1.Patch testing for selection of cosmetic allergens in female facial dermatitis by European standard of cosmetic allergens

Objective To identify the common allergens of the female patients with facial dermatitis with European standard of cosmetic allergens.Methods Female patients with facial dermatitis were tested with European standard of cosmetic allergens.The reactions to allergens were documented by following the International Contact Dermatitis Research Group recommendations.Results Total 41female patients with facial dermatitis were tested with European standard of cosmetic allergens,themain cosmetic allergens were hexamine(12.20),thimerosal(9.76%),diazolidinyl urea(7.32%)and DMDM hydantoin(7.32%);the main European standard allergens were nickel sulfate(22.20%),formaldehyde(14.63%),4-phenylencdiamine base(9.76%)and fragrance mix (9.76%).Conclusions NickeJ sulfate,formaldehyde,hexamine,thimerosal,4-phenylenediamine base,fragrance mix,diazolidinyl urea and DMDM hydantoin are the main allergens of female patients with facial dermatitis.
2.Conditionally replicative adenovirus under the control of glial fibrillary acidic protein and human telomerase reverse transcriptase dual-promoters direct sodium iodide symporter expression for malignant glioma radioiodine therapy

Objective To explore the possibility of using 131I as a targeted therapy method for malignant glioma by infecting U87 and U251 cells with conditionally replicative adenovirus Ad-Tp-E1a-Gp-NIS.Methods Human telomerase reverse transcriptase (hTERT) promoter and glial fibrillary acidic protein (GFAP) promoter were cloned and their transcriptional activities were detected by luciferase assay.The conditionally replicative adenovirus Ad-Tp-E1 a-Gp-NIS was constructed,purified,and transfected into U87 and U251 glioma cells.For these transfected cells,the selective replication ability was evaluated by plaque forming assay,and protein expression was detected by Western blot assay.125I-iodide uptake and exflux,the clonoy formation of 131I-iodide treated cells were also measured.Results Transcriptions activity of the GFAP and hTERT promoters was 59.75%-62.10% (F =11.89,P < 0.01) in U87 cells and 37.31%-49.00% (F =5.87,P < 0.05) in U251 cells.The Ad-Tp-E1a-Gp-NIS could be selectively replicated and the hNIS gene was successfully expressed in the hTERT-positive and GFAP-positive glioma cells which showed two protein bands with relative molecular mass of 120 × 103 and 49 × 103 in Western blot assay.After infection with Ad-Tp-E1a-Gp-NIS,the cell ability of 125I uptake was increased by 78.80 (F =2 914.58,P <0.01) and 92.48 (F =2 275.91,P <0.01) times in U87 and U251 cells,respectively.The GFAP-negative MRC-5 cells could not take in 125I.The in vitro clonogenic assay indicated that,after 131I treatment,more than 90% of the transfected cells were killed,while only about 65% (t =11.73-78.33,P < 0.01) of control cells were killed.Conclusions The Ad-Tp-E1a-Gp-NIS has a good ability in selective replication and the enhancement of antitumor therapy effect by increasing tumor-specific iodide uptake in malignant glioma cells.
3.Relationship between miR-486-5p and biological behavior of gastric carcinoma SGC7901 cells

