2.Detection of Lead Ions in Water Based on Surface Energy Transfer between Gold Nanoparticles and Fluorescenct Dyes
Chinese Journal of Analytical Chemistry 2014;(8):1196-1200
We established a sensitive method for the detection of lead ions in aqueous solution based on the surface energy transfer between gold nanoparticles and fluorescent dyes. The fluorescein-modified thrombin aptamer (5′-FAM-GGT TGG TGT GGT TGG-3′) can be selectively transformed to G-quadruplex induced by lead ions. The conformation changes of the aptamer can alter the distance between the energy donor of fluorescent dyes and the energy receptor of gold nanoparticles, and further enhance the fluorescence intensity. The fluorescence recovery efficiency ( F/F0 ) is proportional to the concentration of lead ions in the range of 12. 5-100 nmol/L following the linear regression equation of y=0. 910﹢0. 007c(R2=0. 997)and the limit of determination is about 10 nmol/L. The proposed method has been applied to the determination of lead ions in tap water with satisfied results.
3.Spectrophotometric detection of diethylstilbestrol on the basis of the plasmon resonance absorption of silver nanoparticles
Journal of Pharmaceutical Analysis 2010;22(3):163-167
In this study, a spectrophotometric detection method for diethylstilbestrol (DES) was proposed by reducing silver nitrate (AgNO3) to obtain silver nanoparticles (AgNPs) in the medium of ammonia and sodium hydroxide. It was found that the resulting AgNPs have plasmon resonance absorption (PRA) characteristic at 415 nm, and the PRA is proportional to the increase of DES concentration in the range of 4.0×10-8-1.0×10-5M with the detection limit (3σ) of 1.2×10-7M. Most of the coexisting substances at high concentrations did not affect the detection of real samples, such as tablets. The recovery was in the range of 96.01%-107.41% and the RSD was lower than 4.7%. This method can be successfully applied to control preparation quality of DES.

Objective:To investigate the correlation between the expression of Golgi phosphoprotein 3 (GOLPH3) and the occurrence of apoptosis in colorectal cancer cells (CRC). Methods:Immunohistochemical assays of GOLPH3 and caspase-3 were performed on the paraffin-embedded sections of 62 CRC samples using the standard streptavidin-peroxidase technique. The apoptotic index of the CRCs was examined using the in situ terminal deoxynucleotidyl transferase nick-end labeling technique. The relationship of the GOLPH3 expression, the cell apoptosis, and the clinicopathologic parameters was analyzed. Results:The positive rates of GOLPH3 expression were significantly higher in the CRC tissues (53.2%) than in the normal colorectal mucosa (37.2%;P<0.05). Likewise, GOLPH3 expression was significantly higher in poorly differentiated cancer tissue, tissue outside the serous membrane, metastatic lymph node tissue, and the stage III CRCs, as compared with those of the moderately to well differentiated tissue, tissue inside the serous membrane, lymph node tissue without metastasis, and the stage I to II CRCs (P<0.05). However, GOLPH3 expression was not significantly correlated with the other clinicopathologic pa-rameters, namely, the age and sex of the patients as well as the site, depth, and length of the invasive tumor (P>0.05). The caspase-3 expression and apoptotic index were significantly lower in the GOLPH3-positive CRC tissue than in the GOLPH3-negative tissue (P<0.05). GOLPH3 expression was negatively correlated with the apoptotic index of CRCs based on the Spearman correlation (r=-0.320, P<0.05). Conclusion:GOLPH3 overexpression in CRC tissue is negatively correlated with apoptotic index.

