1.Combinative application CD4+ CD25+ regulatory T cells test and irregular antibody screening in the diagnosis of invalid clinical transfusion

Objective To investigate whether the T cells (CD4+ CD25+) levels and the irregular antibodies screening cloud improve the diagnosis of invalid red blood cells transfusion.Methods Thirty-one patients with red blood cell invalid transfusion in the People's Hospital of Yubei District of Chongqing were selected.Flow cytometry was used to detect the changes of CD4+CD25+ regulatory T cells.Standard cells Ⅰ,Ⅱ,Ⅲ were used to screen on irregular antibodies in red blood cells (RBC).Results The rate of invalid RBC transfusion was 7.52% (31/412) in Yubei District.The incidence rate of medical diseases was 77.42% (24/31),much higher than surgical disease (22.58% (7/31,P =0.002),and the major departments were oncology department,hematological department and infectious department.The CD4+ CD25+ regulatory T cells were decrease from (22.18±1.58) % to (16.57±1.77) %(P=0.023).Conclusion Combin CD4+CD25+ regulatory T cells test and irregular antibody screening can help prevent and reduce the invalid transfusionof red blood cells.

Objective To investigate the effects of myricetin (Myr) on A549 lung cancer cell proliferation and its mechanism. Method The inhibitory effects of Myr on the growth of A549 cells were investigated and its value of IC50 after 72 h incubation was measured by 3-(4,5-dimethyl-thiazolyl -2)-2,5-diphenyl tetrazolium bromide (MTT) method. The phosphorylation level of Akt protein was studied by Western blotting. Results Myr inbibited A549 cell proliferation in dose-dependent way. The IC50 value of Myr treatment for 72 h was 41.7 ?g/ml. Treatments with 32 ?g/ml Myr for 12 h significantly reduced the protein kinase B (Akt Ser473) phosphorylation. Conclusion Myr could inhibit the growth of A549 cell in a dose-dependent way. Akt phosphorylation may be the mainly molecular target of Myr in A549.

5.CT evaluation of the epibulbar dermoid and dermolipoma

Objective To investigate the CT imaging features and its diagnostic value in patients with epibulbar dermoid and dermolipoma. Methods Twelve cases with epibulbar dermoids and dermolipomas were retrospectively analyzed. There were 5 dermoids and 7 dermolipomas, and a distinct female predominance (11/12). The lesions located at the external canthus, and were almost always uniocular and single (right=9, left=3). Transverse plain CT scan was performed in all patients, contrast enhanced CT scan in 5 cases. The clinical aspects, pathological features, and CT findings were then described. Results Crescent-shaped hypodensity lesion attached to the epibulbar was detected in all cases on CT scan. The diameter ranged from 8 mm?5 mm to 25 mm?15 mm. The lesion had a thin wall (1- 3 mm) under the conjunctiva lateral, and could have middle enhancement. Conclusion CT can show the lesion′s range and characteristics, as well as the relationship with the adjacent structures. CT has great directive value for operating on these tumors, and reducing the complications.
6.Logistic regression study on chronic pancreatitis grade diagnostic model

Objective To establish a MRI grading diagnostic model for chronic pancreatitis (CP) to acquire better combination for CP diagnosis and improve the diagnostic ability of CP grading.Methods To collecte the data of 68 CP patients who were clinically diagnosed and 23 health volunteers from Dec 2012 to Oct 2014.There were 23 mild CP, 14 moderate CP and 31 severe CP according to Cambridge classification.3.0 T MRI data were analyzed, and 14 features were extracted to compare the MRI features among groups.The single ordinal regression analysis was conducted on the variables with significant difference between groups, and the collinearity was diagnosed.The factor analysis was used for multicollinearity.The multiple ordinal logistic regression was finally conducted to establish the regression model.Results There was no significant difference between control groups and CP groups on pancreas divisum (X14), but significant differences were found in other 13 features (P<0.05).Single ordinal regression analysis of 13 features showed that all features except pancreatic parenchymal bubble (X12) were significantly correlated with CP grading diagnosis(P<0.05).The effect of multicollinearity was validated among 9 continuous variables.Three common factors were identified, including F1(X3、X4、X5、X9、X8), F2(X7、X6) and F3 (X1、X2) which represented the exocrine function, the features of main pancreatic ducts and pancreatic parenchyma, respectively.Six features were implemented into the multi Logistic regression model, which included F1, F2, F3, X10 (the visualizations of branch pancreatic duct after secretin stimulation), X11 (pancreatic shape) and X13 (the filling defects of main pancreatic duct).Finally, the most appropriate regression model was gotten, which was the scale model of the probit link function.The model′s diagnostic accuracy for normal, mild CP, moderate CP, severe CP and total CP was 96.65%,100%, 71.42%,100% and 94.50%, respectively.Conclusions The ordinal logistic regression model proposed in this study may accurately predict the CP grades and can offer valuable references for clinic diagnosis and therapy of CP.

