1.Comparison of Her-2 status in primary gastric cancer with the corresponding endoscopic biopsies and lymph node metastases
Chinese Journal of Clinical Oncology 2014;(3):184-188
Objective:This study aimed to validate the accuracy and validity of Her-2 assessment used for endoscopic biopsies and lymph node metastases by comparing these samples with the surgical materials obtained from the same patients. This study also aimed to analyze the relationship between primary tumor and clinicopathological characteristics of gastric cancer. Methods:Her-2 sta-tus was evaluated according to Gastric cancer Her-2 testing guidelines in 107 cases of gastric cancers with coupled endoscopic biopsies and surgical materials and in 76 cases of the corresponding lymph node metastases. Results were then compared and analyzed. Re-sults:The complete concordance of the Her-2 status of endoscopic biopsies and surgical samples (n=107) was noted in 86.9%of the cases. This result is consistent between the two different samples (Z=6.381 3, P<0.000 1). Primary gastric cancer and the corresponding lymph node metastases also displayed uniformity (Z=3.027 4, P=0.002 5). The positive rate of Her-2 was significantly associated with Lauren type, tumor histological grade, depth of invasion, lymph node metastasis, and TNM stage. Conclusion:A high concordance was observed between primary tumor and biopsy specimen (endoscopic biopsies and lymph node metastases), in which Her-2 status exhibit-ed a high predictive value. Her-2 status is correlated with invasion and cancer metastasis. Hence, the most appropriate criteria for Her-2 status assessment in lymph nodes should be considered to improve the Her-2 diagnosis system of gastric cancer.

Objective To evaluate the efficacy of Qingfei-Jiedu decoction in the treatment of Acne. Methods A total of 93 female patients with acne were randomly divided into 3 groups according to random number table, 31 in each group. The observation group took the oral Qingfei-Jiedu decoction, and the Chinese medicine control group took the oral tanshinone capsule, and the western medicine control group took the oral acetic acid ring propyl alcohol progesterone. All 3 groups were treated for 4 weeks. The levels of serum dihydro (DHT), IL-6 and IL-8 were measured by ELISA. The changes of the lesions before and after treatment were observed and the adverse events were recorded during the treatment. The clinical effect was evaluated. Results After treatment, the serum DHT (56.94 ± 15.74 pg/ml vs. 74.48 ± 18.53 pg/ml, t=2.089), IL-6 (22.84 ± 11.77 pg/ml vs. 30.37 ± 14.50 pg/ml, t=2.135), IL-8 (22.64 ± 7.38 pg/ml vs. 29.54 ± 9.65 pg/ml, t=2.057) in the observation group were significantly lower than those in the Chinese medicine control group (P<0.05). After treatment, the disappearance rate of skin lesions (65.4% ± 15.0% vs. 44.7% ± 12.9%, 42.7% ± 13.6%, F=6.862) and comprehensive syndrome effective rate (57.2% ± 12.6% vs. 46.3% ± 12.8%, 44.8% ± 11.7%, F=5.322) in the observation group were significantly higher than those in the other two control groups (P<0.05). The observation group and the traditional Chinese medicine control group did not have adverse events, but the western medicine control group showed one case nausea. Conclusions The Qingfei-Jiedu decoction can reduce the level of serum DHT, IL-6 and IL-8 in female patients with acne vulgaris, and improve the skin lesion disappearance rates and comprehensive syndrome rates.

Objective To evaluate the effect of hydrogen on the postoperative cognitive function in aged mice. Methods Sixty aged male Kunming mice, aged 12-14 months, weighing 35-45 g,were assigned into 3 groups (n=20 each) using a random number table:sham operation group (group Sham),partial hepatectomy group (group PH) and partial hepatectomy plus hydrogen-enriched saline group (group PH-H).On preoperative day 3,during operation and on postoperative day 3,hydrogen-enriched saline 10 ml/kg was intraperitoneally injected once a day in group PH-H,and the equal volume of normal saline was intraperitoneally injected once a day in S and PH groups. The spatial probe test was performed on postoperative day 7 to evaluate the cognitive function.Ten mice were selected on postoperative days 3 and 7 (after the end of the spatial probe test),and blood samples were collected for determination of serum tumor necrosis factor-alpha (TNF-α),interleukin-lbeta (IL-1β) and high-mobility group box 1 protein (HMGB1) concentrations by enzyme-linked immunosorbent assay. The animals were sacrificed after blood sampling, brains were removed and hippocampi were isolated for detection of the expression of TNF-α,IL-1β and HMGB1 by Western blot. Results Compared with group S,the percentage of time of staying at the target quadrant and the number of crossing the platform within 1 min were significantly decreased, the serum concentrations of TNF-α,IL-1β and HMGB1 were increased, and the expression of hippocampal TNF-α,IL-1β and HMGB1 was up-regulated in group PH (P<0.05),and no significant change was found in the variables mentioned above in group PH-H (P>0.05).Compared with group PH,the percentage of time of staying at the target quadrant and the number of crossing the platform within 1 min were significantly increased, the serum concentrations of TNF-α,IL-1β and HMGB1 were decreased, and the expression of hippocampal TNF-α,IL-1β and HMGB1 was down-regulated in group PH-H (P<0.05).Conclusion Hydrogen can improve the postoperative cognitive function in aged inice.

Mild cognitive impairment is a transitional state between normal aging and dementia, which is the best intervention window to delay cognitive decline. As one of the adjustable risk factors affecting cognitive function, perceived stress significantly increases the risk of cognitive impairment by causing hypothalamic-pituitary-adrenal axis dysfunction and psychological disorders. Therefore, perceived stress management will become a new hot spot in the research and practice of non-drug intervention techniques for cognitive impairment. This paper reviews the current situation of perceived stress in MCI population, the relationship between perceived stress and cognitive function, and the evaluation techniques of MCI individual perceived stress.Further more, this paper summarizes the relevant influencing factors of perceived stress based on stress attribution model, and comments on the action mechanism and practical enlightenment of perceived stress intervention program on cognitive function management, in order to provide theoretical reference for cognitive function management of MCI population.

Nicotinamide adenine dinucleotide phosphate oxidase ( NOXs) contributes to the production of reactive oxygen species ( ROS) in liver fibrosis, resulting in the activation of endoplas-mic reticulum stress ( ERS ) and IRE1α-XBP1 signaling path-way. ROS is a series of oxygen metabolites and its derivatives, produced by the single electron reduction of molecular oxygen ( O2 ) , including superoxide anion ( O2- ) , hydroxyl radical (-OH) , hydrogen peroxide ( H2 O2 ) , hypochlorite ion ( OCl-) and so on. They can interact with a large number of molecules, including small inorganic molecules, proteins, lipids, carbohy-drates and nucleic acids, resulting in lipid peroxidation of cell damaging molecules. And as a second messenger, ROS can also affect the proliferation and activation of HSC in liver fibrosis, and induce the hepatocyte apoptosis through a variety of cellular signal transduction. Here we review the current status of the study on the mechanism of NOXs in liver fibrosis.