Background and purpose:MicroRNA-486-5p (miR-486-5p) has been demonstrated to play an important role in many kinds of tumor, however, there are few reports about the relationship between miRNA-486-5p in gastric carcinoma. This study was aimed to explore the effect of miR-486-5p on the proliferation, apoptosis and migration abilities of the human gastric cancer cell line SGC7901.Methods:Quantitative real-time PCR (qRT-PCR) analysis was performed to detect the expression of miR-486-5p in the SGC7901 and GES-1 cells, miR-486-5p over-expressing plasmid was constructed and transfected into the human gastric carcinoma cell line SGC7901 using LipofectamineTM2000. The expression of miR-486-5p of the transfected cells was measured by qRT-PCR, the proliferation level of SGC7901 cells was detected by MTT method, the apoptosis rate of the cells was measured by lfow cytometry and the in vitro migration abilities of SGC7901 cells by transwell test. Results:The miR-486-5p expression in SGC7901 cells was down-regulated compared with GES-1 cells. The expression of miR-486-5p in SGC7901 cells that was transfected miR-486-5p over-expressing plasmid was obviously up-regulated. The proliferation and migration abilities of SGC7901 cells were inhibited signiifcantly, and the apoptosis rate of the cells increased. Conclusion:miR-486-5p can effectively suppress the proliferation and in vitro migration abilities of SGC7901 cells, indicating that miR-486-5p might be used as a target for molecular therapy of gastric cancer.
4.Study of the effect of sevoflurane and isoflurane anesthesia on postoperative renal dysfunction of severe fracture

[Objective] To investigate the effect of sevoflurane and isoflurane anesthesia on the renal function of patients with severe fracture after surgery.[Methods] Sixty-two patients with severe fracture who needed surgery were enrolled and all received general anesthesia.All the patients were divided by random digits table method into two groups with 31 cases each:sevoflurane group(anesthesia maintenance with 1.5%-3.0% sevoflurane)and isoflurane group(anesthesia maintenance with 1.5%-3.0% isoflurane).The patients in two groups received 0.1-0.2 μg/(kg·min)remifentanll continuous intravenous infusion and atracuurium intermittent intravenous injection for anesthesia maintenance.Serum creatinine(SCr)and blood urea nitrogen(BUN)in two groups after surgery were compared,as well as the number of hemodialysis.[Results]SCr and BUN at 1,3,7 d after surgery in two groups were significantly lower than those before surgery[seveflurane group:(517 ± 187),(163 ± 110),(103 ± 99)μ mol/L vs.(853 ± 220)μmol/L,(17.1 ± 7.8),(9.5 ± 4.1),(9.7 ± 3.8)mmol/L vs.(21.8 ± 9.3)mmol/L;isoflurane group:(539 ± 188),(136 ± 108),(101 ± 95)μ mol/L vs.(881 ± 220)μmol/L,(17.5 ± 7.4),(9.5 ± 3.5),(9.8 ± 3.3)mmol/Lvs.(20.0 ± 8.9)mmol/L](P < 0.05).SCr and BUN between two groups at every time point after surgery had no statistical significance(P > 0.05).There was 1 case(3.2%)in sevoflurane group needed hemodialysis within 7 d after surgery,as well as in isoflurane group,and there was no statistical significance between two groups(P>0.05).[Conclusions] For patients with severe fracture who need general anesthesia and surgery,sevoflurane and isoflurane both have no significant adverse reaction to renal function.Therefore,it is acceptable for the patients to use sevoflurane for anesthesia and it is worthy of clinical popularization and application.
5.The application of developmental assessment in the core specialty courses of emergency and inten-sive care orientation in nursing profession

To construct “developmental assessment” in the course of emergency nursing and inten-sive care nursing by applying the combination of formative assessment and summative evaluation, the com-bination of qualitative assessment and quantitative assessment and the incorporation of self-assessment and other evaluation. Specifically, this project contains investigation activity (accounting for 10%), operating activity (10%), checking on work attendance and homework evaluation (15%), reflection activity (15%), comprehensive design experiment evaluation (20%) and theoretical evaluation (30%). Process evaluation and self-evaluation are highlighted, especially feedback and improvement after evaluation. Through multi stage and multi form evaluation, the goal of emergency and critical care professional education has been achieved, which has promoted the development of student nurses' quality and overall development.
6.Influence of teaching integration of theory and practice on staff-students' satisfaction

