2.Effects of Printing and Dyeing Wastewater Before and After Bacterial biotechnological Treatment on Specific Immune Response of Carassius auratus

In order to explore the effects of printing and dyeing wastewater untreated and treated by bacterial biotechnological method on the specific immune response of Carassius auratus, the tested fish were preimmunized intraperitoneally with Aeromonas hydrophila vaccine then were separately placed into control clean water,untreated printing and dyeing wastewater (dilution ratio 13%,pH=7) and treated water 5 fish of every group were killed on the day 2,4,6,8,10,14,18,25,35 th to detect their immune indexes The results indicated that total leucocytic numbers of blood and serum lysozyme level in fish of clean water and treated water increased rapidly and reached their peaks on the day 8th and 6th respectively after vaccination,then reduced to baseline and maintatined for a period of time Leucocytic numbers in fish in untreated wastewater group decreased on day 2nd then increased gradully to normal,serum lysozyme level in this group had decreasing tendency and was lower than the other 2 groups from beginning to end However titer of anti A hydrophila antibody in serum of untreated water group was only lower than that of treated water group on the day 6th when it appeared After this antibody titers of these 3 groups had no significant defference It indicated that printing and dyeing wastewater inhibited immune response of Carassius auratus ,but water treated by bacterial biotechnological method had no this kind of toxic effect
3.Effect of preoperative glucose infusion on glucose transport-4 mRNA expression in skeletal muscle in patients undergoing thoracic surgery

Objective The purpose of this study was to examine the effect of preoperative glucose infusion on glucose transport-4 mRNA ( Glut-4 mRNA) expression in skeletal muscle before and after thoracic operation. Methods Twelve male ASA Ⅰ - Ⅱ patients undergoing elective radical surgery for esophagus cancer were randomly divided into 2 groups : (Ⅰ ) control group; (Ⅱ) glucose infusion group. Patients with hypertension, hyperlipidemia or diabetes mellitus were excluded. The patients in both groups were premedicated with phenobarbitol and atropine. Anesthesia was induced with midazolam 0.1 mg ? kg-1 , propofol 1.5 mg ?kg-1 , fentanyl 4?g?kg-1 and vecuronium 0.15 mg?kg-1 . The patients were mechanically ventilated after trachea! intubatioa Anesthesia was maintained with continuous iv infusion of propofol (80?g ? kg-1? min-1 ) and vecuronium (1.0 ?g?kg-1?min-1) supplemented with droperidol 0.1 mg?kg-1 and fentanyl 6 ?g ?kg-1 given in several boluses. In group Ⅱ 5 % glucose (0.25 g?kg-1) was infused 2 h before operation. Blood samples and 1 g of skeletal muscle were obtained while chest was being opened and closed for determination of plasma glucose and insulin levels and Glut-4 mRNA expression in muscle. The total RNA of the muscle cells was extracted by trizol one-step template. The Glut-4 mRNA amplification products were determined using RT-PCR with ?-action mRNA as an internal control. Objective:In order to prevent misdiagnosis and degrade death rate, we explored the early diagnosis and correct treatment methods of traumatic pseudoaneurysm of internal carotid artery in sphenoid sinus,. Method: The clinic data of 6 cases who had traumatic pseudoaneurysm of internal carotid artery in sphenoid sinus and were admited in our hospital were analyzed retrospectively. Large numbers of literature about the disease were re-viewed. Some early diagnosis methods and optimal treatment schemes were proposed. Result: All patients had a history of cranium trauma and recurrent attacks nasal hemorrhage. There are two patients with a complication of sight loss in single eye. In the six cases, one case was treated with unilateral common carotid artery ligation, three cases were treated with aneurysm and internal carotid artery embolism by using sacculus proprius which is able to shedding , one case was treated with internal carotid artery embolism by using tiny circlip ring . These five pa-tients were crued with no severe complication. One patient death of hemorrhea. His nasal cavity was tamponaded repeatly because repeat nasal hemorrhage. However, he had never treated with internal carotid artery embolism. Conclusion:The patients who have traumatic pseudoaneurysm of internal carotid artery usually die of unmanageable hemorrhea. The patients may be cured if they could obtain an early diagnosis and correct treatment.
5.Polymerase chain reaction analysis of the clonality of Porphyromonas gingivalis and collagenase gene.

