1.The research on the different expression of matrix metalloproteinase-9 in cervical columnar ectopy and cervical intraepithelial neoplasia

Objective To explore the different probability of carcinoma and the necessity of treatment in cervical columnar ectopy (CCE),normal smooth cervix and cervical intraepithelial neoplasia(CIN) by investigating the different expression of matrix metalloproteinase-9(MMP-9).Methods The expression of MMP-9 was detected by using immunohistochemical method in 30 cases of CCE,28 cases of CIN and 30 cases of normal smooth cervix from 2007 to 2012.Results The positive expression of MMP-9protein in CIN was 96.4% (27/28),CCE was 23.3% (7/30) and normal smooth cervix was 20.0% (6/30)respectively.The positive expression of MMP-9 protein was higher in CIN than CCE and normal smooth cervix,the difference has statistical significance (P< 0.01),there was no significant difference between CCE and normal smooth cervix (P > 0.05).Conclusions CCE,which was a physiological performance of cervical erosion,should not be overtreated.But the women of childbearing age should be routinely performed in cervical cancer screening.
2.Relationship between multi-gene proteins in colorectal carcinoma complicat-ed with chronic schistosomiasis:an immunohistochemical study by using tis-sue microarray techniques

Objective To investigate the relationship between p53,COX-2,Bax,c-myc genes and colorectal carcinoma complicated with chronic schistosomiasis. Methods One hundred and sixty patients with colorectal carcinoma were selected and divided into two groups;a schistosomiasis group(colorectal carcinoma complicated with chronic schistosomiasis,n=80)and a non-schistosomiasis group(colorectal carcinoma uncomplicated with chronic schistosomiasis,n=80). The tissue microarray tech-niques and immunohistochemistry method were used in all the patients to detect the expressions of p53,COX-2,Bax and c-myc proteins. Results The positive rate and level of p53 protein expression in the schistosomiasis group were lower than those in the non-schistosomiasis group,but there were no significant differences between the two groups(both P>0.05). The COX-2 protein in both groups was positive,but the positive expression level of COX-2 in the schistosomiasis group was higher than that in the non-schistosomiasis group,and there was a significant difference between the two groups(P<0.01). The positive rate and level of Bax protein expression were not significantly different between the two groups(both P>0.05). The positive rate of c-myc expression in the schistosomiasis group was higher than that in the non-schistosomiasis group,with a significant difference(P<0.01),but the positive expression level was lower than that in the non-schistosomiasis group,and there was a significant difference between the two groups(P<0.01). Conclusions Schistosome infection may impact on the deficiency of p53 of human colorectal cancer cells. It may promote the excessive expression of COX-2 protein,which is an indirect carcinogenic factor. The expression of Bax gene has no correlation with schistosome infection. The schistosome chronic infection may cause a persistent low level expression of c-myc gene.
3.Influence of chronic fluorosis on protein kinase Cβ/p66shc signal pathway in the brain of rats

Objective To investigate the influence of chronic fluorosis on protein kinase Cβ (PKCβ)/p66shc signal pathway in the brain of rats,and reveal the molecular mechanism of brain damage.Methods According to body weight by the random number table method thirty SD rats were divided into three groups of 10 each (half females and half males),the normal control group [less than 0.5 mg/L of fluorine (prepared with NaF) in drinking water],low fluoride exposure group (10.0 mg/L fluorine),and high fluoride exposure group (50.0 mg/L fluoride).The experiment period was 6 months.The protein level of PKCβ,p66shc,phospho-p66shc and preserved ammonia acyl isomerase (Pin1) in rat brain was detected by Western blotting.The level of neuron nuclear antigen (NeuN),p66shc and phospho-p66sh in brain of rats was detected by immunohistochemistry.Results By Western blotting,the levels of PKCβ,Pin1 and phospho-p66shc protein in brain tissue in high fluoride exposure group [(193.00 ± 57.53)%,(228.21 ± 71.14)%,(201.54 ±:50.86)%] were higher than those of the normal control groups [(100.00 ± 21.24)%,(100.00 ± 40.55)%,(100.00 ± 13.35)%,all P < 0.05].By immunohistochemistry,the numbers of NeuN staining in brain tissue of the rats in both high and low fluoride exposure groups [(49.50 ± 12.57)%,(65.66 ±14.58)%] were lower than that of the control group [(100.00 ± 18.32)%,all P < 0.01].The level of phospho-p66shc protein in brain tissue in high fluoride exposure group [(242.66 ± 93.01)%] was higher than those of the low fluoride exposure and the normal control groups [(152.53 ± 60.65)%,(100.00 ± 25.63)%,all P < 0.01].Conclusion Chronic fluorosis has increased the expressions of PKCβ,Pin1 and phospho-p66shc at protein level in brain of rats,which may be related to the molecular mechanism of brain damage resulted from chronic fluorosis.
4.P16/Ki-67 double dye the value of diagnosis of cervical intraepithelial lesions and cervical biopsy consistency analysis

Objective To evaluate the role of P16/Ki-67 double dye in the diagnosis of cervical intraepithe-lial lesion(CIN)value,and to analyze its consistency with cervical biopsy.Methods A total of 92 patients with suspected CIN in Shanghai Songjiang District Central Hospital from November 2021 to April 2023 were selected as the research objects.All patients underwent high-risk human papillomavirus(HR-HPV)and P16/Ki-67 double dye.Taking cervical biopsy as the gold standard,the diagnostic value of the two detection meth-ods for CIN was analyzed,as well as the consistency of the two detection methods in the diagnosis of CIN clas-sification and cervical biopsy.Results The results of cervical biopsy showed that 74 cases(80.43%)of 92 suspected CIN patients were positive,including 26 cases of CIN Ⅰ,32 cases of CIN Ⅱ and 16 cases of CIN Ⅲ,and 18 cases(19.57%)were negative.Taking cervical biopsy results as the gold standard,P16/Ki-67 double dye was more accurate than HR-HPV test in the diagnosis of CIN(P<0.05).The receiver operating charac-teristic(ROC)curve showed that the area under the curve of HR-HPV and P16/Ki-67 double dye for CIN di-agnosis was 0.760 and 0.890,respectively,all of them had certain diagnostic value.The consistency between HR-HPV evaluation of CIN classification and cervical biopsy was consistent(Kappa=0.707,P<0.05).The consistency between P16/Ki-67 double dye and cervical biopsy was excellent(Kappa=0.832,P<0.05).Con-clusion P16/Ki-67 double dye with a high diagnostic value in CIN,consistency in determining CIN grade and cervical biopsy is wonderful,which can provide reference for clinical diagnosis.