Objective To apply teaching integration of theory and practice in curriculum integration of medical and surgical nursing,in order to improve staff-students' satisfaction and promote teaching quality of curriculum integration.Methods The teaching method was designed as the combination of autonomous learning,entering the clinical scene and centralized instructions,and was applied into teaching integration of theory and practice in curriculum integration of medical and surgical nursing in grade 2009 undergraduates.Teachers and students were investigated by the satisfaction questionnaire.Results The students got high average score which was (90.20±2.72) points in the examination of teaching integration of theory and practice.The total satisfaction degree showed significant difference between the normal teaching mode and the new one.Conclusions As an explored innovative teaching mode,teaching integration of theory and practice can well arouse staff-students' learning enthusiasm,promote teaching quality.It is the requirement of curriculum integration and be worthy of being developed.
7.Relationships between implicit and explicit attitude toward death in college students

Objective:To explore the implicit attitude and explicit attitude toward death of college students,and the relationship between the two kinds of attitude.Methods:Totally 65 college studems participated in this study.The Single Category Implicit Association Test (SC-IAT,including cognition,affection and behavior) was used to exam the implicit attitude toward death of undergraduate students.The Death Attitude Profile-Revised (DAP-R,including five dimensions,i.e.,fear of death,death avoidance,death acceptance,escape acceptance,and neutral acceptance) was used to measure the explicit attitude.Results:Data of the SC-IAT indicated that the differences between the mean reaction time of the incompatible task and compatible in all three dimensions and the total SC-IAT were significant (Ps <0.01),and both the D scores of the total SC-IAT and three dimensions were larger than zero.The mean score of neutral acceptance was the highest in the five factors of SC-IAT.Relationships between the implicit effect of SC-IAT and the Mean scores of the DAP-R's five factors were not significant(Ps > 0.05).Conclusion:The implicit attitude toward death of college students is negative,the explicit attitude toward death of college students is neutral.The implicit and explicit attitude toward death of college students is incongruent,and they are relatively independent.
8.Inhibitory effect of 131 I-labeled epidermal growth factor receptor-targeted liposome nanoparticle on EGFR-overexpressing cancer cells in vitro

Objective To construct 131 I labeled anti?EGFR immunoliposome nanoparticle ( 131 I?Cetuaximab ( C225)?BSA?PCL) , and investigate its inhibitory effect on EGFR?overexpressing cancer cells in vitro. Methods Anti?EGFR liposome nanoparticle C225?BSA?PCL and non?targeted liposomes BSA?PCL were constructed. The products were observed with transmission electron microscopy and dynamic light scat?tering. The EGFR?targeted binding and cellular uptake in EGFR?overexpressing cancer cells were observed with flow cytometry and confocal microscopy. Anti?EGFR and non?targeted liposomes were labeled with 131 I using the chloramine?T method. The targeted cell killing effects of 131 I labeled liposomes were analyzed using MTT assay. The time?dependent cellular uptake analysis was used to evaluate the slow?release effects of the 131 I labeled liposomes. The independent?samples t test was used for data analysis. Results The EG?FR?targeted liposome C225?BSA?PCL and non?targeted liposome BSA?PCL were successfully constructed, and the effective diameters were approximately 130-180 nm. Flow cytometry and confocal microscopy re?vealed significant uptake of C225?BSA?PCL in EGFR?overexpressing tumor cells. BSA?PCL could also bind to cells with minimal and weak tumor retention. The EGFR?targeted radioactive liposome 131I?C225?BSA?PCL showed greater targeted cell killing effect than non?targeted liposome 131I?BSA?PCL,the IC50 values of 131I?C225?BSA?PCL and 131 I?BSA?PCL were 0. 03-1. 32 and 0. 25-12. 19, respectively. The uptakes of 131 I?C225?BSA?PCL was higher than that of 131 I?BSA?PCL ( t=3.03-16.86, all P<0.05) and reached the maxi?mal level at 4 h after incubation. Conclusions The EGFR?targeted liposome C225?BSA?PCL demonstrated superior cellular binding and uptake on EGFR?overexpressing cancer cells compared with BSA?PCL. The EGFR?targeted radioactive liposome 131 I?C225?BSA?PCL had favorable intracellular retention and excellent targeted cell killing effect, and could effectively suppress the growth of EGFR?overexpressing cancer cells.
9.Evaluation of the internal therapeutic effectiveness of 131I-antiEGFR-BSA-PCL in nude mice with colorectal cancer