OBJECTIVETo investigate the genotypic characterization of Porphyromonas gingivalis (Pg) and the heterogeneity of a potential virulence factor-PrtC.
METHODSArbitrarily primed polymerase chain reaction (AP-PCR) was applied to 80 Pg strains isolated from 24 unrelated Chinese periodontitis patients. PCR reaction was used to detect a fragment of the collagenase gene (PrtC gene). To evaluate the sequence heterogeneity of the Pg PrtC genes, sequence analysis of four PrtC gene of clinical isolates was performed.
RESULTSRandom primer OPA-05 and OPA-17 distinguished 7 AP-PCR profiles (I through VII). The majority of the strains belonged to type VII which accounted for 25.8%. A 548bp fragment of PrtC gene was detected from 24 clinical strains. The PCR products were verified by the restriction endonucleases PstI and PvuI. Sequence analysis showed 4 PrtC genes were heterogeneous in their nucleotide composition and differed from reference strain Pg 53977.
CONCLUSIONSThe results demonstrated genetic diversity existed among these clinical strains isolated from Chinese periodontitis patients and the PrtC genes are heterogeneous in their nucleotide sequence.

7.The expression of COX-2 mRNA and protein in human laryngeal squamous cell carcinoma.

OBJECTIVE:
To investigate the expression of COX-2 in both molecule and protein levels in laryngeal squamous cell carcinoma and the relationship between its intensity and histological grade, clinical stage and lymphoma metastasis.
METHOD:
By using emi-quantify RT-PCR and Immunohistochemistry, the expression of the COX-2 mRNA and COX-2 protein in laryngeal squamous cell carcinoma and polyp of vocal cord were examined.
RESULT:
Compared with the polyp of vocal cord tissue, the expression level of COX-2 mRNA and COX-2 protein in human laryngeal squamous cell carcinoma were significantly increased (P < 0.01). The intensity of COX-2 mRNA and protein expression increased as the laryngeal squamous cell carcinoma progresses developed.
CONCLUSION
COX-2 is high expressed in human laryngeal squamous cell carcinoma, and it may play an significant role in the growth, invasion and metastasis of human laryngeal squamous cell carcinoma. A combination of emi-quantify RT-PCR and immunohistochemistry is sensitive and specific for laryngeal neoplasms detection.

8.Expression of Th1/th2 transcription factors and cytokines in laryngeal squamous cell carcinoma and its clinical significance.

OBJECTIVE:
To study the differentiation of Th1/Th2 on the levels of transcription factors and cytokines production in patients with laryngeal squamous cell carcinoma in peripheral blood mononuclear cells (PBMC). In addition, the relation of p53 and T-bet, GATA3 expression was discussed for understanding the role of T-bet and GATA3 in metastasis.
METHOD:
The gene expression of Th1/Th2 type transcription factors T-bet, GATA3 and cytokines IFN-gamma, IL-4 were determined by RT-PCR and realtime RT-PCR from 49 patients with laryngeal squamous cell carcinoma and 30 normal controls. The expression of p53 was analyzed by immunohistochemistry.
RESULT:
Predominant expression of Th2 type cytokines was detected in 49 laryngeal squamous cell carcinoma patients. The expression rates of T-bet, GATA3, IFN-gamma and IL-4 was 42.86% (21/49), 71.43% (35/49), 26.53% (13/49), 63.27% (33/49) respectively. The expression rates of T-bet and IFN-gamma in patients were lower than in control group (P < 0.05), but the results of GATA3 and IL-4 were opposite (P < 0.05). The similar results were obtained by realtime PCR. The expression of p53 in patients was accompanied with lower expression of T-betmRNA and higher expression of GATA3mRNA.
CONCLUSION
There is predominant expression of Th2 type transcription factors and cytokines in PBMC of laryngeal squamous cell carcinoma patients. T-bet and GATA3 can be used as reference indicators for the metastasis of laryngeal squamous cell carcinoma.