OBJECTIVE To investigate the distribution features and clinical significance of allergens in patients with chronic rhinosinusitis(CRS). METHODS A retrospective analysis was performed in 136 cases of CRS. Allergen features were compared in the two subtypes of CRS: chronic rhinosinusitis with nasal polyps(CRSwNP) and chronic rhinosinusitis without nasal polyps(CRSsNP). Differences of allergens features of CRS were analyzed, and they were compared to the allergens of 36 patients with allergic rhinitis(AR). RESULTS The total allergen positive rate in CRS was 48.5%. The allergen positive rate, distribution proportion of inhaled and food allergens, distribution proportion of allergens subsets, distribution proportion of single and multiple allergens between two subtypes of CRS had no significant difference(P >0.05). The major allergens of CRS were single inhaled allergens(84.8%). Some CRS patients were allergic to food allergen(9.1%). The minority allergens of CRS were a mixture of inhaled and food allergens(6.1%). Compared to AR, the distribution proportion of inhaled and food allergens(χ2=14.801, P =0.001), the distribution proportion of allergens subsets(χ2=12.951, P=0.005), and the distribution proportion of single and multiple allergens(χ2=9.067, P=0.003) had significant difference. CONCLUSION The allergen positive rate of CRS is much higher than the prevalence of allergic diseases in general population, suggesting that allergic factors may be closely correlated to the pathogenesis of CRS. The clinical features of allergens are similar in the two subtypes of CRS, while there are significant differences in allergen distribution between CRS and AR patients. The detection of allergens may be helpful in prevention and treatment of CRS.

Objective To investigate the relationship between the channel design of tandem-andovoid (T&O) applicator and the doses to organs at risk (OARs) and target volume in three-dimensional brachytherapy for advanced cervical carcinoma. Methods The data on 15 patients with advanced cervical carcinoma treated with three-dimensional brachytherapy in our hospital from 2015 to 2016 were collected, and 30 randomly selected high-dose-rate titanium T&O plans were retrospectively studied.CT-guided,conformal brachytherapy plans were generated. To simulate T&O applicator, the tandem applicator was virtually compared with the T&O plans with the target volume and OARs remaining unchanged. The DVH parameters of the rectum, bladder, and sigmoid were compared using the paired t test. Results For T&O plans and tandem applicator plans, the mean D2cc of the rectum was 387.8±96.8 cGy and 340.8±88.1 cGy,respectively;the mean D2cc of the bladder was 443.2± 87.5 cGy and 719.4± 243.0 cGy,respectively;the mean D2cc of the sigmoid was 330.3±88.8 cGy and 383.1±105.6 cGy,respectively. In the T&O plans, the doses to the rectum, bladder, and sigmoid were within the limits (rectum:D2cc ≤ 500 cGy;bladder:D2cc ≤ 550 cGy;sigmoid:D2c ≤ 500 cGy),while D2cc of the bladder and sigmoid was higher or partially higher than the limits.T&O plans showed a significant reduction in bladder D2cc and sigmoid D2cc compared with the tandem applicator plans (all P<0.05).Conclusions Compared with tandem applicator plans, plans using T&O applicator provide significant sparing of bladder and sigmoid tissues in three-dimensional brachytherapy for cervical carcinoma, but the toxicities require further investigation.

Heteroatom-doped carbon dots (CDs) have attracted extensive interest because of their improved elec-tronic and fluorescence properties with heteroatom doping. In this study, a new synthetic method for nitrogen (N) and sulfur (S) -doped CDs was developed via a hydrothermal method using methionine and citric acid as raw materials. The as-prepared CDs exhibit excellent optical properties and good bio-compatibility. The spherical N,S-doped CDs have an average diameter of 5 nm. They consist of C, O, N and S, and take on excellent water solubility due to the hydroxyl and carboxyl, amino groups on the surface. The CDs have a photoluminescence quantum yield of 13.8% using quinine sulfate as a reference; the average fluorescence lifetime of the CDs was 3.67 ns. The CDs solution present good photoluminescence properties, and the maximum excitation wavelength and emission wavelength locate at 330 nm and 405 nm, respectively. In addition, their fluorescence intensity almost does not change under the condi-tions of acid, alkali, and high salt, which indicated their anti-photobleaching property and good light stability. Based on the good biocompatibility and strong fluorescence emission of the CDs, they can be used as fluorescent imaging reagents.
