1.NKG2D and its ligands in tumor immunotherapy

Natural killer group 2,member D (NKG2D) is a C-typelectin-like activating receptors on natural killer cells surface. NKG2D engaging with the ligands on tumor cells surface can eliminate tumor cells. However,there must exist immune escape mechanisms in tumor mice and cancer patients. Recently, molecular biological techniques are used to regulate the expression of the receptors and ligands to break the immune evasion.
2.Effect of Exercise on the Expression of Adiponectin mRNA and GLUT4 mRNA in Type 2 Diabetic Rats

To investigate the impact of exercise on the expression of adiponectin and GLUT4 mR NA in type 2 diabetic rats, type 2 diabetic rat model was made. The diabetic rats were treated with swimming training for 8 weeks. The expression of adiponectin mRNA in perirenal fat and GLUT4mRNA in skeletal muscles were assessed by reverse transcription polymerase chain reaction (RT PCR) and the levels of blood glucose, serum insulin, and blood lipid were measured. Our results showed that the expression of adiponectin mRNA and GLUT4 mRNA in diabetic model group was decreased by 45 % (P<0.01), 43 % (P<0.01) respectively. The gene expression of adiponectin and GLUT4 was increased significantly in swimming group (P<0.05 and P<0.01, respectively).Compared with the model group, fasting insulin, TG, TC and FFA were decreased significantly in the training group (P<0.05 or P<0.01) as compared with model group. It is concluded that exercise can promote the expression of adiponectin mRNA and GLUT4 mRNA in type 2 diabetic rats,which may be one of the mechanisms responsible for the amelioration of insulin resistance in the rats.
3.Comparative study on the coronary artery anomalies between the Han and Tibetan nationalities living in middle and high altitude

Objective To analyze the differences of coronary artery anomalies between Han and Tibetan nationality living in middle and high altitude.Methods A total of 7 028 adults living in the Qinghai Plateau (1 800-7 720 m altitude),who underwent coronary CT angiography in Qinghai Cardiocerebro-vascular Disease Special Hospital between 2010 to 2015,were included in this study.There were 6 391 cases of the Han nationality and 637 cases of the Tibetan nationality.The differences of coronary artery anomalies between Han and Tibetan nationality were analyzed retrospectively.Results (1) Incidence of coronary artery anomalies was lower in Han nationality than in the Tibetan nationality (1.596% (102/6 391) vs.4.239% (27/637),P <0.001).(2) There was 64.7% (66/102) male residents with coronary artery anomalies in Han nationality,and 74.1% (20/27) male residents with coronary artery anomalies in Tibetan nationality (P =0.359).(3) Left side coronary artery anomalies in Han nationality was similar as in Tibetan nationality (64.4% (67/104) vs.55.6% (15/27),P =0.396).(4) Incidence of benign coronary artery anomalies was significantly lower in Han nationality than in Tibetan nationality (0.720% (46/6 391) vs.2.200% (14/637),P < 0.001).Incidence of potentially dangerous coronary artery anomalies was also significantly lower in Han nationality than in Tibetan nationality (0.876% (56/6 391) vs.2.041% (13/637),P =0.004).(5)Ten kinds of coronary artery anomalies were found in this study.There were significant differences between Han and Tibetan nationality in left coronary artery originated from right coronary sinus(0.046% (3/6 391) vs.0.471% (3/637),P =0.012),in left circumflex branch originated from right coronary sinus (0.046% (3/6 391) vs.0.471% (3/637),P =0.012),and opening of right coronary artery in left coronary sinus or left anterior descending (0.704% (45/6 391) vs.1.570% (10/637),P =0.018).Conclusion The incidences of coronary artery anomalies and benign coronary artery anomalies were significantly lower in Han nationality residents than that of the in Tibetan nationality residents living in middle and high altitude.
4.The predictive value of artificial intelligence plaque quantitative analysis in coronary heart disease

