1.The Expression and Clinical Significance of Vascular Endothelial Growth Factor-C in Non-small Cell Lung Cancer

Objective To investigate the expression of vascular endothelial growth factor-C(VEGF-C) in human non-small cell lung cancer(NSCLC) and to elucidate its roles in lymph node metastasis and prognosis evaluation. Methods The expression of VEGF-C was examined in 50 NSCLC specimens and 20 archival surgical specimens of human pulmonary inflammatory pseudotumor tissues by immunohistochemistry. Results The positive rate of VEGF-C expression in NSCLC and inflammatory pseudotumor was 44% and 5% respectively. There was significant difference in VEGF-C expression between the two groups. Objective To study the value of two-dimensional Echocardiography(2DE)and doppler tissue synchronization imaging (TSI) during early diagnosis of close myocardial contusion.Methods 9 small Guizhou-Panama pigs were used.The close myocardial contusion animal model was successfully established by using the serf made small impactor.Echocardiography wag applied before and after injury for 0.5,2,4,8 and 12h respectively,these data were analyzed together with the TYC pathological results.Results After the strike for 0.5h,the location and area of the damage call be directly and rapidly shown by 2DE and TSI,which showed that after myocardial contusion (MC),main damaged areas are anterior and lateral myocardial walls.After myocardial contusion MC,three echocardiography techniques were used to observe the scale of the abnormal segment,the movement of the myocardial wall,Time tO Peak of Systolic Velocity and wall motion segmental inter(WMSI),Time to Peak of Systolic Velocity index(TPI),which all were increased than that pre-injury.Conclusion 2DE and TSI can be used for accurately early diagnosis of the location of myocardial contusion.TSI is more specific for the diagnosis of myocardial contusion.
3.Clinical and etiology analysis of 54 patients with pulmonary mycosis

Objective To investigate the clinical manifestations, imaging features, diagnosis and management of pulmonary mycosis.Methods Fifty-four patients were identified with pulmonary mycosis by pathological examination in our hospital from January 2002 to June 2008. Data of clinical manifestations, imaging features, diagnosis and management were retrospectively reviewed.Results Of the 54 cases, 30 (55.6%) were pulmonary aspergillosis, 15 (27.8%) were pulmonary cryptococcosis. Underlying disease was reported in 42 of 54 (77.8%) cases. The diagnosis of all the patients was confirmed by pathological exami-nation. Lung or bronchi tissue was obtained by operation in 30 (55.6%) cases, by bronchofibroscope in 12 (22.2%) cases, by CT-guided percutaneous needle biopsy in 9 (16.7%) cases, and by lymphoid node biopsy in 4 cases. Main symptoms included cough (41, 75.9%), expectoration (30, 55.6%), hemoptysis (20, 37.0%), fever (16, 29.6%), and asymptomatic (6, 11.1%). The X-ray and chest CT showed masses or nodule lesions (41, 75.9%), patchy lesions (8, 14.8%), cavity (12, 22.2%), diffuse milliary nodules (1 case). Bilateral lungs were involved in 7 cases (13.0%);right lung alone in 26 cases (48.1%);left lung alone in 21 cases (38.9%). Among the 54 cases, 41 (75.9%) were misdiagnosed before pathological examination. Thirty (55.6%) cases underwent surgical resection of pulmonary lesions. One patient suffered from cryptococcal meningitis after operation. Twenty (37.0%) patients received systemic anti-fugal therapy. Cure or significant improvement was found in 16 cases. Conclusions The most frequently isolated fungi were Aspergillus, followed by Cryptococcus. Final diagnosis is mainly dependent on pathological examination. Clinical manifestations, imaging features, diagnostic procedures and management are different due to the difference of pathogenic fungi. Satisfactory Results can be obtained by anti-fungal therapy combined with surgical management.
4.Activation of SDF-1/CXCR4 axis induces proliferation,migration,and tubulogenesis of human endothelial progenitor cells

Objective To explore the effect of chemokine SDF-1 on the proliferation,migration and in vitro of human endothelial progenitor cells(EPCs).Methods The expression of CXCR4 and SDF-1 on EPCs was detected with immunocytochemistry.Proliferation,migration and in vitro tubulogenesis of EPCs was detected by MTT,Millicell chemotaxis and three-diamensional in vitro Matrigel assays,respectively.Results SDF-1 and CXCR4 were expressed on EPCs.SDF-1 induced proliferation,migration and tubulogenesis of EPCs.However,AMD3100 abolished the effects induced by SDF-1.Conclusion The SDF-1/CXCR4 axis possibly plays a critical role in regulating vasculogenesis of EPCs.
5.Antibiotic Susceptibility of Clinical Isolates from Lower Respiratory Tract Infection Patients in Respiratory Ward

