1.Study of in vitro antibacterial activity of faropenem against 194 clinical isolates

Objective: To study in vitro antibacterial activity of faropenem.Methods: The minimum inhibitory concentration(MIC) of faropenem,cefoperazone/sulbactam or cefotaxime against 194 clinical isolates was determined by agar dilution method.MIC50,MIC90 and the susceptible rate were counted.Results: MIC50 and MIC90 of faropenem were lower than or equal to 1?g/ml and 4?g/ml for all 194 clinical isolates respectively.All susceptible rates of faropenem were more than 90%.MIC90 of faropenem was similar to that of cefoperazone/sulbactam against beta-lactamase producing Escherichia coli and klebsierlla peumoniae,but was 1/32 MIC90 of cefotaxime.MIC90 of three antibiotics was similar against Proteus spp,Moraxella catarrhalis and Streptococcus pneumoniae.MIC90 of faropenem was marked lower than that of cefoperazone/sulbactam or cefotaxime against Staphylococcus aureus,Staphylococcus epidermidis and Enterococcus faecalis.Conclusion: Antibacterial activity of faropenem against clinical isolates is very strong and it is also strongly against beta-lactamase producing bacteria.It can be expected that faropenem will be used widely in clinic in china.
2.Study on in vitro antibacterial activity of prulifloxacin against pathogenic bacteria of respiratory tract and urinary tract infections

Object:To study in vitro antibacterial activity of prulifloxacin against pathogenic bacteria of respiratory tract and urinary tract infections.Methods:The minimum inhibitory concentration(MIC) of prulifloxacin and levofloxacin against 221 pathogenic bacteria of respiratory tract and urinary tract infections was determined by agar dilution method.Results:MIC50,MIC90 and the susceptible rate of prulifloxacin were≤0.03~1?g/ml,0.5~4?g/ml and 67%~100%,respectively.Those of levofloxacin were 0.125~2?g/ml,0.5~8?g/ml and 61%~100%,respectively.MIC90 of prulifloxacin was half that of levofloxacin against Escherichia coli,Enterobacter cloacae,Pseudomonas aeruginosa and Moraxella catarrhalis.MIC90 of prulifloxacin was equal to that of levofloxacin against Klebsiella pneumoniae,Proteus spp,Staphlococcus epidermidis,Streptococcus pneumoniae and Enterococus faecalis.MIC90 of prulifloxacin was two times that of levofloxacin against Haemophilus influenzae and Staphylococcus aureus.Conclusion:Prulifloxacin had a broad-spectrum antibacterial activity against pathogenic bacteria of respiratory tract and urinary tract infections.
3.Renal Lesion Induced by Oral Preparations of Traditional Chinese Medicines:Analysis of 619 Cases

OBJECTIVE:To analyze the renal lesions induced by oral preparations of traditional Chinese medicines so as to provide theoretic basis for clinical rational drug use.METHODS:The CNKI full-text database between January 1991 and December 2008 were retrieved for clinical reports of renal lesions induced by oral preparations of traditional Chinese medicines.And the pertinent literature was sorted,analyzed and compared.RESULTS:176 reports(totaled 619 cases)were collected.Repeated long-term medication and overdosing etc were the major factors accountable for the renal lesion.CONCLUSION:Importance should be attached to the nephrotoxicity of oral preparations of traditional Chinese medicines,and which should be used rationally in the clinic and periodic examination of patients' renal function should be performed in taking oral preparations of traditional Chinese medicines.
4.Microbial Limit Test for Guizhi Fuling Pills

OBJECTIVE: To establish a method for the microbial limit test of Guizhi funing pills.METHODS: The recovery rates of tested strains treated by Guizhi funing pills were determined by plating method and medium dilution method.The test method of the control bacteria was verified.RESULTS:Guizhi funing pills had no inhibitory action on escherichia coli,canidia albicans and aspergillus niger,but which had potent bacteriostatic action on staphylococcus aureus and bacillus subtilis.CONCLUSION: The medium dilution method is suitable for microbial limit test of Guizhi funing pills,whereas the conventional plating method is applicable for the test of control bacteria.

Objective To explore the sluice gate technology in the control of nutrient vessel retrograde island skin flap with sural nerve small saphenous vein blood flow.Methods Totally 32 cases were divided into group A and group B,group A flap with no special treatment to the return of small saphenous vein,group B applied river gate principle,in operation around the small saphenous vein reserved suture,postoperative through a rubber band traction control flap blood circulation and skin flap swelling. Observation and comparison of two groups of skin flap necrosis.Results There had 4 cases in group A flap incomplete necrosis,the incidence rate was 25.0%,1 cases of skin flap incomplete necrosis in B group,the incidence rate was 6.2%.Conclusion The use ofsluice gate technique to control the small saphenous vein reflux of blood,relieve skin flap swelling,improve the survival rate of the flap is simple and effective.
6.Different screw placement schemes in the treatment of middle-aged and young patients with displaced femoral neck fracture:reduction quality and femoral head necrosis rate