8.Construction of ply gene-deletion mutant of Streptococcus pneumoniae and research of its virulence change

Objective To lay the foundation for further exploration on parasitifer's defence reaction to pneumolysin through constructing ply gene-deletion strain of Streptococcus pneumoniae and researching on its virulence change. Methods A linker fragment with erm gene in middle and homologous upstream and downstream fragment of ply gene at both sides was prepared by long flanking homology-polymerase chain reaction(LFH-PCR). The linker fragment was transformed into Streptococcus pneumoniae. ply-deficient strain was then screened out from blood plate which contains erythromycin and identified by PCR. ply-deficient strain growth in vitro was observed and virulence change was observed through infecting mouse model. Results PCR results showed that ply gene was replaced completely by erm gene. The ply deficient strain was successfully constructed. The growth of single strain culture medium showed that ply genetic defect made no influence on bacterial's external growth. While in the mice nasal cavity infecting experiment, deficient strain enter into blood after 6 h from infecting which obviously slower than that did wild-type(2 h). And the number of bacteria at each point was much smaller than that of wild-type(P <0. 01 ). The mice peritonaeum infecting experiment showed that median lethal time of wild-type was 3 d, while that of deficient strain was 18 d(P<0. 01). Conclusion It is a good way to completely substitute ply gene using LFH-PCR. ply deletion made no influence on baterial's growth in vitro, but it resulting in reduction of bacterial virulence in vivo.
9.Valuing the role of visual environment defocus characteristics in myopia prevention and control for children and adolescents

Abstract
Myopia is a complex disease influenced by multiple factors, and its prevention and control require a thorough identification and monitoring of its risk factors. The paper examines the defocus characteristics of the visual environment and their association with myopia among children and adolescents, and further describes their role in the prevention and control of myopia, as well as analyses the limitations of the current study and proposes directions for further research. Finally, the paper calls attention to the etiological significance of the defocus characteristics in the visual environment during the development of myopia among children and adolescents, which could provide a scientific basis for optimizing the learning environment and designing other measures to promote myopia prevention and control among children and adolescents.
10.Association between sleep quality and dry eye symptoms among adolescents

Objective:
To explore the association between sleep quality and dry eye symptoms in adolescents,so as to provide the evidence for reducing the prevalence of dry eye symptoms.
Methods:
The study population was adolescents aged 12-24 years from the Psychology and Behavior Investigation of Chinese Residents (PBICR) survey, which was conducted from 20 June to 31 August 2022. A stratified random sampling and quota sampling method was used to select 6 456 adolescents within mainland China. The Ocular Surface Disease Index (OSDI) and Brief version of the Pittsburgh Sleep Quality Index (B-PSQI) were used to assess dry eye symptoms and sleep quality. Multiple Logistic regression model was used to explore the relationship between sleep quality and dry eye symptoms in adolescents. The influence of gender on the association was explored by using interaction terms.
Results:
A total of 2 815 adolescents reported having dry eye symptoms, with a prevalence of 43.6%. Logistic regression analysis results showed an increased risk of exacerbation of dry eye symptoms in adolescents with poor sleep quality. The OR (95% CI ) for mild, moderate, and severe dry eye symptoms groups were 1.39(1.16-1.67), 1.52(1.28-1.81), and 2.35(2.02-2.72), respectively, compared with the ocularly normal group ( P <0.05). There was a significant interaction between sleep quality and gender on dry eye symptoms in adolescents ( P <0.01).
Conclusions
Sleep quality is associated with dry eye symptoms in adolescents, and those with poor sleep quality have a higher risk of dry eye symptoms. The effect of sleep quality on dry eye symptoms is greater in boys.
