing loss in the APCH group was better than those in the acupuncture group (P<0.05) and the WM group (P<0.05). The total effective rate was respectively 83.3% in the APCH group, 60.0% in the acupuncture group and 56.7% in the WM group. The total effective rate in the APCH group was better than those in other two groups (bothP<0.05).
Conclusion:The therapeutic effect was better for sudden deafness by wind-expelling method of acupuncture and Chinese herbs than by acupuncture alone or by Western medication.
9.Experience on the research-aimed experiment of undergraduate students

It is meaningful to prompt the research-aimed experiments in the laboratory edu-cation of undergraduate students majoring in pharmacy,which means under the guidance of tutors, students collect information,design the experiment procedure and conduct the whole experiment independently. Under this novel mode of laboratory education, students’ abilities of indepen-dent-thinking and comprehensive-experimental conduction are largely improved. Meanwhile,the sense of team-work is enhanced. To sum up,the research-aimed experiments is significantly ben-eficial to training undergraduate students for future scientific researches.
10.Chronic hepatic injury modeling in mice induced by D-galactosamine and lipopolysaccharide combination

Objective To research the method of Chronic hepatic injury modeling in mice induced by D -galactosamine and lipopolysaccharide combination . Methods Injected D-galactosamine ( 30 mg/mL ) and lipopolysaccharide ( 2μg/mL ) combination by intraperitoneal injection , two days at a time for 8 weeks .Monitored variation of diet and weight; detected serum level of alanine aminotransferase ( ALT ) and aspartate aminotransferase (AST), been put to death in mice and removed the liver tissue .strained hepatic tissue by the HE and Masoon dye to observe Liver tissue structure and cellular morphology and the degree of fibrosis .Results Lipopolysaccharide and D-galactosamine combination resulted in ALT rise , hepatocyte degeneration and necrosis ,collagen fiber hyperplasia obviously . Conclusion D-galactosamine and Lipopolysaccharide combination could induce mice chronic hepatic injury modeling .