BACKGROUND:The key difference between closed reduction and open reduction for femoral neck fracture is the incision of joint capsule or not. As for this problem, scholars have different opinions, but there is no unified conclusion. OBJECTIVE:To compare reduction quality and the rate of femoral head necrosis of open reduction and closed reduction in the treatment of femoral neck fracture in middle-aged and young patients with displaced femoral neck fracture. METHODS: Clinical data of 102 middle-aged and young patients with femoral neck fracture, who were treated in the Department of Traumatic Orthopedics, Guizhou Orthopedic Hospital from June 2008 to June 2014, were analyzed. They were folowed up. According to the manner of reduction, they were divided into open reduction group (n=39) and closed reduction group (n=63). General data, postoperative fracture healing, the rate of femoral head necrosis and reduction quality were compared between the two groups. RESULTS AND CONCLUSION:No significant difference in general information, preoperative and postoperative hemoglobin difference, fracture healing time and fracture nonunion rate was detected between the two groups (P > 0.05). The rate of femoral head necrosis was lower in the open reduction group than in the closed reduction group, but reduction quality was higher in the open reduction group than in the closed reduction group (P < 0.05). These results indicate that in the treatment of displaced femoral neck fractures in middle-aged and young patients, postoperative femoral head necrosis rate of open reduction and fixation is low, and the quality of reduction is better. Open reduction has a certain advantage, but the case number and case study have a certain limitation. We should choose the appropriate way of operation treatment according to the patient’s condition.

Objective To explore the best technology to produce recombinant mouse endostatin by thioredoxin fusion expression system.Methods The recombinant plasmid pThioHis-endo was further transformed into different E.coli.,including BL21,JM109,DH5?.After induction with IPTG of different concentration or for different time period,thioredoxin-endo fusion protein was expressed in E.coli.and the product was identified by SDS-PAGE.The recombinant endostatin expression and extraction,wash,degeneration and refolding of inclusion bodies were carried out by the optimized route,and the product was further purified by affinity chromatography through Ni~(+) column and identified by SDS-PAGE.Results No difference of endostatin expression in BL21,JM109,DH5? was found.The optimal concentration of IPTG was 0.9 mmol/L and optimal inductive phase was 5 h.The soluble recombinant endostatin could be obtained by affinity chromatography through Ni~(+) column.Conclusion Soluble endostatin recombinant fusion protein of high purity and yield could be obtained by the optimized technique route.

9.Fluorescent Microspheres Lateral Flow Assay Based on Immunomagnetic Separation for Detection of S.choleraesuis

Immunomagnetic separation (IMS) was coupled with fluorescent microspheres lateral flow assay (FM-LFA) for rapid detection of S.choleraesuis in this study.The target bacteria were firstly enriched from sample by immunomagnetic beads (IMBs),then eluted by heat treatment and detected by fluorescent microspheres lateral flow test strip.The IMBs was labeled with 30 μg/mg antibody,and the capture efficiency was greater than 90% against 102-106 CFU/mL of S.choleraesuis with great specificity.The immunofluorescent microspheres were prepared by coupling 300 μg of 11 D8-D4 monoclonal antibody with 1 mg of fluorescent microspheres at pH 6.Monoclonal antibody 5F11-B11 (2.0 mg/mL) and donkey anti-mouse IgG (1.0 mg/mL) were sprayed on nitrocellulose membrane as test line and control line,respectively.The FM-LFA based on IMS was used to detect S.choleraesuis in PBS and milk.The limits of detection in PBS buffer and milk were 1.5×105 CFU/mL and 7.6×105 CFU/mL respectively,which were 10 and 200 times lower than that of traditional fluorescent microspheres lateral flow assay,respectively.The results showed that the method,which could enrich S.choleraesuis in milk effectively,could avoid matrix interference and improve the detection sensitivity,thus had a good application prospect.
10.Femoral head replacement for unstable intertrochanteric fractures in aged patients

BACKGROUND:Undergoing femoral head replacement in senile osteoporosis patients with intertrochanteric fracture is a chal enging problem.
OBJECTIVE:To explore the clinical effect of the use of joint instal ation and fracture reduction on the treatment of unstable intertrochanteric fractures in elderly patients.
METHODS:A total of 21 elderly patients with unstable intertrochanteric fractures underwent femoral head replacement. Simultaneously, we col ected clinical data of 20 elderly patients with unstable intertrochanteric fractures undergoing internal fixation of dynamic hip screw. Operation time, blood loss amount, time of walking practice after replacement, complications, and Harris score were compared between the two groups. Clinical effects in patients with femoral head replacement were observed.
RESULTS AND CONCLUSION:Al patients were fol owed up for averagely 20 months (ranged from 6 months to 36 months). After replacement, the incision was first healing. The operation time of femoral head replacement was short;blood loss amount was less;the time of walking practice was early. However, no significant difference in Harris score was detected between the two groups (P>0.05). After femoral head replacement, no infection, inversion and shift, or hip joint loose occurred. Results suggested that correct anteversion angle, eccentricity and femoral calcar size should be identified before the implementation of artificial joint replacement for the elderly femoral intertrochanteric fracture. After implantation of the prosthesis, reduction and fixation of trochanter and femoral calcar bony landmarks are simple easily operated surgery. It can reduce blood loss amount, shorten operation time and elevate clinical effects.