9.Effects of probiotics on gastrointestinal function, nutrition status and inflammatory reaction in critically ill patients with enteral nutrition support

Objective To explore the effects of probiotics on intestinal function, nutritional status and inflammatory response in critically ill patients with enteral nutrition (EN) support. Methods A total of 90 critically ill patients admitted to Intensive Care Unit (ICU) of the First Affiliated Hospital of Anhui Medical University from July 2016 to November 2018 were enrolled, and they were divided into a probiotics combined with EN group (27 cases) and an simple EN group (63 cases) according to random number table method. The patients in the simple EN group were treated with conventional nutrient preparations such as fresubin or fresubin energy fibre; the patients in probiotics combined with EN group were supplemented with probiotics on the basis of conventional EN support for consecutive 7 days in both groups. The changes of gastrointestinal function, nutritional index and inflammatory response index after treatment were observed in both groups. Results After treatment, the subjective global assessment (SGA) method was used to identify the gradation of the patients, it was shown that the proportion of SGA-B grade patients in simple EN group had an upward trend; while the proportion of SGA-B grade patients in probiotics combined with EN group had no significant change; and there was no significant difference in the proportion of SGA-B patients between simple EN group and probiotics combined with EN group after treatment [20.6% (13/63) vs. 7.4% (2/27),P > 0.05]. Compared with those before treatment, the levels of hemoglobin (Hb), white blood cell count (WBC), C-reactive protein (CRP), tumor necrosis factor-α (TNF-α) and interleukins (IL-6, IL-12) in both groups were significantly decreased, while the levels of albumin (Alb), pre-albumin (PA), total lymphocyte count (TLC) in both groups were increased after treatment, and the changes of Hb, TNF-α, IL-6 in probiotics combined with EN group were more significant than those in the simple EN group [Hb (g/L): 95.0 (78.0, 107.0) vs. 93.0 (80.0, 107.0), TNF-α (pg/L): 21.2±4.0 vs. 28.0±5.7, IL-6 (pg/L): 161.3±37.6 vs. 186.2±51.8];the differences in levels of Hb, CRP, TNF-α before and after treatment between the probiotics combined with EN group and simple EN group were statistically significant [Hb (g/L): 1.0 (-4.0, 12.0) vs. 11.0 (1.0, 20.0), CRP (mg/L): 44.3 (13.7, 57.7) vs. 7.5 (-20.1, 62.4), TNF-α (pg/L): 13.3±6.3 vs. 7.9±5.5, all P < 0.05]. There were no statistical significant differences in the other indicators between the two groups (all P > 0.05). Conclusion Probiotics can improve the gastrointestinal function and inflammatory status of critically ill patients with EN support, regretfully, in a short term, the improvement of nutritional status of such patients is not obvious, but probiotics has certain significance in preventing the risk of aggravation of malnutrition and reduction of Hb level.
10.Review on research and application of chloral hydrate preparations

Chloral hydrate is a safe and effective sedative hypnotic medicine. Patients can usually calm down or fall asleep quickly after taking it. It has great clinical value and practical needs in children ’s sedation. However ,the nature of chloral hydrate itself and various factors such as ambient temperature and light affect its stability of the preparation ,most of chloral hydrate preparations are still used in clinic in the form of hospital preparations. Therefore ,developing chloral hydrate preparation with single dose ,long validity period and automatic mass production is the prospect and goal of its subsequent development. Based on it,this study collects and reviews the relevant literatures on various preparations of chloral hydrate and their clinical application by searching the Chinese and English databases.