Objective To investigate the biological effects of internal radiation and therapeutic effectiveness of 131I-labeled anti-epidermal growth factor receptor (EGFR) in colorectal cancer of model mice.Methods Nano-liposome characterized for EGFR-targeting was constructed.The efficacy of cellular binding and uptake of the liposome was evaluated by the analysis of confocal microscopy observation and the iodide uptake assay.After intra-tumor injections of 74 MBq (740 MBq/ml) 131 I-antiEGFR-BSA-PCL,131 I-BSA-PCL,131 I or an equivalent volume of normal saline.The biological effects of internal irradiation and therapeutic efficacy of the liposomes on colorectal cancer modeled in a male BALB/c mouse were evaluated by means of tumor size,body weight,histopathology,and SPECT imaging.Results The confocal fluorescence images showed that the antiEGFR-BSA-PCL was successfully internalized into LS180 cells.The 131I uptake efficacy of 131I-antiEGFR-BSA-PCL was significantly higher than that of 131I-BSA-PCL in LS180 cells (t =2.77-5.40,P < 0.01).Tumor size measurement showed that tumor growth was inhibited by the treatment with 131 I-EGFR-BSA-PCL and 131I-BSA-PCL,but had no significant differences between these two groups (P >0.05).It was found that the 131I-antiEGFR-BSA-PCL was markedly taken up by the tumor and reac hed its uptake value of (21.61 ± 1.0 1) and (20.58 ± 0.65)% ID/g at 72 h following drug injection,which was higher than the uptake value of 131 I (t =9.36,8.69,P < 0.01).SPECT imaging assay showed that,after being injected into mouse tumor,the 131 I-EGFR-BSA-PCL and 131I-BSA-PCL were uniformly distributed inside the tumor.Conclusions 131 I-antiEGFR-BSA-PCL obviously suppresses the development of colorectal cancer in mice.
10.A retrospective study:analyzing the risk factors of liver dysfunction in Graves’ disease

Objective Liver dysfunction is a common complication of hyperthyroidism [ mainly Graves’ disease(GD)], that may restrict the choice as well as affect the ultimate outcome of treatment. The purpose of this study was to describe the clinical and biochemical patterns in patients suffering from Graves’ disease and liver dysfunction and to determine influential factors. Methods A total of 1 928 patients received radioactive iodine, 131 I treatment. Before 131 I therapy, 24 h radioactive iodine uptake of thyroid(24 h RAIU), serum free triiodothyronine (FT3 ), free thyroxine( FT4 ), sensitive thyroid-stimulating hormone( sTSH), anti-thyrotrophin receptor antibody (TRAb), thyroglobulin antibody(TgAb), anti-thyroid peroxidase antibody(TPOAb), and serum hepatic function parameters etc were performed. Data were analyzed by the unpaired t-test, the independent samples t-test, the χ2 test, logistic regression, and Pearson bivariate correlation. Results Ages, the course of Graves’ disease, the weight of thyroid, FT4 , TPOAb, and TRAb in Graves’ disease patients complicated with liver dysfunction were higher than those in patients with normal hepatic function, as shown in table 1. The influential factors including age, course of Graves’ disease, heart rate, weight of thyroid, FT4, 24 h RAIU, TgAb, TPOAb, and TRAb. 24 h RAIU were the protecting factors. Age, course of Graves’ disease, heart rate, weight of thyroid, FT4 , TRAb, and TPOAb were the risk factors. Conclusion The risk of liver dysfunction in patients with Graves’ disease was increased in the following cases: age over 45 years, heart rate above 90 bpm, weight of thyroid more than 35 g, course of Graves’ disease longer than 3 years, FT4 greater than 70. 5 pmol/ L, TPOAb above 360 IU/ ml, and TRAb above 15 IU/ L. In these coses 131 I therapy will be recommended.