Objective To explore the predictive value of artificial intelligence(AI)plaque quantitative analysis combined with CT derived fractional flow reserve(CT-FFR)and pericoronary fat attenuation index(FAI)in coronary heart disease(CHD)in Qinghai.Methods A total of 118 suspected CHD patients were selected,and were divided into a stenosis group(n=76)and a non-stenosis group(n=42)based on whether their vascular stenosis rate was>50%.The plaque volume,load,CT-FFR value,and pericoronary FAI of the two groups were measured and compared,and their predictive value in CHD was further analyzed.Results The plaque total volume,calcified and non-calcified plaque volume,lipid plaque volume,and corresponding volume load of the stenosis group were significantly higher than those of the non-stenosis group(P<0.05),the percentage of napkin ring sign and pericoronary FAI were significantly higher than those of the non-stenosis group,and the CT-FFR was significantly lower than that of the non-stenosis group(P<0.05).Logistic regression analysis found that the volume load of calcified and lipid plaques,napkin ring sign,CT-FFR,and pericoronary FAI were independent risk factors for CHD(P<0.05).The combination of AI plaque quantitative analysis,CT-FFR,and pericoronary FAI had high sensitivity and specificity,with high value in CHD diagnosis.Conclusion The combination of AI plaque quantitative analysis,CT-FFR,and pericoronary FAI has high value in the diagnosis of CHD,and may be worthy of clinical promotion application.

6.Effects of self-assembled nanoparticles from Shaoyao gancao decoction on the in vitro release and intestinal absorption of the main components of Glycyrrhiza uralensis

OBJECTIVE To study the effects of self-assembled nanoparticles from Shaoy ao gancao decoction (SGD-SAN)on the in vitro release and intestinal absorption of the main components of Glycyrrhiza uralensis . METHODS Gancao single decoction (GSD),Shaoyao single decoction (SSD),mixed suspension of Shaoyao and Gancao single decoction (MSSGD)and SGD (i.e. Shaoyao-Gancao decoction )were prepared ,and SAN was characterized. HPLC method was adopted to determine the contents of 7 main components (liquiritin apioside , liquiritin, isoliquiritin apioside , isoliquiritin, liquiritigenin, glycyrrhizic acid , isoliquiritigenin)in G. uralensis . The dialysis bag method was used to investigate the effects of the formation of SGD-SAN on in vitro release of 7 main components in G. uralensis with pH 1.2 HCl solution and pH 6.8 phosphate buffered solution (PBS)as release media. Single-pass intestinal perfusion study was performed to investigate the effects of the formation of SGD-SAN on the intestinal absorption of 7 main components from G. uralensis . RESULTS SAN with particle size of 200-300 nm and polydispersity index of 0.3-0.5 was found in GSD ,MSSGD and SGD. GSD-SAN and MSSGD-SAN were in rod shape while SGD-SAN was irregularly spherical under transmission electron microscope. The results of in vitro release study showed that the formation of SGD-SAN could significantly increase in vitro release of liquiritigenin ,isoliquiritigenin and glycyrrhizic acid ,and had no effect on other components of G. uralensis in pH 1.2 HCl solution. The formation of SGD-SAN also had no effect on the release of each component from G. uralensis in pH 6.8 PBS. The results of intestinal perfusion experiments showed that the formation of SGD-SAN could significantly promote the absorption of each component from G. uralensis in the ileum. CONCLUSIONS- The formation of SGD-SAN significantly improves the in vitro release of poorly soluble components from G. uralensis and promotes the intestinal absorption of main components from G. uralensis ,which is the physical structure basis for the compatibility and synergy of Paeonia lactiflora and G. uralensis .
7.Characteristics of the rhizosphere and phyllosphere microbial community of three submerged plants in the dragon-shaped water system of Beijing.

Aquatic plants and the epiphytic microorganisms are important contributors to the purification of constructed wetlands. Taking the dragon-shaped water system of Beijing Olympic Park as a model, this study analyzed the structure and function of the microbial communities reside the sediment, the water body and the rhizosphere and phyllosphere of three submerged plants-Vallisneria natans, Myriophyllum verticillatum, and Potamogeton pectinatus using high-throughput sequencing technology. The results showed that the microbial diversity from the highest to the lowest were samples from sediment, plant rhizosphere, plant phyllosphere and water. The microbial diversity of plant phyllosphere samples were significantly higher than those of the water body. LEfSe analysis showed that different habitats enriched different microbial groups. The sediments mainly enriched anaerobic microbes, while the water body and the phyllosphere of plants mainly enriched aerobic microbes, and the rhizosphere of plants had the both. Functional prediction analysis showed that the abundance of denitrification marker genes in phyllosphere samples was higher than that in samples from rhizosphere, sediment and water body, and the abundance of denitrification marker genes in phyllosphere samples of M. verticillatum and P. pectinatus was higher than that of V. natans. This study could serve as a guidance for the selection of submerged plants and functional microorganisms for constructed wetlands.
