1.ULTRASTRUCTURE OF CARDIOMYOCYTES AND IMMUNOCYTOCHEMICAL LOCALIZATION OF ATRIAL NATRIURETIC PEPTIDE IN RAT ANTERIOR VENAE CAVAE

The wall of rat anterior venae cavae has cardiomyocytes running in different directions. These cardiomyocytes were connected by the intercalated discs. Some myocytes in the inner layer of the AVC near the atrium contained specific granules, which were very similar to those in the atrial myocytes in shape and distribution, but the number of the specific granules in the myocytes of venae was much less than that in the myocytes of atrium. Well-developed Golgi complexes, rough surfaced endoplasmic reticulum and multivesicular bodies were frequently found near the specific granules. Immuno-electron microscopy of the specific granules of the venae by using rabbit anti-synthetic rat ANP (1-28) serum, postembedding method and immunogold staining disclosed gold particles on the specific granules of myocytes in the venae were found in the atrial specific granules. It was also shown that the diameter of the labelled granules was somewhat related to the number of the labelling gold particles. No gold particles of the myocytes of the venae when the section was beforehand treated with the solution containing a combination of rat ANP and rabbit anti-ANP serum. The result indicated the presence of atrial natriuretic peptide (ANP) in the specific granules of myocytes of rat anterior venae cavae as well as in those of the atrium. The formation process of the specific granules and the possible way of the release of their contents were discussed.
2.STUDY ON APOPTOSIS IN ACUTE ISCHEMIC AND REPERFUSED RAT MYOCARDIUM

Objective To demonstrate in vivo that there is different degree myocardium apoptosis during acute ischemia and reperfusion and to explore prelimarily the relations between myocardium apoptosis and the expression of Bcl 2/Bax protein in rats. Methods The morphologic features of apoptotic cardiocytes were pursuited by transmission electron microscopy.DNA extracted from cardiocytes were used to identify the apoptotic DNA ladder by agarose gel eletrophoresis. DNA fragmentation was identified histochemically by TUNEL staining. Expression of Bcl 2/Bax protein in apoptotic myocardium was observed by means of immunohistochemistry and analysed by image analysis system. Results In both ischemia and reperfusion groups morphologic features of apoptotic myocardium were observed by transmission electron microscopic and the apoptotic cardiocytes were detected by the TdT mediated dUTP nick end labeling (TUNEL) staining. 3.OBSERVATION OF THE RENAL GLOMERULUS CASTS OF DOG UNDER THE SCANNING ELECTRON MICROSCOPE

The kidneys removed from six female adult dogs were injected through the renal artery with ABS at a constant pressure between 33.3—33.7 kPa(250—280mmHg). Small specimens cut out from the kindneys were digested in 3% trypsin solution for 48 hours and then eroded in 50% HCl. The cast specimens were studied under the scanning electron microscope. The results were outlined as follows: 1. The corrosion-cast models of the renal glomerulus of the dogs observed under the SEM could be divided into three kinds: the spherical glomerulus, the chrysanthemum-like glomerulus and mixed glomerulus. 2. In general, the afferent and efferent arteriolae entered or exited at the hilum of glomerulus. Rarely, the efferent vessel left the glomerulus at the opposite side of the hilum. Anastomoses between the afferent and efferent or between the efferents vessels were found in our casts. 3. The capillary network of the juxtaglomerular apparatus was observed at the hilum of glomerulus in our casts. The diameters of the meshes of the capillary network were smaller than that of the vessels themselves.

5.Identification of a novel NF1 mutation in a Chinese family affected with neurofibromatosis type I.

OBJECTIVE:
To explore the molecular basis for a Chinese family affected with neurofibromatosis type I.
METHODS:
Peripheral blood samples were collected from the proband and his parents. Potential mutations of NF1 gene were screened by PCR and Sanger sequencing. Pathogenicity of candidate mutations was analyzed using Polyphen-2 and Provean software.
RESULTS:
Two mutations of the NF1 gene, including c.702G>A (synonymous mutation) and c.1733T>G (missense mutation), were discovered in the proband. Neither mutation was found in his parents and 50 healthy controls. Bioinformatics analysis indicated that the c.1733T>G mutation (p.Leu578Arg) was probably damaging. The affected codon L578 is highly conserved across various species.
CONCLUSION
The c.1733T>C mutation of the NF1 gene probably underlies the neurofibromatosis type I in this family.

6.The value of blastocyst culture on preimplantation genetic diagnosis.

OBJECTIVETo estimate the value of blastocyst culture for preimplantation genetic diagnosis (PGD).
METHODSDay 3 embryos were biopsied and analyzed with fluorescence in situ hybridization (FISH) technique. Embryos with normal FISH results were cultured into blastocysts, and the ones with better morphology scores were transferred. Fourteen embryos with abnormal FISH results were cultured into blastocysts. Part of the cells taken from the blastocysts were amplified by whole genomic amplification (WGA) and assessed by array-based comparative genomic hybridization (array-CGH) analysis.
RESULTSSix blastocysts with normal FISH results were transferred in 5 cycles. Four healthy babies of 3 cycles were delivered. Another one was a singleton pregnancy but with embryo growth arrest, whose villus karyotype was normal. Fourteen embryos with abnormal FISH results were cultured into blastocysts and analyzed by array-CGH. Six blastocysts were normal by array-CGH.
CONCLUSIONFISH combined with blastocyst culture may further ensure the accuracy of PGD result. Detection at the blastocyst stage can avoid false positive results and mosaic interferences on Day 3 stage and are therefore more authentic.

7.Clinical application of modified radical neck dissection by gasless unilateral axillary approach in papillary thyroid cancer

Objective:To investigate the effectiveness, safety, and advantages of modified radical neck dissection by gasless unilateral axillary approach (GUA-MRND) in the surgical management of selected patients with papillary thyroid cancer.Methods:We retrospectively analyzed patients with papillary thyroid cancer who underwent GUA-MRND (endoscopic group, n=16) versus unilateral open modified radical neck dissection (MRND) (open group, n=32) during the period from Jan. 2019 to Jun. 2021, including the differences in surgical efficiency, complication rate, and incisional satisfaction.Results:Compared MRND with GUA-MRND, the patients were younger ( P<0.05) , operative time and postoperative drainage anterior ( P<0.01) were slightly inferior in the latter, but it had obvious advantages in cervical swallowing discomfort and incision satisfaction evaluation ( P<0.05) . There was no significant difference in the incidence of temporary recurrent laryngeal nerve injury, intraoperative and postoperative bleeding, hematoma, infection, lymphatic or chylous leakage and supraclavicular numbness after surgery ( P>0.05) . The number of dissected lymph nodes in area II in the GUA-MRND was lower ( P<0.05) , but it was significantly higher ( P<0.01) in area III. And the average regional cleaning efficiency in the GUA-MRND was level Ⅲ (35.5%) , level Ⅵ (28.59%) , level Ⅳ (23.21%) , level Ⅱ (7.18%) and level Ⅴ (7.12%) , suggested that GUA-MRND had higher efficacy for level III, level Ⅵ and Level IV. Conclusion:GUA-MRND is safe, effective, and has high cosmetic satisfaction in the treatment of selected patients with lateral cervical lymph node metastases from papillary thyroid cancer.
