1.Ethnic disparity of risk factors for ischemic stroke a retrospective comparison between Han and Uygur patients in Xinjiang, China

Objective To investigate the differences of risk factors for ischemic stroke between the Han and Uygur nationalities in Xinjiang China.Methods The data of demography,vascular risk factors,lifestyle as well as blood pressure,blood glucose and blood lipids on admission of 589 patients with stroke (Han 294 cases and Uygur 295 cases) in 5 hospitals from 2007 to 2009 in Xinjiang,China were collected.The exposure levels or the proportion of the risk factors for ischemic stroke between the 2 nationalities were compared.Results The proportions of smoking (18.0% vs.11.5%,x2 =4.945,P =0.026) and alcohol consumption (37.8% vs.21.5%,x2 =9.884,P =0.002) in the Han patients were significantly higher than those in the Uygur patients,while the body mass index in the Uygur patients with ischemic stroke was significantly higher than that in the Han patients with ischemic stroke (25.168 kg/m2 vs.24.443 kg/m2,t =2.515,P =0.012).Conclusions The onset of ischemic stroke of the Han people in Xinjiang China may be more associated with the exposure of smoking and alcohol consumption,while the Uighur people may be more associated with the high-fat and high energy intake caused obesity.
2.Effects of military stress on memory function in battlefield environment confined intensive training of peace-keeping soldiers

Objective To explore the effects of military stress on memory function in battlefield environment confined intensive training of peace-keeping soldiers.Methods 41 peace-keeping soldiers were chosen as the research objects.They were tested the memory function,respectively after they trained intensely in the domestic and in Sudan for five months.The memory function included verbal memory:WHO-UCLA auditory verbal learning test (immediate recall,the insert test,the short time delay memories,long delayed recall,long delayed recognition,visual memory) using the Rey Ostereitb Complex Figure (graphics copy and graphic delayed recall).Results The peace-keeping soldiers got lower scores in the insert tested(4.59±2.03) and graphics copy(35.85± 0.36) than in the domestic (4.85 ± 2.21,35.71 ± 0.46,P> 0.05),but there was no significant difference.They got lower scores in the immediate recall (41.68±9.87),the short time delay memories (9.27±2.77),long delayed recall (9.12±2.99),long delayed recognition (27.56± 1.70),and graphic delayed recall (16.90± 6.16) than those in the domestic(51.68±8.63,11.73±2.15,12.24± 1.96,29.00± 1.43,23.15±7.16,P<0.01),and the difference was statistically significant.Conclusion The peace-keeping soldiers battlefield environment stress has a certain effect on the soldiers' memory function.
3.Podophyllotoxin derivative LN-13 induced multidrug resistant cell K562/A02 apoptosis and its molecular mechanism

Aim To study the mechanism of action of the new derivative of podophyllotoxin(LN-13)in indu-cing the apoptosis of K562/A02 cells.Methods The MTT method was taken to detect the inhibition of LN-13 and VP-16 on K562/A02 proliferation and inhibi-tion rate and IC50 values were obtained 48 hours later. The K562/A02 cell morphological change induced by LN-13 were observed through Hochest33342 and PI staining after 48 hours later.Flow cytometry was taken to detect the apoptosis of K562/A02 cells induced by LN-13.The reverse transcription-polymerase chain re-action was taken to detect the Bcl-2,Bax,caspase-3 and mdr-1 mRNA expression.The expression of P-gp was detected by Western blot.Results The growth of K562 /A02 cells was obviously inhibited by LN-13 when IC50 value was 3.32 μmol · L-1 .LN-13 could obviously induced cell apoptosis observed by Ho-chest33342 and PI staining.Flow cytometry detection showed that LN-13(2,4,8 μmol·L-1 )could induce cell apoptosis and apoptosis ratio reached 15.0%, 48.0%,68.96%,respectively.The reverse transcrip-tion-polymerase chain reaction showed that LN-13 in-creased the Bax and Caspase-3 mRNA expression,and meanwhile the expression of mdr-1 mRNA decreased. Western blot showed that P-gp expression was de-creased as the LN-13 dose increased.The data were significantly different from those of control group.Con-clusion Podophyllotoxin derivative LN-13 can induce the apoptosis of K562 /A02 cells,which may be close-ly-related to regulating P-gp expression and apoptosis related gene mRNA expression.
4.Association study between WNK1 gene polymorphisms and ischemic stroke in Uygur

Objective To explore the association between single nucleotide polymorphisms (SNPs) in WNK1 gene and ischemic stroke in Uygur population. Methods Ten tagging single nucleotide polymorphisms (tSNPs) of the WNK1 gene in 295 ischemic stroke patients and 318 control subjects were genotyped,and the association between these tSNPs and ischemic stroke were conducted.The 10 tSNPs were rs3858703, rs11611246, rs7305065, rs1990021, rs34408667, rs12309274, rs1012729, rs956868,rs12828016 and rs953361. They were determined by the Multiplex SNaPshot platform. All data were analyzed using t-test,x2-test and Logistic regression. Linkage disequilibrium and Haplotype were analyzed by Haploview software.Results There were no significant differences between cases (25.6%) and controls(30.0% ) of the 10 tSNPs in WNK1 gene.When the samples were further stratified according to gender,rs11611246 T allele was found to be associated with a reduced risk of ischemic stroke,with a per-allele OR of 0.448(95% CI 0.269-0.746,P =0.002) in female cases than in female controls. The significance remained after adjustment for the covariates of age,and for the covariates of age,BMI,cigarette smoking,alcohol drinking,hypertension,diabetes and hyperlipidemia.In addition,no association between the other 9 tSNPs and ischemic stroke were found in Uygur subjects.Conclusion The study reports a new genetic variant,rs11611246,located in the fourth intron of the WNK1 gene,decreasing the risk of ischemic stroke in Uygur population.The T allele might be the protecting factor of ischemic stroke in female Uygur.
6.Establishment of RAW264.7 cell line stably expressing Mycobacterium tuberculosis protein ESAT-6.

For studying the effects of Mycobacterium tuberculosis secretory protein ESAT-6 on the related functions of macrophages, RAW264.7 cells were transfected with pEGFP-C1-ESAT-6 and pEGFP-C1 by liposome respectively. After screening with a high level of G418, the macrophage cell lines that stably expressed EGFP-ESAT-6 fusion protein or EGFP were established. The gene and protein expression levels were further analyzed by RT-PCR, fluorescence microscopy and Western blotting. The results indicated that the EGFP-ESAT6 fusion gene was integrated into the chromosome and the protein could be stably expressed in the selected macrophage cell line. These results gave us a tool for the future study in the mechanisms of ESAT-6 protein in modulating the macrophage cells.