OBJECTIVE To investigate the pathogenic bacteria distribution and susceptibility of lower respiratory tract infection in patients in our respiratory ward.METHODS All the clinical isolates from sputum or secretion of lower respiratory tract from 2003 to 2007 were analyzed retrospectively.RESULTS A total of 1529 clinical isolates were collected,including 1326 strains of Gram-negative bacilli(86.7%) and 203 strains of Gram-positive cocci(13.3%).The proportion of Gram-negative bacilli was higher in lung underlying diseases group when compared with non-lung underlying diseases.Haemophilus parainfluenzae and H.influenzae were susceptible to spectinonycin,doxycycline and third-generation cephalosporins.Pseudomonas aeruginosa and Acinetobacter baumanni were susceptible to meropenem,cefoperazone/sulbactam and cefepime.Klebsiella pneumoniae and Escherichia coli were susceptible to meropenem,cefoperazone/sulbactam and amikacin.The major Gram-positive isolates were highly resistant to azithromycin,erythromycin,penicillin.All Staphylococcus aureus isolates were susceptible to vancomycin.Streptococcus pneumoniae isolates were highly susceptible to meropenem,levofloxacin,cefazolin and vancomycin.CONCLUSIONS Infection caused by Gram-negative bacilli are predominant in lower respiratory tract infection patients,especially in the patients with lung underlying diseases.Antibiotics resistance of some pathogens become increasingly severe.Therefore,the rational use of antibiotics is important for the containment of bacterial resistance.
6.Effects of NDGA on the growth of a human malignant glioma cell line CHG-5

Objective To investigate the effects of nordihydroguaiaretic acid (NDGA) on the growth of a human malignant glioma cell line CHG 5 in vitro and in vivo . Methods Colorimetric MTT assay, flow cytometry, and light microscopy were used to investigate the proliferation in vitro , cell cycles, apoptosis of CHG 5 cells, and the growth of xenografted tumor in nude mice. Results NDGA significantly inhibited the proliferation of CHG 5 cells in vitro . Cells in G 0/G 1 phase increased, but cells in S, G 2/M phases decreased, and apoptotic cells increased significantly. After treatment of NDGA (50 mg/kg, intraperitoneally) at 5 d after the inoculation of tumor cells, the xenografted tumor volume reduced remarkably without causing significant toxic and side effects. Conclusion The inhibitory effect of NDGA on the growth of CHG 5 cells may be correlated with the regulation of cell cycles and induction of apoptosis.
7.Expression of COX-2 and its prognostic significance in non-small cell lung cancer.

BACKGROUNDTo investigate the expression of COX-2 and its relation to clinical pathophysiological features and prognosis in non-small cell lung cancer (NSCLC).
METHODSThe expression of COX-2 protein was detected in 52 NSCLC tissues by immunohistochemical (S-P) method.
RESULTSThe positive COX-2 expression was observed in 25 (48.1%) cases of NSCLC tissues. The positive rate of COX-2 expression was 76.5% and 34.3% in adenocarcinoma and squamous cell carcinoma respectively (P < 0.01). The positive rate of COX-2 expression in T3+T4 disease (92.3%) was remarkably higher than that in stage T1+T2 (33.3%) (P < 0.01). There was a remarkable difference in COX-2 expression rate between clinical stage I+II (28.1%) and clinical stage III+IV (80.0%) groups (P < 0.01). The positive rate of COX-2 expression was 83.3% in those with lymph node metastasis, but only 17.9% in those without lymph node metastasis (P < 0.01). In addition, there were significant differences in positive rate of COX-2 expression among patients with ≤2, > 2 but < 5, ≥5 years of survival span respectively (P < 0.01).
CONCLUSIONSOverexpression of COX-2 in NSCLC, especially in adenocarcinoma, is closely related to invasion, lymph node metastasis and clinical stage of lung cancer. It may play a role in development of NSCLC, and also may be a prognostic marker.
8.Relationship between mucosal patterns of Barrett esophagus under magnifcation endoscopy and pa-thology

Objective To investigate the relationships among clinical features, endoscopic characteristics and pathologic epithelial types of Barrett esophagus. Methods Magnification chromoendoscopy ( MCE) was performed in 2506 patients with gastroesophageal reflux disease ( GERD) and 106 patients with Barrett esophagus in our hospital during Feb,2003 -Feb,2004 were analyzed. The clinical features, endoscopic characteristics, pathologic epithelial types and their relationships were analyzed. Results The symptoms of gastro esophageal reflux disease ( GERD) were the main presentation of Barrett esophagus but 27 (25. 5% ) cases without such presentation. Four types of mueosal patterns, dot pattern, ridge or villous pattern and irregular/distorted pattern. were noted within the columnar mucosa using high magnification endos-copy: There were three epithelial types within the columnar lined esophagus: intestinal metaplasia (IM ) , cardiac and fundie types. Three epithelial types were noted in the methylene blue staining areas; ridged/vil-lous pattern and irregular/distorted pattern, all of them were in epithelial types of intestinal metaplasia. Conclusion Magnification chromoendoscopy helps to identify areas with IM, and having important significance in diagnosis and clinical follow up of Barrett esophagus.

10.BMI-1 gene expression and its significance in leukemia

Objective To investigate the expression and its significance of BMI-1 gene in leukemia patients.Methods BMI-1 gene level was assessed by reverse transcription polymerase chain reaction (RT-PCR)in acute leukemia patients,chronic myeloid leukemia(CML)patients,chronic lymphoblastic leukemia(CLL)patients,and bone marrow mononuclear cells(BMMNC)as normal controls,as well as leukemia cell line K562.The expression of BMI-1 gene in leukemia and the relationship was explored.Results The expression of BMI-1 gene was positive in leukemia cell line K562.but negative in 10 controls as well as in CLL.The BMI-1 gene was expressed in 15.4% of CML patients and 47.1% of AL patients.The positive expression of BMI-1 gene in CML-CR was significandy lower than that in CML-BP (P<0.05).Conclusion BMI-1 gene was over expressed in leukemia patients,and may be related to curative effect of the disease.It may be used for leukemia treatment targeting the provision of new sites.
